A method to assess algicidal activity of microalgal extracts coupling microalgae produced in stirred closed photobioreactor operating in continuous with pulse amplitude modulated (PAM) fluorometry ArchiMer
Menguy, Eva; Dumontet, Vincent; Coulombier, Noémie; Meriot, Vincent; Le Dean, Loic; Barthelemy, Vanille; Jauffrais, Thierry.
We describe in the present study a quick and reliable method based on chlorophyll a fluorescence to assess putative algicidal effect of different microalgal extracts. We couple microalgal production under chemostat cultivation mode to continuously produce a given microalgae species (e.g. Dunaliella salina in this study) at a stable physiological state to ease comparison between extracts tested; with a non-destructive method based on chlorophyll a fluorescence. Pulse amplitude modulated (PAM) fluorometry was used to assess over time the effect of different microalgal crude extracts on the efficiency of the photosystem II (PSII) of a tested microalgae (Dunaliella salina). • Microalgal production at stationary phase in stirred closed photobioreactor (PBR)...

A Physiologically Structured Equation to Consider Quota Heterogeneity in the Droop Model ArchiMer
Mairet, Francis; Baron, Regis.
The Droop model allows to represent microalgae growth limited by a nutrient, using a cell quota (also referred to as variable-yield) approach. Single-cell measurements have revealed quota heterogeneity in phytoplankton collected from field studies. Such heterogeneity can be due, among other factors, to spatial structure (e.g. in biogeochemical cycles in the ocean, or for photobioreactors connected in series). Nonetheless, quota heterogeneity is generally omitted in modelling studies, using an average quota approach, or included in size-structured or individual-based models. Here, we propose a distributed Droop equation to tackle this problem, considering subpopulation growth -in line with Droop macroscopic view- rather than cell division dynamics. We...

Algicidal effects of aqueous leaf extracts of neem (Azadirachta indica) on Scenedesmus quadricauda (Turp.) de Brébission Acta Botanica
Chia,Mathias Ahii; Akinsanmi,Jamiu Taiwo; Tanimu,Yahuza; Ladan,Zakari.
The application of synthetic algaecides for the control of algae produces by-products that are sometimes toxic to the environment. There is a need for natural and cheap alternatives to synthetic algaecides. In the present study, we investigated the potential of aqueous crude extract of Azadirachta indica to inhibit the growth of Scenedesmus quadricauda. Phytochemical screening of the extract revealed the presence of groups of bioactive compounds that are capable of inhibiting microalgal growth. Chlorophyll a concentration, dry weight production and cell density of microalga decreased with increasing crude extract concentration. After three days of exposure, the 1000 mg/L extract concentration resulted in complete growth inhibition and cell lysis....

Allocation du carbone et métabolisme azoté chez l'haptophyte Tisochrysis lutea ArchiMer
Garnier, Matthieu.
The aim of this thesis is to improve knowledge on mechanisms involved in the response to nitrogen limitation and in lipid accumulation in the microalgae haptophyte Tisochrysis lutea. The wild type strain and a lipid accumulating mutant strain were grown under different nitrogen limitation and starvation and analyzed by functional genomics. Four genes of high-affinity nitrate/nitrite transporter (Nrt2) were identified and characterized to reveal the mechanisms involved in mineral absorption in this species. Transcriptomes of both strains were sequenced and proteins affected by nitrogen starvation and differentially expressed between the two strains were identified. We so identified the functions regulated by nitrogen deficiency and potentially involved in...

Anti-Biofilm Effect of Biodegradable Coatings Based on Hemibastadin Derivative in Marine Environment ArchiMer
Le Norcy, Tiffany; Niemann, Hendrik; Proksch, Peter; Linossier, Isabelle; Vallee-rehel, Karine; Hellio, Claire; Fay, Fabienne.
Dibromohemibastadin-1 (DBHB) is an already known potent inhibitor of blue mussel phenoloxidase (which is a key enzyme involved in bioadhesion). Within this study, the potentiality of DBHB against microfouling has been investigated. The activity of DBHB was evaluated on key strains of bacteria and microalgae involved in marine biofilm formation and bioassays assessing impact on growth, adhesion and biofilm formation were used. To assess the efficiency of DBHB when included in a matrix, DBHB varnish was prepared and the anti-microfouling activity of coatings was assessed. Both in vitro and in situ immersions were carried out. Confocal Laser Scanning Microscopy (CLSM) was principally used to determine the biovolume and average thickness of biofilms developed...

