Fructan degradation and hydrolytic activity in tuberous roots of Viguiera discolor Baker (Asteraceae), a herbaceous species from the cerrado Rev. Bras. Bot.
Degasperi,Maria I.; Itaya,Nair M.; Buckeridge,Marcos S.; Figueiredo-Ribeiro,Rita De Cássia L..
Fructans of the inulin type are the major reserve carbohydrates in tuberous roots of Viguiera discolor, a perennial herb native to the cerrado. Changes in molecular mass of the polymer, followed by releasing free fructose suggested that hydrolysis could be related to the sprouting of the buds after the dormant period, when aerial parts of the plant are naturally absent. Excision of aerial parts resulted in the increase of fructan 1-exohydrolase (1-FEH) activity in tuberous roots after sprouting. 1-FEH was partially purified from this material by binding to ConA-Sepharose and the highest activity was detected at pH 5.4 and between 20 and 40 °C. Values of Km for V. discolor inulin could not be determined since no saturation was observed up to 10%. The study...

Fructan dynamics in the underground organs of Chresta exsucca (Asteraceae), a dry season flowering species Acta Botanica
Santos,Camila Silva dos; Abraão,Cinara Ferreira; Moraes,Moemy Gomes de.
ABSTRACT Climatic seasonality has an influence on the phenology of native Cerrado plants. Herbs and subshrubs tend to flower in the rainy season, although some species of these habits flower in the dry season. Reserve carbohydrates, stored in the underground organs, are used to support phases of high energy-demand, but also may protect plants from damage during periods of environmental limitation. In this study we evaluated variation in fructan levels in the underground organs of field-grown plants of Chresta exsucca among different phenological phases. Chresta exsucca flowers in the dry season and possesses a diffuse underground system, which stores inulin-type fructans. Resprouting was continual during the sampling period. Oligosaccharide content was...

Fructan production in Vernonia herbacea (Vell.) Rusby is related to adequate nitrogen supply and period of cultivation Rev. Bras. Bot.
Cuzzuol,Geraldo R. F.; Carvalho,Maria Angela M.; Zaidan,Lilian B. P..
Previous studies showed that plants of Vernonia herbacea grown for one year under a limited nitrogen supply presented reduced growth and higher fructan content than plants treated with sufficient nitrogen supply. However, the total fructan production was similar in both plant groups due to the higher biomass of the underground reserve organ in nitrogen-sufficient (N-sufficient) plants. In the present study we aimed to evaluate if a stress growing condition under nitrogen-limited (N-limited) supply, following cultivation under N-sufficient supply would have a positive effect on fructan production. Plants cultivated during one year under N-sufficient supply (10.7 mmol L-1 N-NO3-) were separated in two groups. During the following six months, one group...

Fructans and water suppression on intact and fragmented rhizophores of Vernonia herbacea BABT
Dias-Tagliacozzo,Gláucia M.; Itaya,Nair M.; Carvalho,M. Angela Machado de; Figueiredo-Ribeiro,Rita de Cássia L.; Dietrich,Sonia M. C..
In order to evaluate the involvement of fructans in drought tolerance, experiments were conducted using intact plants and excised rhizophores of Vernonia herbacea. The water content in rhizophores of intact plants was maintained for 30 days when plants were watered every 7 and 15 days, whereas plants remained alive up to 60 days without water. Total fructan, oligo- to polysaccharides ratio and mean molecular mass of polysaccharides increased in these plants indicating depolymerization of median size molecules. In apical fragments of rhizophores kept dry the oligosaccharides increased in relation to polysaccharides one day after excision in treated tissues. This was reflected in the HPLC profile of the oligosaccharides in which the proportion of free...

Amarante,Marla Karine; Ariza,Carolina Batista; Oliveira,Carlos Eduardo Coral de; Gualtieri,Karina de Almeida; Oda,Julie Massayo Maeda; Watanabe,Maria Angelica Ehara.
ABSTRACT Inulin is an effective prebiotic and its potential in modulating systemic immunity have been proposed. A subpopulation of T cells, named T regulatory cells (Tregs), expressing the Forkhead boxP3 transcription factor are key mediators of peripheral tolerance and suppress undesirable immune responses. These Tregs can be induced by cytokine transforming growth factor beta (TGF-β) and interleukin 10 (IL-10). This work aimed to evaluate inulin effects on human peripheral blood mononuclear cells (PBMC) in vitro. PBMC were incubated with inulin, and the expression of TGF-(1, FOXP3 and IL-10 was analyzed. Increased supernatant IL-10 levels were observed in PBMC of inulin-treated group (p=0.03). Moreover, FOXP3 gene expression was 7.6 fold higher in...

Prebiotic inulin: Useful dietary adjuncts to manipulate the livestock gut microflora BJM
Samanta,A.K.; Jayapal,Natasha; Senani,S.; Kolte,A.P.; Sridhar,Manpal.
In recent years, there has been a growing appreciation on the relevance of gastrointestinal microflora in both ruminants and non-ruminants owing to revelation of their role in several physiological functions including digestion, nutrient utilization, pathogen exclusion, gastrointestinal development, immunity system, gut gene expression and quality of animal products. The ban imposed on the use of antibiotics and hormones in feed has compelled animal researchers in finding an alternative which could overcome the issues of conventional feed additives. Though the concept of prebiotic was evolved keeping in mind the gastrointestinal flora of human beings, presently animal researchers are exploring the efficiency of prebiotic (inulin) for modulating the gut...

Production and action pattern of inulinase from Aspergillus Niger-245: hydrolysis of inulin from several sources Rev. Microbiol.
Cruz,Vinícius D’Arcadia; Belote,Juliana Gisele; Belline,Márcia Zilioli; Cruz,Rubens.
A strain of Aspergillus niger isolated from soil samples showed great capacity to produce extracellular inulinase. Although the enzyme has been synthesized in presence of monosaccharides, sucrose and sugar cane molasse, the productivity was significantly higher (p<0.05) when the microorganism was inoculated in media formulated with dahlia extract and pure inulin, as carbon sources. With regard to the nitrogen source, the best results were obtained with casein and other sources of proteic nitrogen, comparatively to the mineral nitrogen. However, statistic significance (p<0.01) only was found between the productivity obtained in the medium prepared with casein and ammonium sulphate. The optimum pH of the purified enzyme for inulin hydrolysis...
