Kauffmann,Carla; Giacomin,Ana Caroline; Arossi,Kelen; Pacheco,Leandra Andressa; Hoehne,Lucélia; Freitas,Elisete Maria de; Machado,Gérzia Maria de Carvalho; Cavalheiro,Marilene Marcuzzo do Canto; Gnoatto,Simone Cristina Baggio; Ethur,Eduardo Miranda.
In South American folk medicine members of the genus Myrciaria are used for the treatment of malaria, diarrhoea, asthma, inflammation and post-partum uterine cleansing. The aim of this work was to evaluate its antileishmanial properties (in vitro) of essential oil derived from leaves of Myrciaria plinioides D. Legrand, a plant species that is native in South of Brazil. The essential oil was obtained by hydro-distillation using fresh leaves of M. plinioides. The chemical composition of this essential oil (MPEO, M. plinioides essential oil) was determined by gas chromatography coupled to mass spectrometry (GC-MS). MPEO was assayed in vitro for antileishmanial properties against promastigotes of Leishmania amazonensis and Leishmania infantum, and for...

Avaliação da confiabilidade entre dois observadores em exames citopatológico e imunocitoquímico de aspirado de medula óssea no diagnóstico da leishmaniose visceral canina Arq. Bras. Med. Vet. Zootec.
Vasconcelos,T.C.B.; Machado,L.C.; Abrantes,T.R.; Menezes,R.C.; Madeira,M.F.; Ferreira,L.C.; Figueiredo,F.B..
ABSTRACT Visceral leishmaniasis is a zoonosis in which the dog appears as the main source of infection in urban areas. Its diagnosis is complex and the cytopathological exam is a fast and cheap alternative to parasite direct visualization and its sensitivity can be increased by immunocytochemistry, though with a higher cost. The accuracy of such methods is dependent on the microscopist's experience and therefore, this study evaluated the reliability of such techniques between two observers, from bone marrow aspirates of 50 dogs from an endemic area for the disease. The parasitological culture in Novy-MacNeal-Nicolle medium was used as the reference standard. Among the main findings, the sensitivities obtained by observers I and II were respectively 62.5%...

Xavier,S.C.; Chiarelli,I.M.; Lima,W.G.; Gonçalves,R.; Tafuri,W.L..
A remarkable histopathological picture of one asymptomatic dog naturally infected with Leishmania infantum (syn. chagasi) has been presented. Intracellular parasites were ease found in macrophages of all exanimated organs, especially in skin. Embedded paraffin tissues of liver, spleen, axillary and popliteal lymph nodes, and skin (ear, muzzle and abdomen) were stained by hematoxylin and eosin and by immunocytochemical reaction (streptoavidin-peroxidase method) to detect parasites. All organs showed an intense parasitism associated to severe pathological changes. All lymph nodes had conspicuous histological architecture alterations. Lymphocytes were replaced by macrophages stuffed with an intense number of amastigotes forms of Leishmania. The lymphoid...

Silva,L.C.; Assis,V.P.; Ribeiro,V.M.; Tafuri,W.L.; Toledo Júnior,J.C.; Silva,S.O.; Melo,M.N.; Rachid,M.A.; Valle,G.R..
Considering the venereal transmission of visceral leishmaniasis from dogs to bitches, the aim of this study was to verify if the penile surface and smegma from infected dogs can be the source of parasites in bitches. Twelve Leishmania infantum infected dogs had semen and smegma samples collected for submission to PCR identification of the DNA of the parasite. Semen (41.7%) and smegma (50.0%) have similar positive incidence (P>0.05; Fisher's exact test), with 58.3% of the dogs positive for semen and/or smegma samples. The proportion of positivity for both semen and smegma was 33.3%, but 8.3% was positive only for semen, and 16.7% only for smegma, revealing a moderate agreement between tests (K=0.5; Kappa index). It was concluded that Leishmania infantum...

Leishmania infantum heat shock protein 83 for the serodiagnosis of tegumentary leishmaniasis BJMBR
Celeste,B.J.; Angel,S.O.; Castro,L.G.M.; Gidlund,M.; Goto,H..
The serologic assay is an important tool in the diagnosis of leishmaniasis. One of the most commonly used tests is enzyme-linked immunosorbent assay (ELISA). Since total Leishmania promastigotes are used as antigen in the routine assay, false-positive reactions are frequent due to cross-reaction with sera from other diseases, mainly Chagas' disease. Therefore, an antigen that determines less cross-reactivity has been pursued for the serodiagnosis of leishmaniasis. In the present study we analyzed the use of recombinant Leishmania infantum heat shock protein (Hsp) 83 in ELISA for the serodiagnosis of cutaneous (N = 12) and mucocutaneous leishmaniasis (N = 14) and we observed the presence of anti-L. infantum Hsp 83 antibodies in all samples as well as...

Miltefosine induces metacaspase and PARP genes expression in Leishmania infantum BJID
Khademvatan,Shahram; Gharavi,Mohammad Javad; Saki,Jasem.
OBJECTIVES: Apoptosis is the process of programmed cell death (PCD) that occurs in both animal and plant cells. Protozoan parasites possess metacaspase and these caspase-related proteases could be involved in the PCD pathways in these organisms. Therefore we analyzed the activities of metacaspase and PARP genes in Leishmania infantum (MCAN/IR/96/LON49) treated with miltefosine. MATERIALS AND METHODS: Anti-leishmania activity of miltefosine was studied by treatment of cultured promastigotes with various concentration of miltefosine. MTT assay and Annexin-V FLUOS staining by using FACS flow cytometry methods were used. Cytotoxic potential of HePC on the amastigots of L.infantum was evaluated in J774 cell line. In addition, metacaspase and PARP genes...

Synovial fluid as an auxiliary diagnostic tool for different stages of canine visceral leishmaniasis Ciência Rural
Rennó,Mário César; Vasconcellos,Amanda Otoni; Santos,Rogerio Rodrigues; Silva,Rodrigo Otávio Silveira; Carneiro,Rubens Antônio; Paes,Paulo Ricardo de Oliveira.
ABSTRACT: Leishmaniasis represents a complex of chronic diseases with a broad geographic distribution and a high significance in public health worldwide. The varied clinical signs in conjunction with the low sensitivity and specificity of canine visceral leishmaniasis (CVL) detection methods make diagnosis of the disease complex. Among the several available laboratory tests, studies have suggested that the detection of parasites in synovial fluid (SF) is a good auxiliary tool in the diagnosis of CVL. However, no study has evaluated the influence of the clinical stage of CVL in the detection of Leishmania sp. in SF. This study aimed to evaluate the detection of Leishmania sp. amastigotes in the SF of dogs at different stages of the disease. The negative...