Heterocapsa triquetra (Ht), a non-toxic dinophyte, was selected to investigate the presence of pigments inhibiting melanoma cells growth. We developed an innovative microwave-assisted extraction process (MAE), determined the Ht pigment composition using Reverse Phase-High Performance Liquid Chromatography (HPLC) and high-resolution Ultra Performance Liquid Chromatography-Mass Spectrometry E (UPLC-MSE) analysis and assessed their antiproliferative activity in human invasive melanoma cells (A2058) grown in vitro. MAE allowed a fast and efficient extraction of Ht pigments as compared with conventional processes (soaking, sonication). Seventeen pigments and derivatives were unequivocally identified by UPLC-MSᴱ in the Ht extract, among which two were supposed...

The search for novel compounds of marine origin has increased in the last decades for their application in various areas such as pharmaceutical, human or animal nutrition, cosmetics or bioenergy. In this context of blue technology development, microalgae are of particular interest due to their immense biodiversity and their relatively simple growth needs. In this review, we discuss about the promising use of microalgae and microalgal compounds as sources of natural antibiotics against human pathogens but also about their potential to limit microbial infections in aquaculture. An alternative to conventional antibiotics is needed as the microbial resistance to these drugs is increasing in humans and animals. Furthermore, using natural antibiotics for...

Baudelet, Paul-hubert; Gagez, Anne-laure; Berard, Jean-baptiste; Juin, Camille; Bridiau, Nicolas; Kaas, Raymond; Thiery, Valerie; Cadoret, Jean-paul; Picot, Laurent.
The glaucophyte Cyanophora paradoxa (Cp) was chemically investigated to identify pigments efficiently inhibiting malignant melanoma, mammary carcinoma and lung adenocarcinoma cells growth. Cp water and ethanol extracts significantly inhibited the growth of the three cancer cell lines in vitro, at 100 mu g.mL(-1). Flash chromatography of the Cp ethanol extract, devoid of c-phycocyanin and allophycocyanin, enabled the collection of eight fractions, four of which strongly inhibited cancer cells growth at 100 mu g.mL(-1). Particularly, two fractions inhibited more than 90% of the melanoma cells growth, one inducing apoptosis in the three cancer cells lines. The detailed analysis of Cp pigment composition resulted in the discrimination of 17 molecules, ten of...

Bacterial community structure of the marine diatom Haslea ostrearia ArchiMer
Lepinay, Alexandra; Capiaux, Herve; Turpin, Vincent; Mondeguer, Florence; Lebeau, Thierry.
Haslea ostrearia produces a water-soluble, blue-green pigment, called marennine, with proven economic benefits (as a bioactive compound used to green oysters, which improves their market value). Incomplete knowledge of the ecological features of this marine diatom complicates its cultivation. More specifically, the ecology of bacteria surrounding H. ostrearia in ponds is what remains unknown. The structure of this bacterial community was previously analyzed by means of PCR-TTGE before and after isolating H. ostrearia cells recovered from 4 localities in order to distinguish the relative parts of the biotope and biocenose and to describe the temporal dynamics of the bacterial community structure at two time scales (2 weeks vs. 9 months). The bacterial...

Bioethanol from Microalgal Biomass: A Promising Approach in Biorefinery BABT
Silva,Carlos Eduardo de Farias; Bertucco,Alberto.
Abstract The development of new technologies which increase the production of biofuel without directly compete with food production is required. Microalgal biomass has recently been in the highlight. The role of this biomass is here discussed within the concept of biorefinery and industrial sustainability of bioethanol production. The process of cultivation in order to accumulate around 50% of carbohydrates in the biomass (dry weight) and the importance of water and nutrient recycling are reviewed. Saccharification of biomass using enzymes or acids and alternative processes such as hydrothermal liquefaction and flash hydrolysis are addressed. Since the main monosaccharide in microalgal biomass is glucose, high rates of hydrolysis and fermentation were,...

Li, Meng; Callier, Myriam; Blancheton, Jean-paul; Galès, Amandine; Nahon, Sarah; Triplet, Sebastien; Geoffroy, Thibault; Menniti, Christophe; Fouilland, Eric; Roque D'Orbcastel, Emmanuelle.
Integrated multi-trophic aquaculture (IMTA) systems are a promising solution for sustainable aquaculture combining nutrient recycling with increased biomass production. An innovative land-based recirculating aquaculture system (RAS) was studied in France for a 60-day experiment. It combined a European sea bass (Dicentrarchus labrax) RAS with two other production systems: high rate algal ponds (HRAP) with natural marine polyspecific algal assemblages, and oysters in separate open tanks. The objective was the assessment of: 1) the efficiency and the stability of the microalgae bioremediation of the effluent from a fish RAS in spring and summer, 2) the abundance and the diversity patterns of the microalgae biomass for consumption in the oyster compartment of...
