Antigenic stimulation is more efficient than LPS in inducing nitric oxide production by human mononuclear cells on the in vitro granuloma reaction in schistosomiasis BJMBR
Oliveira,D.M.; Silva-Teixeira,D.N.; Araújo-Filho,R.; Goes,A.M..
Nitric oxide (NO) is an extremely important and versatile messenger in biological systems. It has been identified as a cytotoxic factor in the immune system, presenting anti- or pro-inflammatory properties under different circumstances. In murine monocytes and macrophages, stimuli by cytokines or lipopolysaccharide (LPS) are necessary for inducing the immunologic isoform of the enzyme responsible for the high-output production of NO, nitric oxide synthase (iNOS). With respect to human cells, however, LPS seems not to stimulate NO production in the same way. Addressing this issue, we demonstrate here that peripheral blood mononuclear cells (PBMC) obtained from schistosomiasis-infected patients and cultivated with parasite antigens in the in vitro granuloma...

Dimethyl sulfoxide effects on the febrile response to lipopolysaccharide injection in rabbits Arq. Bras. Med. Vet. Zootec.
Souza,A.H.; Valadão,C.A.A.; Selegatto,M.A.; Almeida,R.M.; Macari,M..
The present study was designed to determine whether DMSO causes an inhibition on the development of fever in rabbits. The intravenous administration of LPS (1.5µg.kg-1 body weight) caused fever in both saline+LPS and DMSO+LPS group, but the onset and magnitude of the induced fever were significantly different. The saline+LPS group presented a prototypic biphasic fever whereas the DMSO+LPS group presented an attenuated febrile response, but it was not abolished. These results suggest that DMSO may provide a protective mechanism against pyrogen LPS, probably through the modulation of NF-kB mediated events, such as fever.

Effect of a rosmarinic acid supplemented hemodialysis fluid on inflammation of human vascular endothelial cells BJMBR
Wang,W-J.; Cheng,M-H.; Lin,J-H.; Weng,C-S..
Chronic systemic inflammation and repetitive damage of vascular endothelia by incompatible dialysis system are probable causes of cardiovascular disease in patients on dialysis. The present study aimed to assess in vitro biocompatibility and anti-inflammatory effect of hemodialysis fluid supplemented with rosmarinic acid (RA) using human umbilical vein endothelial cells (HUVEC). HUVECs (5×106 cells/mL) were pre-exposed to 1 μg/mL of lipopolysaccharides (LPS) and incubated with RA-supplemented hemodialysis fluid (HDF). Cytotoxicity was assessed qualitatively by morphologic assessment and quantitatively by MTT assay. Expressions of proinflammatory mediators were assessed using quantitative real-time PCR and production of NO was quantified. Phosphorylation of...

Effect of different methods of sterilization on the inactivation of bacterial endotoxin (LPS) in endodontic files BJM
Silva,Raquel Assed Bezerra da; Leonardo,Mário Roberto; Faccioli,Lúcia Helena; Medeiros,Alexandra Ivo; Nelson-Filho,Paulo.
This study evaluated the presence of lipopolysaccharide (LPS) in endodontic files after in vitro contamination with E. coli LPS and sterilization in dry heat or wet heat, with or without previous immersion in a Ca(OH)2 suspension. LPS was quantified using the Kinetic QCL TM test and data were analyzed statistically by the t-test. LPS was quantified only in the contaminated group, not submitted to any immersion or sterilization procedure (0.262±0.1296 endotoxin units/mL) (p=0.0003). In conclusion, both wet heat and dry heat sterilization were effective in inactivating LPS without the need of previous immersion of the files in a Ca(OH)2 suspension.

Effect of LPS on the Viability and Proliferation of Human Oral and Esophageal Cancer Cell Lines BABT
Gonçalves,Márcia; Cappellari,Ángelica Regina; Santos Junior,André Avelino dos; Marchi,Fernanda Olicheski de; Macchi,Fernanda Souza; Antunes,Krist Helen; Souza,Ana Paula Duarte de; Morrone,Fernanda Bueno.
The esophagus and mouth tumors are very frequent malignancies worldwide. Lipopolysaccharides (LPS) are capable of regulating gene expression of pro-inflammatory cytokines by binding to toll-like receptor 4 (TLR4). Recent studies show that LPS can increase the migration ability of human esophageal cancer cell line HKESC-2 by increasing its adhesion properties. However, the effect of LPS has not been tested on viability of human esophageal and oral cancer cells. This study aimed to determine the action of LPS on the cell proliferation and viability in OE19 (adenocarcinoma) and OE21 (squamous carcinoma) cell lines, representative of human esophageal cancer, and HN30 cell line, representative of human oral carcinoma. LPS was used as treatment to OE19 and OE21...

