Caracterización del sistema enzimatico de los hongos comestibles (Pleurotus) como una estrategia de selección de cepas para la producción rural Colegio de Postgraduados

En el presente trabajo se investigo la producción extracelular de celulasas en medio líquido con 12 cepas nativas mexicanas y una comercial de Pleurotus. También se determino el contenido de proteína en los extractos y el peso seco de las colonias. Y se determino el coeficiente de correlación entre las variables producción de celulasa y proteína. Se encontró que el mejor medio de cultivo líquido para la producción de celulasas fue el sugerido por Elisashvili et al (2008a), con algunas modificaciones. Todas las cepas de Pleurotus crecieron sobre el medio de cultivo líquido con celulosa (Solka Floc) como fuente de carbono debido a que pudo producir enzimas celulolíticas para degradar el substrato. Entomopathogenic fungi are biocontrol agents capable to infect many insect pests and can produce different infection propagules in artificial media. The soybean crop suffers with attack of many pests and an ideal option would be the use of a single fungal species for the control of a broad host range of this crop. This study aimed to compare the virulence of conidia and blastospores (fresh or air dried) of Beauveria bassiana (Bals.) The use of microsclerotia (MS) of the entomopathogenic fungi Metarhiziumspp. may be an alternative for pest control due to its long persistence in soil and production of infective conidia in situ. In this study we produce MS of 48 isolates of Metarhiziumspp. and selected three most productive isolates to be used in maize seed coating for two purposes: enhancement of plants growth and protection against Spodoptera frugiperda(Lepidoptera: Noctuidae). The use of the resistant propagules, termed microsclerotia (MS), produced by the nematophagous fungus Purpureocillium lilacinum stands out as an eco-friendly tool for the control of various soil-inhabitant pests. The present study aimed to screen 36 isolates of P. lilacinum from the ESALQ collection of microorganisms for maximum MS production. Three culture media were tested varying Carbon:Nitrogen ratio corresponding to 10:1 for medium named J4 and 50:1 for both media J6 and A1. All liquid cultures were cultivated in baffled flasks (250 mL) containing 45 mL of each medium inoculated with 5 mL of spore suspension. Agro-industrial residues and by-products available in southeastern Brazil were used as ingridients for low-cost culture media for liquid fermentation of Bacillus thuringiensis var. kurstaki. Highest spore yield was obtained with a medium containing cheese whey, soya bean milk and molasses (WSM). Crystals and spores were produced in all media and potency of the final product was highest for nutrient broth + yeast extract medium (NBY). There was no correlation between the number of spores in the fermented media and the potency of the preparations. Considering all three factors, the potencies, costs and yields of the final products, lowest relative cost was obtained with BMM medium (Bombyx mori pupae + molasses). Abstract Production of a bioherbicide for biological control of weeds requires a series of steps, from selection of a suitable microbial strain to final formulation. Thus, this study aimed to select fungi for production of secondary metabolites with herbicidal activity using biological resources of the Brazilian Pampa biome. Phytopathogenic fungi were isolated from infected tissues of weeds in the Pampa biome. A liquid synthetic culture medium was used for production of metabolites. The phytotoxicity of fungal metabolites was assessed via biological tests using the plant Cucumis sativus L., and the most promising strain was identified by molecular analysis.
