Rubber production is dependent on the availability of carbohydrates in the tissues engaged in latex synthesis. In this study three rubber tree clones were used to study the protein profile, the activities of invertase and sucrose synthase in the latex and anatomical characteristics of the bark, as well as to establish a possible relationship with latex production. The clones used in the study, RRIM 600, GT 1 and Fx2261, differed in latex productivity, from high to low in this order. Although present, few differences were observed in the latex protein profile obtained by denaturing electrophoresis. Duas selecoes para alto e baixo teor em amido nas raizes do algodoeiro (Gossypium hirsutum L.), assim como a populacao original, foram estudadas num experimento em casa de vegetacao. Na populacao alto amido na raiz, a atividade amilolitica nas folhas aumentou, e. diminuiu na selecao para baixo amido, facilitando, assim, na primeira, a exportaca de assimilados, e reduzindoa na ultima. No tronco, a atividade de ß-amilase aumentou na populacao baixo amido, e nas raizes foi a atividade da invertase que aumentou na mesma populacao, o que representa a baixa disponibilidade de assimilados nessa selecao. The yeast strain AK 46 was isolated from dough containing fermented cherry fruits and identified as Saccharomyces cerevisiae. The nucleotide sequence of the rDNA spacer region from this strain coincided with those from the yeasts used for the production of whisky, wine, and bread. In a comparison with six baking strains, strain AK 46 possessed less leavening ability in dough with and without addition of 5% sucrose (based on the weight of flour) but a high ability in sweet dough containing 30% sucrose. The activities of the two enzymes α-glucosidase and invertase, which are related to dough fermentation, were lower than those in the other strains. Objetivou-se avaliar o crescimento da plântula e o metabolismo de carboidratos associados ao aumento da tolerância à hipoxia exercido pela presença do cálcio no período de germinação e/ou alagamento de plântulas de milho com diferentes idades. O experimento foi desenvolvido na Universidade Federal de Lavras, MG, em 2002. Cariopses de milho da variedade 'Saracura' (BRS-4154) foram germinadas em água ou solução de CaCl2. Após dois e quatro dias, as plântulas foram submetidas ao alagamento em tubos de PVC com tampão (com ou sem CaCl2) por três dias, sendo então avaliadas a sobrevivência, massa seca e as características bioquímicas. O cálcio aumentou a sobrevivência ao alagamento de plântulas com quatro dias, entretanto, não influenciou naquelas com dois dias.... |

Two α -galactosidase (P1 and P2) and one invertase present in the culture of Aspergillus terreus grown on wheat straw for 168 h at 28ºC were partially purified by gel filtration and hydrophobic interaction chromatographies. Optimum pH and temperatures for P1, P2 and invertase preparations were 4.5-5.0, 5.5 and 4.0 and 60, 55 and 65ºC, respectively. The K M app for Ï -nitrophenyl-α -D-galactopyranoside were 1.32 mM and 0.72 mM for P1 and P2, respectively, while the K M app value for invertase, using sacarose as a substrate was 15.66 mM. Abstract The identification of yeasts isolated from spoiled Jubileu peach puree using the API 20C AUX method and a commercial yeast as witness were studied. Subsequently, the yeast’s growth potential using two batch culture treatments were performed to evaluate number of colonies (N), reducing sugar concentration (RS), free-invertase (FI), and culture-invertase activity (CI). Stock cultures were maintained on potato dextrose agar (PDA) slants at 4 °C and pH 5 for later use for batch-culture (150 rpm) at 30°C for 24 h, then they were stored at 4 °C for subsequent invertase extraction. Conversion of sucrose into fructose and gluconic acid using invertase, glucose oxidase and catalase was studied by discontinuous (sequential or simultaneous addition of the enzymes) and continuous (simultaneous addition of the enzymes in a 100 kDa-ultrafiltration membrane reactor) processes. The following parameters were varied: concentration of enzymes, initial concentration of substrates (sucrose and glucose), pH, temperature and feeding rate (for continuous process). The highest yield of conversion (100%) was attained through the discontinuous (batch) process carried out at pH 4.5 and 37 ºC by the sequential addition of invertase (14.3 U), glucose oxidase (10,000 U) and catalase (59,000 U). |

The filamentous fungus Aspergillus caespitosus was a good producer of intracellular and extracellular invertases under submerged (SbmF) or solid-state fermentation (SSF), using agroindustrial residues, such as wheat bran, as carbon source. The production of extracellular enzyme under SSF at 30ºC, for 72h, was enhanced using SR salt solution (1:1, w/v) to humidify the substrate. The extracellular activity under SSF using wheat bran was around 5.5-fold higher than that obtained in SbmF (Khanna medium) with the same carbon source. Aspergillus niger - 245, a strain isolated from soil samples showed good β-fructosidase activity when inoculated in medium formulated with dahlia extract tubers. The enzyme was purified by precipitation in ammonium sulphate and percolated in DEAE-Sephadex A-50 and CM-cellulose columns, witch showed a single peack in all the purification steps, maintaining the I/S ratio between 0.32 to, 0.39. Optimum pH for inulinase activity (I) was between 4.0 - 4.5 and for invertase activity (S) between 2.5 and 5.0.
