Abnormal spindle orientation during microsporogenesis in an interspecific Brachiaria (Gramineae) hybrid Genet. Mol. Biol.

This paper reports a case of abnormal spindle orientation during microsporogenesis in an interspecific hybrid of the tropical grass Brachiaria. In the affected plant, prophase I was normal. In metaphase I, bivalents were regularly co-oriented but distantly positioned and spread over the equatorial plate. In anaphase I, chromosomes failed to converge into focused poles due to parallel spindle fibers. As a consequence, in telophase I, an elongated nucleus or several micronuclei were observed in each pole. In the second division, the behavior was the same, leading to polyads with several micronuclei. A total of 40% of meiotic products were affected. The use of this hybrid in production systems needing good-quality seeds is discussed.

The genetic monitoring of interspecific hybrids involves the application of methodologies able to provide an easy and indubitable genetic characterization of both parental and hybrid individuals. In the present work, cytogenetic techniques were used to identify a hybrid lineage of "Piaupara" in order to caracterize them in relation to the parental species, Leporinus macrocephalus (piauçu) and L. elongatus (piapara). The cytogenetic analysis revealed that L. macrocephalus presented 2n = 54 chromosomes and a nucleolar organizer regions (NOR) at the telomere of the long arm of the submetacentric chromosome pair 2. The in vitro culture of zygotic embryos can be a useful tool to reduce the germination time and provide higher rates of embryos developed into plants (Lima 2013), as the in vitro culture allows the removal of physical barriers such as hard endocarp and facilitates the supply of nutrients to the developing embryo (Pádua et al. 2014). In vitro germination can overcome this problem by embryo rescue. In this study we optimized the in vitro germination of zygotic embryos of BRS Manicoré hybrid palm comparing various culture media and various seed lots.

ABSTRACT The interspecific oil palm hybrid BRS Manicoré (E. guineensis x E. oleifera) has superior agronomic characteristics. However, the germination rate is low (30%) and the process is slow when the seeds are sown in a conventional form. The purpose of this study was to optimize the in vitro germination of zygotic embryos of this hybrid comparing seed lots. The viability of zygotic embryos was evaluated by the tetrazolium test (0.075%) for 4 h. The embryos were cultured on MS and Y3 culture media, with and without the addition of NaH2PO4, as well as on MS, MS1/2 and N6 medium. Meiotic arrest compromises pollen fertility in an interspecific hybrid between Brachiaria ruziziensis x Brachiaria decumbens (Poaceae: paniceae) BABT

Microsporogenesis was analyzed in an interspecific hybrid between an artificially tetraploidized sexual accession of Brachiaria ruziziensis (2n=4x=36) and a natural apomictic tetraploid accession of B. decumbens. Syncytes involving a large number of cells were recorded in 15.4% of meiocytes. Meiosis progressed normally in syncytes during prophase I; in metaphase I, however, several nuclei were found fusioned, showing chromosome stickiness and several chromosome fragments. Meiosis was arrested in metaphase I and pycnotic nuclei and micronuclei were formed. Abnormal cytokinesis fractionated the syncyte into abnormal meiotic products that were covered by the pollen wall.