Effects of a neuronal nitric oxide synthase inhibitor on lipopolysaccharide-induced fever BJMBR
Perotti,C.A.A.; Nogueira,M.S.; Antunes-Rodrigues,J.; Cárnio,E.C..
It has been demonstrated that nitric oxide (NO) has a thermoregulatory action, but very little is known about the mechanisms involved. In the present study we determined the effect of neuronal nitric oxide synthase (nNOS) inhibition on thermoregulation. We used 7-nitroindazole (7-NI, 1, 10 and 30 mg/kg body weight), a selective nNOS inhibitor, injected intraperitoneally into normothermic Wistar rats (200-250 g) and rats with fever induced by lipopolysaccharide (LPS) (100 µg/kg body weight) administration. It has been demonstrated that the effects of 30 mg/kg of 7-NI given intraperitoneally may inhibit 60% of nNOS activity in rats. In all experiments the colonic temperature of awake unrestrained rats was measured over a period of 5 h at 15-min intervals...

Effects of propofol on lipopolysaccharide-induced expression and release of HMGB1 in macrophages BJMBR
Wang,T.; Wei,X.Y.; Liu,B.; Wang,L.J.; Jiang,L.H..
This study aimed to determine the effects of different concentrations of propofol (2,6-diisopropylphenol) on lipopolysaccharide (LPS)-induced expression and release of high-mobility group box 1 protein (HMGB1) in mouse macrophages. Mouse macrophage cell line RAW264.7 cells were randomly divided into 5 treatment groups. Expression levels of HMGB1 mRNA were detected using RT-PCR, and cell culture supernatant HMGB1 protein levels were detected using enzyme-linked immunosorbent assay (ELISA). Translocation of HMGB1 from the nucleus to the cytoplasm in macrophages was observed by Western blotting and activity of nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) in the nucleus was detected using ELISA. HMGB1 mRNA expression levels increased...

Etude de la réponse immunitaire chez l'huître Crassostrea gigas : caractérisation et fonctions de protéines de reconnaissance aux LPS et d'effecteurs antimicrobiens ArchiMer
Gonzalez, Marcelo.
Research on the immune response of oysters is motivated by the problems of infectious diseases potentially found in the farms. Over the course of the last few years, the development of genomic approaches has allowed us to make headway on characterising genes involved in the cup oyster's (Crassostrea gigas) innate immunity, particularly thanks to various programmes for analysing transcriptome, of type EST (Express Sequence Tag) and subtractive banks. Within the context of this work, over the course of which we have used the data from genomic research and we have also developed biochemical approaches, we were particularly interested in studying the proteins involved in reconnaissance reactions to LPS as well as to antimicrobial effectors. In an earlier part,...

Ginsenoside Rg1 protects human renal tubular epithelial cells from lipopolysaccharide-induced apoptosis and inflammation damage BJMBR
Ni,X.J.; Xu,Z.Q.; Jin,H.; Zheng,S.L.; Cai,Y.; Wang,J.J..
Ginsenoside Rg1, one of the most notable active components of Panax ginseng, has been widely reported to exert anti-inflammatory actions. This study aimed to reveal whether ginsenoside Rg1 also exhibits beneficial roles against lipopolysaccharide (LPS)-induced apoptosis and inflammation in human renal tubular epithelial cells, and to evaluate the potential role of the component on tubulointerstitial nephritis treatment. HK-2 cells were treated with various doses of ginsenoside Rg1 (0, 50, 100, 150, and 200 μM) in the absence or presence of 5 μg/mL LPS. Thereafter, CCK-8 assay, flow cytometry, western blot, migration assay, reactive oxygen species (ROS) assay, and ELISA were carried out to respectively assess cell viability, apoptosis, migration, ROS...

