Sapindus saponaria (soapberry) is a species that presents a great diversity of chemical compounds, such as saponins; however, few studies have examined the allelopathic effect of this species. Therefore, this study provides an evaluation of the allelopathic potential of aqueous extracts of the roots and mature leaves of S. saponaria on the germination of diaspores and seedlings growth of lettuce (Lactuca sativa) and onion (Allium cepa). The aqueous extract was prepared in the proportion of 100 g of dry plant material in 1,000 mL of distilled water (a concentration of 10% w v-1), and diluted with distilled water to 7.5, 5.0 and 2.5% concentrations. The search for decision-making tools is essential in order to control agricultural practices and plan for sustainable agriculture that respects the environment. To this end, FAO has developed an evaluation model called AquaCrop, which covers most technical routes, including fertilization. In this perspective, this study focuses on the AquaCrop model (v6.1) to calibrate it for the simulation of lettuce crop yields (Lactuca sativa L), under different fertilization levels for the trial periods (2015/2016) and (2016/2017). Efetuou-se a clonagem e seqüenciamento do gene que codifica a proteína capsidial de dois isolados do vírus do mosaico da alface (Lettuce mosaic virus, LMV) provenientes do estado de São Paulo, previamente caracterizados como pertencentes aos patótipos II (AF198, incapaz de infetar cultivares com os genes de resistência mo1¹ ou mo1²) e IV (AF199, capaz de quebrar a resistência propiciada pelos genes mo1¹ e mo1²), com base na virulência em cultivares diferenciadoras. Avaliou-se cultivares de alface americana transplantadas para o ampo em março de 2002, no município de Santana da Vargem, MG. Foi conduzido um experimento contendo oito cultivares, no delineamento em blocos casualizados com três repetições. Foram avaliados a massa fresca total e comercial por planta, comprimento de caule, diâmetro da cabeça e sanidade das folhjas externas. Quanto à característica massa fresca total, houve melhor desempenho da cultivar Raider, com 1075,5 g planta-1, sendo estatisticamente superior às demais cultivares. A alface (Lactuca sativa L.), pertencente à família Asteraceae, é uma planta originária da Ásia e trazida para o Brasil pelos portugueses no século 16. É a hortaliça folhosa de maior consumo no país, apresentando bons teores em vitaminas A e C, e minerais como o ferro e o fósforo. A Multiplex Polymerase Chain Reaction (PCR) assay to be used as an alternative to the conventional culture method in detecting Shigella and enteroinvasive Escherichia coli (EIEC) virulence genes ipaH and ial in lettuce was developed. Efficacy and rapidity of the molecular method were determined as compared to the conventional culture. Lettuce samples were inoculated with different Shigella flexneri concentrations (from 10 CFU/ml to 10(7) CFU/ml). DNA was extracted directly from lettuce after inoculation (direct-PCR) and after an enrichment step (enrichment PCR). During surveys undertaken from 1998 to 2003 in the major vegetable growing areas of the city of São Paulo green belt, lettuce (Lactuca sativa) and endive (Cichorium endivia) plants were observed, which showed chlorotic thickening of foliar veins, defective growth and, in some cases, failure to form complete heads. Biological and serological [DAS-Enzyme linked immunosorbent assay (Elisa)] tests together with electron microscope observations, revealed the presence of Lettuce big-vein virus and Mirafiori lettuce virus, in these plants both responsible for the lettuce big-vein syndrome. |
