A di-O-dihydrogeranylgeranyl glycerol from Thermococus S 557, a novel ether lipid, and likely intermediate in the biosynthesis of diethers in Archaea ArchiMer

The lipids of a deep-sea hydrothermal vent arch ae on, Thermococcus S 557, were isolated, purified and structurally determined. Based on acid methanolysis and spectroscopic studies, the polar lipids were shown to comprise diphytanyl glycerol and dibiphytanyl diglycerol, typical membrane lipids of Arch aea. From the neutral lipids, 2,3-di-O-dihydro-14,15-geranylgeranyl glycerol was isolated. This novel ether lipid is, very likely, a close intermediate in the biosynthesis of diphytanyl glycerol diether in Arch aea.

A sensitive bioanalytical method development and validation of cabozantinib in human plasma by LC-ESI-MS/MS BJPS

ABSTRACT A simple, sensitive and specific liquid chromatography-tandem mass spectrometry (LC-MS/MS) method was developed for the quantification of cabozantinib (CZ) in human plasma using cabozantinib-d4 (CZD4) as an internal standard (IS). Chromatographic separation was performed on Xbridge C18, 50 x 4.6 mm, 5 mm column with an isocratic mobile phase composed of 10mM Ammonium formate and Methanol in the ratio of (20:80 v/v), at a flow-rate of 0.7 mL/min. Although Glycosyl-Inositol-Phospho-Ceramides (GIPCs) are the main sphingolipids of plant tissues, they remain poorly characterized in term of structures. This lack of information, notably with regard to polar heads, currently hampers the understanding of GIPC functions in biological systems. This situation prompted us to undertake a large scale-analysis of plant GIPCs: 23 plant species chosen in various phylogenetic groups were surveyed for their total GIPC content. GIPCs were extracted and their polar heads were characterized by negative ion MALDI and ESI mass spectrometry. Aspergillus is a large genus, with a complex and ever evolving taxonomy. Section Flavi is one of the most significant Sections in the Genus. Taxonomy and species identification is subject of great interest for scientists aiming to clarify the species concept and limits within the section. Furthermore, this Section comprises both toxigenic and non-toxigenic species/strains, with great interest to biotechnology and food industry. Various genes, namely the rRNA (internal transcribed spacer (ITS) and partial LSU-rRNA regions), calmodulin and ~-tubulin genes, have been widely reported as good markers for Aspergillus species identification, because they are rapid and cost-effective.

Vesicomyids live in endosymbiosis with sulfur-oxidizing bacteria and therefore need hydrogen sulfide to survive. They can nevertheless live in a wide range of sulfide and oxygen levels and depths, which may explain the exceptional diversity of this clam family in deep-sea habitats. In the Gulf of Guinea, nine species of vesicomyid clams are known to live in cold-seep areas with pockmarks from 600 to 3200 m deep, as well as in the organic-rich sediments of the Congo deep-sea fan at 5000 m deep. Aspergillus is a large genus, with a complex taxonomy. The genus is easily identifiedd by its characteristic conidiophore, but species identification and differentiation is complex, mainly because it is traditionally based on a range of morphological features. Aspergillus subgenus Circumdati section Flavi, also refered to as the A.dflavus group, has attracted worldwide attention for its industrial use and toxigenic potential. The aim of this study was to extract and identify volatile compounds from pineapple residues generated during concentrated juice processing. Distillates of pineapple residues were obtained using the following techniques: simple hydrodistillation and hydrodistillation by passing nitrogen gas. The volatile compounds present in the distillates were captured by the solid-phase microextraction technique. The volatile compounds were identified in a system of high resolution gas chromatography system coupled with mass spectrometry using a polyethylene glycol polar capillary column as stationary phase. The pineapple residues constituted mostly of esters (35%), followed by ketones (26%), alcohols (18%), aldehydes (9%), acids (3%) and other compounds (9%)....

Bacteria from deep-sea hydrothermal vents constitute an attractive source of bioactive molecules. In particular, exopolysaccharides (EPS) produced by these bacteria become a renewable source of both biocompatible and biodegradable molecules. The low molecular weight (LMW) derivatives of the GY785 EPS produced by the deep-sea hydrothermal vent strain Alteromonas infernus have previously displayed some biological properties, similar to those of glycosaminoglycans (GAG), explored in cancer and tissue engineering. Characterization of new phenolic derivatives in Portuguese propolis by electrospray mass spectroscopy IPB - Escola Superior Agrária

This research outlines an extensive characterization of the phenolic composition of a propolis sample from the northeast of Porto gal. For that, ao ethanolic extract was prepared, fractionated by HPLC and the identification of the phenolic compounds was done by electro spray mass spectrometry in the negative mode. This technical approach allowed the identification of 37 phenolic compounds in the Portoguese propolis sample, including seven that were described for the first time. Two of these new compounds had [M-Hr ions at mlz 403, aod the others had [M-Hr ions at mlz 433, mlz 461, mlz 417, mlz 475, and mlz 565. Convenient preparation of picolinyl derivatives from fatty acid esters ArchiMer

It is essential to have simple rapid methods for the determination of fatty acid structures. Traditionally, fatty acids are analysed by gas chromatography using their methyl ester derivatives (FAME). However, their corresponding mass spectra exhibit molecular ions but are usually devoid of ions indicative of structural features and, notably, the position of double bounds on the aliphatic chains [1]. In the most useful approach to structure determination, the carboxyl group is derivatised with a reagent containing a nitrogen atom. Recently, a convenient method for preparing picolinyl esters from intact lipids has been published [2]. The epi-benthic dinoflagellates Gambierdiscus and Fukuyoa produce ciguatoxins (CTXs) and maitotoxins (MTXs), which are among the most potent marine toxins known. CTXs are bio-accumulated and bio-transformed along the marine food chain and cause Ciguatera Fish Poisoning (CFP), the most common non-bacterial foodborne intoxication worldwide.
