A new methodology for the improvement of diagnostic immunohistochemistry in canine veterinary pathology: automated system using human monoclonal and polyclonal antibodies Arq. Bras. Med. Vet. Zootec.
Cassali,G.D.; Silva,P.; Rêma,A.; Gärtner,F.; Gobbi,H.; Tafuri,W.L.; Schmitt,F.C..
The authors describe their experience with an automated immunohistochemical system applied to canine tissue samples. Twenty human cellular markers specific monoclonal and polyclonal antibodies and two different antigen retrieval methods were used in normal and neoplastic breast tissue, as well as skin samples obtained from female dogs of pure and mixed breeds. The antibodies tested were the most frequently used in human and veterinary medicine studies, employed with diagnostic purposes in breast pathology, as well as in cancer research. Most of them may be used to study other normal and abnormal tissues and included cytokeratins, progesterone receptor, c-erbB2, p53, MIB-1, PCNA, EMA, vimentin, desmin, alpha-actin, S-100, pan-cadherin, and E-cadherin. The...

A poor prognosis in human hepatocellular carcinoma is associated with low expression of DPP4 BJMBR
Yu,Hao; Mei,Xiao-Ping; Su,Peng-Fei; Jin,Guang-Zhi; Zhou,Hong-Kun.
This study aimed to explore the prognostic role of dipeptidyl peptidase 4 (DPP4) expression in hepatocellular carcinoma (HCC). DPP4 expression was measured in formalin-fixed paraffin-embedded specimens that were gathered from 327 HCC patients. Immunohistochemistry analyses were utilized to examine DPP4 expression characteristics and prognostic values (overall survival (OS) and time to recurrence) of DDP4 in HCC tissues. In addition, a patient-derived xenograft (PDX) model was used to assess the correlation between DPP4 expression and tumor growth in vivo. DPP4 was expressed in low levels in HCC tissues in contrast to paired peritumoral tissues (38 cases were down-regulated in a total of 59 cases, 64.4%. P=0.0202). DPP4 expression was significantly...

Pires,Prhiscylla Sadanã; Ecco,Roselene; Silva,Rodrigo Otávio Silveira; Araújo,Marina Rios de; Salvarani,Felipe Masiero; Heneine,Luíz Guilherme Dias; Oliveira Júnior,Carlos Augusto de; Lobato,Francisco Carlos Faria.
ABSTRACT: A standardized immunochemistry method for the diagnosis of clostridial myonecrosis was applied to 38 formalized tissue samples from ruminants with clinical and post mortem history suggestive of blackleg or gas gangrene. The diagnosis of clostridial myonecrosis was confirmed in 37 out of 38 (97.4%) samples tested. Clostridium chauvoei and Clostridium perfringens type A were the most common agents found alone, being detected in ten (26.3%) and six (15.8%) samples, respectively. The other cases showed an association of two or three clostridia, with C. perfringens type A detected in 11 (29%) cases. Based on the findings of the present study, polyvalent vaccines against clostridial infections of animals incorporating C. perfringens would be more...

Abundant immunohistochemical expression of dopamine D2 receptor and p53 protein in meningiomas: follow-up, relation to gender, age, tumor grade, and recurrence BJMBR
Trott,G.; Pereira-Lima,J.F.S.; Leães,C.G.S.; Ferreira,N.P.; Barbosa-Coutinho,L.M.; Oliveira,M.C..
Meningiomas are common, usually benign tumors, with a high postoperative recurrence rate. However, the genesis and development of these tumors remain controversial. We aimed to investigate the presence and implications of a mutated p53 protein and dopamine D2 receptor in a representative series of meningiomas and to correlate these findings with age, gender, tumor grade, and recurrence. Tumor tissue samples of 157 patients diagnosed with meningioma (37 males and 120 females, mean age 53.6±14.3 years) who underwent surgical resection between 2003 and 2012 at our institution were immunohistochemically evaluated for the presence of p53 protein and dopamine D2 receptor and were followed-up to analyze tumor recurrence or regrowth. Tumors were classified as...