Histopathological analysis of rat mesentery as a method for evaluating neutrophil migration: differential effects of dexamethasone and pertussis toxin BJMBR
Brito,G.A.C.; Falcão,J.L.A.A.; Saraiva,S.N.R.; Lima,A.A.M.; Flores,C.A.; Ribeiro,R.A..
In the present study, histopathological analysis of rat mesentery was used to quantify the effect of two anti-inflammatory agents, dexamethasone (Dex) and pertussis toxin (Ptx), on leukocyte migration. The intravenous injection of Dex (1 mg/kg) and Ptx (1,200 ng) 1 h prior to the intraperitoneal injection of the inflammatory stimuli lipopolysaccharide (LPS) or formyl-methionyl-leucyl-phenylalanine (fMLP) significantly reduced the neutrophil diapedesis (LPS: Ptx = 0.86 ± 0.19 and Dex = 0.35 ± 0.13 vs saline (S) = 2.85 ± 0.59; fMLP: Ptx = 0.43 ± 0.09 and Dex 0.01 ± 0.01 vs S = 1.08 ± 0.15 neutrophil diapedesis/field) and infiltration (LPS: Ptx = 6.29 ± 1.4 and Dex = 3.06 ± 0.76 vs S = 15.94 ± 3.97; fMLP: Ptx = 3.85 ± 0.56 and Dex = 0.40 ± 0.16 vs S = 7.15 ±...

Increased production of tumor necrosis factor-alpha in whole blood cultures from children with primary malnutrition BJMBR
Azevedo,Z.M.A.; Luz,R.A.; Victal,S.H.; Kurdian,B.; Fonseca,V.M.; Fitting,C.; Câmara,F.P.; Haeffner-Cavaillon,N.; Cavaillon,J.-M.; Gaspar Elsas,M.I.C.; Xavier Elsas,P..
Because low tumor necrosis factor-alpha (TNF-alpha) production has been reported in malnourished children, in contrast with high production of TNF-alpha in experimental protein-energy malnutrition, we reevaluated the production of TNF-alpha in whole blood cultures from children with primary malnutrition free from infection, and in healthy sex- and age-matched controls. Mononuclear cells in blood diluted 1:5 in endotoxin-free medium released TNF-alpha for 24 h. Spontaneously released TNF-alpha levels (mean ± SEM), as measured by enzyme immunoassay in the supernatants of unstimulated 24-h cultures, were 10,941 ± 2,591 pg/ml in children with malnutrition (N = 11) and 533 ± 267 pg/ml in controls (N = 18) (P < 0.0001). TNF-alpha production was increased by...

Role of gelatinases MMP-2 and MMP-9 in tissue remodeling following acute lung injury BJMBR
Corbel,M.; Boichot,E.; Lagente,V..
Acute lung injury is characterized by a severe disruption of alveolo-capillary structures and includes a variety of changes in lung cell populations. Evidence suggests the occurrence of rupture of the basement membranes and interstitial matrix remodeling during acute lung injury. The dynamic equilibrium of the extracellular matrix (ECM) under physiological conditions is a consequence of the balance between the regulation of synthesis and degradation of ECM components. Matrix metalloproteinases (MMPs) represent a group of enzymes involved in the degradation of most of the components of the ECM and therefore participate in tissue remodeling associated with pathological situations such as acute lung injury. MMP activity is regulated by proteolytic activation...

Spontaneous neutrophil activation in HTLV-1 infected patients BJID
Guerreiro,Jaqueline B.; Porto,Maria Aurélia F.; Santos,Silvane B.; Lacerda,Lino; Ho,John L.; Carvalho,Edgar M..
Human T cell lymphotropic Virus type-1 (HTLV-1) induces lymphocyte activation and proliferation, but little is known about the innate immune response due to HTLV-1 infection. We evaluated the percentage of neutrophils that metabolize Nitroblue tetrazolium (NBT) to formazan in HTLV-1 infected subjects and the association between neutrophil activation and IFN-gamma and TNF-alpha levels. Blood was collected from 35 HTLV-1 carriers, from 8 patients with HAM/TSP (HTLV-1- associated myelopathy); 22 healthy individuals were evaluated for spontaneous and lipopolysaccharide (LPS)-stimulated neutrophil activity (reduction of NBT to formazan). The production of IFN-gamma and TNF-alpha by unstimulated mononuclear cells was determined by ELISA. Spontaneous NBT levels,...
