Zasmidium angulare, a novel species of Mycosphaerellaceae, and several novel taxa that reside in Dissoconiaceae, were identified from a collection of apples and Cucurbita maxima (cv. Blue Hubbard) from China and the USA that exhibited sooty blotch and flyspeck (SBFS) signs on their host substrata. Morphology on fruit surfaces and in culture, and phylogenetic analyses of the nuclear ribosomal DNAs 28S and internal transcribed spacer regions, as well as partial translation elongation factor 1-alpha gene sequences in some cases, were used to delineate seven previously unidentified species and three known species. The objetive of this work was to rescue immature embryos of apple rootstocks Malus prunifolia (Marubakaido) and Malus pumila (M9) after 40-60 days of pollination and to put them into MS culture media supplemented with agar (6 g L-1) and casein hydrolysate (500 mg L-1). Embryos originated from interspecific crosses and open pollination showed differences in the in vitro responses, depending on the female parent, the developmental stage of the embryo, and the culture medium composition. Recent studies have found a wide range of ascomycetes to be associated with sooty blotch and flyspeck (SBFS) blemishes on the surfaces of pomaceous fruits, specifically apples. Based on collections of such fungi from apple orchards in Germany and Slovenia we introduce two novel genera according to analyses of morphological characters and nuclear ribosomal DNA sequences (large subunit and internal transcribed spacer regions). O ágar é um dos componentes mais caros do meio de propagação in vitro. Visando a reduzir custos, os estudos foram desenvolvidos no Laboratório de Cultura de Tecidos da EMBRAPA/CPACT, Pelotas RS, objetivando-se testar a ação do ágar e da sua substitução parcial pelo amido de mandioca na composição do agente solidificante (AS) do meio MS, associados às concentrações de 0; 2,5; 5,0; 7,5 e 10µM de ácido indolbutírico (AIB). Os dois AS de meio de cultivo testados: AS1 - 6g/l de ágar e; AS2 - combinação de 3g/l de ágar mais 50g/l de amido de mandioca (fécula), tiveram redução de Vi dos sais de MS e acréscimos de 100mg/l de mio-inositol e 30g/l de sacarose, juntamente com pH ajustado em 5,9 antes da autoclavagem. O trabalho teve por objetivo estudar o enraizamento in vitro do porta-enxerto 'Marubakaido' (Malus prunifolia Borkh), visando a diminuir os efeitos prejudiciais causados pelo excesso de auxina. Foram utilizadas brotações do porta-enxerto 'Marubakaido' com três pares foliares (2,5 a 3 cm), oriundas do processo de multiplicação in vitro. O meio utilizado foi o MS/2 acrescido de l00mg/l de mio-inositol, 30 g/l de sacarose e 6g/l de ágar. O pH foi ajustado para 5,9, antes da colocação do ágar. Abstract: The objective of this work was to identify rootstocks with competitive agronomic potential to substitute those currently used in Southern Brazil for the Galaxy and Fuji Suprema apple scion cultivars cultivated on replanting soils. The experiment was carried out in the municipality of Lebon Regis, in the state of Santa Catarina, Brazil, from the 2009/2010 to the 2014/2015 crop season, in a randomized complete block design, with four replicates. The yield performance of the 'Galaxy' and 'Fuji Suprema' scions grafted on the G.056, G.202, G.213, G.814, G.896, G.969, M.9, and Marubakaido/M.9 rootstocks was evaluated.