Administering ascorbic acid to rats undergoing ageing processes: effects on myosin-V immunoreactive myenteric neurons Anais da ABC (AABC)
Mello,Sônia A.; Marese,Angélica C.M.; Brancalhão,Rose M.C.; Zanoni,Jacqueline N.; Natali,Maria Raquel M..
During the ageing process the enteric nervous system undergoes morphofunctional changes, such as enteric neurodegeneration. Neuronal death can be attributed to increase radicals free, and ascorbic acid (AA), known antioxidant, could minimize damage cause by oxidative stress. The objective of this study is to analyse the behaviour of morphoquantative myenteric neurons in the duodenum of adult Wistar rats with aged 90 (C90), 345 (E345) and 428 (E428) days, as well as animals of the same age who received ascorbic acid supplementation for 120 days (EA345 and EA428). Whole-mount preparations of muscle layer from the duodenum of the animals were immunostained by the method myosin V. 80 microscopic fields were quantified (14.8 mm2/animal) and measured 100...

Analysis of EGFR and HER-2 expressions in ductal carcinomas in situ in canine mammary glands Arq. Bras. Med. Vet. Zootec.
Silva,I.L.D.; Dias,A.P.M.; Bertagnolli,A.C.; Cassali,G.D.; Ferreira,E..
Biomolecular evidence has shown that ductal carcinoma in situ(DCIS) may develop into invasive carcinoma of the canine mammary gland, and mutations in proto-oncogenes HER2 and EGFR; two members of the family of epidermal growth factor receptors, may be involved in this process. The purpose of this study was the characterization of the immunohistochemical expression of the EGFR and HER2 proteins in the process of neoplastic transformation, supposedly present in ductal carcinomas in situin canine mammary glands. Fifteen cases of DCIS were evaluated, with a higher expression of HER2 and EGFR being observed in low-grade carcinomas when compared with high-grade neoplasms, and with a high positive statistical correlation in the latter. Results suggest that...

Ancillary techniques on the evaluation of canine cutaneous mast cell tumors from Brazil Ciência Rural
Flores,Mariana Martins; Mazaro,Renata Dalcol; Langohr,Ingeborg Maria; Roy,Alma; Strother,Keith; Fighera,Rafael Almeida.
ABSTRACT: The use of histologic classification by a 2-tier grading system only, immunohistochemistry (IHC) for KIT and Ki-67 and polymerase chain reaction (PCR) for internal tandem duplications (ITD) on exon 11 has improved the prognostication of canine cutaneous mast cell tumors (CCMTs) particularly in the United States. However, these techniques are not commonly used in most Brazilian laboratories. Likewise, no studies, to date, have investigated the occurrence of ITD in CCMTs from the country. Thus, this study tested the 2-tier grading system, the immunohistochemistry for KIT and Ki-67 and the PCR for exon 11 in a group of Brazilian CCMTs with the goal of investigating the applicability of these tests in a Brazilian laboratory. Of the 39 CCMTs, 69.2%...

Androgen Receptor Distribution, PAS and Alcyan Blue Reaction in the Vomeronasal Organ and the Nasal Septum Mucosa of the Developing Male Rat International Journal of Morphology
Núñez Chichet,M E; Genovese,P; Bielli,A.
The vomeronasal organ (VNO) is a tubular chemoreceptory organ which detects environmental pheromones and is essential for mammalian normal reproductive activity. A sensitive chemoreceptory neuroepithelium lines the concave VNO wall and the opposite, convex wall is lined by a seudostratified, receptor-free epithelium. The secretion of the Jacobson glands (JG) -tubuloacinary glands lying in the lamina propia of the VNO - is essential for pheromones contacting vomeronasal organ chemoreceptors. Tubuloacinary glands lying in the connective tissue of the nasal septum mucosa (Bowman glands), can be classified as ventral and dorsal, according to their location. Positivity to PAS and Alcyan blue reactions and androgen receptor immunolocalization was evaluated with...

Antifibrotic effects of total or partial application of amniotic membrane in hepatic fibrosis Anais da ABC (AABC)
MAMEDE,KARINA M.; SANT’ANNA,LUCIANA B..
Abstract: Liver fibrosis is the final common pathway of chronic liver diseases, having cirrhosis as a possible progression, which has liver transplantation as the only effective treatment. Human amniotic membrane represents a potential strategy as a therapy for liver fibrosis, due to its anti-inflammatory, anti-fibrotic and immunomodulatory properties. The aim of this study was to evaluate amniotic membrane effects as a treatment for hepatic fibrosis induced in rats by bile duct ligation (BDL), verifying alterations between two different forms of amniotic membrane application, around all the lobes of the liver and around only one lobe of the liver. Two weeks after inducing fibrosis, an amniotic membrane fragment was applied to the surface of the liver,...

Aortic body tumors in dogs from Northern Paraná, Brazil Ciência Rural
Headley,Selwyn Arlington; Bracarense,Ana Paula; Nakagawa,Tizianne; Moreno,Kleber; Pereira,Patrícia Mendes; Reis,Antônio Carlos Faria dos.
This article describes the occurrence of aortic body tumors in four dogs from Southern Brazil. All dogs were males, more than 9 year of age; most of these were from the Boxer breed of dogs. The animals had large neoplastic growths at the base of the heart, which demonstrated histologic characteristics consistent with that of aortic body tumor. By immunohistochemistry, tissue sections from all dogs reacted positively to neuron specific enolase. These cases might represent the first description of this tumor in dogs from Brazil, as confirmed by immunohistochemistry.

Aversive stimulation during the stress-hyporesponsive period does not affect the number of corticotroph cells in neonatal male rats BJMBR
Aguiar,C.E.; Cadore,L.P.; Padoin,M.J.; Barbosa-Coutinho,L.M.; Lucion,A.B..
Immunohistochemistry was used to evaluate the effects of neonatal handling and aversive stimulation during the first 10 days of life on the number of corticotrophs in the anterior lobe of the pituitary of 11-day-old male Wistar rats. Since adult rats handled during infancy respond with reduced corticosterone secretion in response to stressors and with less behavior inhibition in novel environments, we assumed that neonatal stimulation could affect pituitary morphology during this critical period of cell differentiation. Three groups of animals were studied: intact (no manipulation, N = 5), handled (N = 5) and stimulated (submitted to 3 different aversive stimuli, N = 5). The percentage of ACTH-immunoreactive cells in the anterior lobe of the pituitary...

Bovine respiratory syncytial virus: immunohistochemichal detection in mouse and bovine tissues using a Mab against human respiratory syncytial virus Arq. Bras. Med. Vet. Zootec.
Almeida,R.S.; Spilki,F.R.; Roehe,P.M.; Verinaud,L.M.C.; Arns,C.W..
An immunoistochemical (IHC) test was developed to detect bovine respiratory syncytial virus (BRSV) in cell cultures and tissues of experimentally infected mice and calves, using a commercial monoclonal antibody (Mab) against human respiratory syncytial virus (HRSV), as a less expensive alternative, instead of producing specific monoclonal antibodies to BRSV. Clinical samples from calves suffering respiratory disease were also submitted to this test. IHC detected BRSV antigens in mouse tracheas (3, 5 and 7 days post-infection) and lungs (5 and 7 days post-infection), and in one of three lungs from experimentally infected calves. Lungs samples from two naturally infected calves were tested and resulted positive for BRSV by the IHC test. These results suggest...

Andrade,G.I.; Serra,C.V.; Barbosa-Stancioli,E.F.; Serakides,R.; Lobato,Z.I.P..
The immunohistochemistry standardization for bovine viral diarrhea virus (BVDV) diagnostic was described. The formalin-fixed tissue samples from a heifer with mucosal disease were used as positive control. The validation of the first phase results was performed using samples from an aborted fetus and a calf infected with reference strains of BVDV. The best results were seen using monoclonal antibodies and a commercial kit consisting of labelled streptavidin biotin (LSAB) reagents and the diaminobenzidine (DAB) substrate-chromogen reagent. The immunohistochemistry demonstrated to be an useful method for routine diagnosis for the controll and detection of BVDV infection.

Rocha,Paulo Ricardo Dell'Armelina; Lopes,Letícya Lerner; Arruda,Laura Peixoto de; Pescador,Caroline Argenta; Cruz,Raquel Aparecida Sales da; Colodel,Edson Moleta.
Cholangiocarcinomas are neoplasms that originate from the bile duct epithelium. The present case described a cholangiocarcinoma in an adult female American Rhea (Rhea Americana araneipes) by means of gross, histopathology and immunohistochemistry. Irregular, firm, multifocal, yellow-white masses, measuring from 0.4 to 6cm in diameter were observed in both liver lobes. At the cut surface, multiple firm nodules filled with connective tissue were present. Microscopically, the neoplasia was composed of small, irregular, gland-like structures of neoplastic cells surrounded by connective tissue. The cells resembled epithelial cells of the hepatic biliary ducts. Neoplastic cells were positive for cytokeratin and negative for vimentin. This is the first report of...
