Acclimatization of Tapeinochilos ananassae plantlets in association with arbuscular mycorrhizal fungi PAB

The objective of this work was to assess the potential of three isolates of arbuscular mycorrhizal fungi to promote growth of micropropagated plantlets of Tapeinochilos ananassae during acclimatization. The experiment was carried out in greenhouse, in a completely randomized block design, with four inoculation treatments: non‑inoculated control and plants inoculated with Glomus etunicatum, Acaulospora longula or Gigaspora albida, with ten replicates. After 90 days, the following parameters were evaluated: survival rate, height, leaf and tiller number, leaf area, fresh and dry biomass, contents of macro‑ and micronutrients in the root and shoot, glomerospore number, and mycorrhizal colonization. Acclimatization of 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstock Scientia Agricola

The pre-acclimatization stage can be used to improve micropropagation protocols and increase the yield of produced plants. The influence of sucrose and photon flux density (PFD) levels on the acclimatization of in vitro-grown 'VR043-43' (Vitis vinifera x Vitis rotundifolia) grapevine rootstocks was evaluated. Rooted shoots were obtained from 4-week-old in vitro shoots cultivated in QL (Quoirin and Lepoivre, 1977) culture medium supplemented with 15, 30 and 45 g L-1 of sucrose. No previous studies have investigated the use of graphite instead of activated charcoal in orchids. In this work, different concentrations of activated charcoal or graphite were added to KC medium to darken the culture medium and stimulate the in vitro propagation of Cattleya bicolor and of a double hybrid orchid (‘BLC Pastoral Innocence’). A highly efficient somatic embryogenesis system and subsequent plant regeneration of chinaberry (Melia azedarach L.) was developed. Plants were regenerated from indirect somatic embryogenesis induction. Novel features of this improved protocol, include: a) Embryogenic callus induction with no addition of 2, 4-D in the culture media; b) Somatic embryos differentiation was achieved by using high concentration of cytokinins (BAP 10 mg/L) and adenine; c) 100% conversion of somatic embryos to plants was practically obtained and 100% of plants survived under greenhouse conditions; d) Addition of putrescine improved somatic embryos germination. An efficient protocol for culturing meristems of sorghum hybrids Phyton

A robust protocol for culturing meristems of Sorghum is required to assist with rapid genetic improvement of the genus. Through meristem culture, an efficient method for rapid micropropagation was developed for Sorghum bicolor (L.) Moench. hybrids, namely NC+262, NC+6C21 and NC+6B50. Complete plants were regenerated directly from shoot meristems without an intervening callus phase. Regeneration frequencies varied between the studied genotypes and according to the growth regulator combinations present in the medium. The combination of BAP and TDZ showed a synergistic effect on shoot multiplication. This study aimed to establish and propagate in vitro plants of Bouchea fluminensis, a medicinal species known in Brazil as gervão-falso ("false verbena"), evaluating the influences of different growth regulators on in vitro multiplication and rooting stages, as well as examining ex vitro acclimatization of rooted plants. Explants were established on Murashige and Skoog medium at half strength of salts and vitamins without growth regulators. ANATOMICAL STUDIES OF IN VITRO ORGANOGENESIS INDUCED IN LEAF-DERIVED EXPLANTS OF PASSIONFRUIT PAB

With the aim of studying the organogenesis in vitro in Passiflora edulis Sims f. flavicarpa Deg., the passionfruit, leaf-derived explants were cultured on media containing NAA or BAP and incubated either in continuous darkness or in light. The histological events leading to de novo organ formation were evaluated. Darkness induces rhizogenesis in the presence of NAA, whereas direct shoot regeneration is stimulated by light and BAP. This latter condition is recommended for passionfruit micropropagation as several adventitious shoot buds were formed from meristemoids of parenchymal origin.

Antimicrobial activity of the essential oil (EO) of Lippia gracilis Schauer over fungi and heliconia endophytic bacteria was evaluated. For the fungi Geotrichum candidum; Trichoderma viride; Torula herbarum; Paecillomyces sp.; Fusicoccum sp.; P. aeruginens; Curvularia lunata; Aspergillus nidulans; A. flavus; and A. niger mycelial discs were inoculated in PDA medium with oil (0; 420; 440; 460; 480 e 500 µL L-1). The inhibition percentage in 420 µL L-1 was 100% for all fungi, except for C. lunata and A. niger, which was 95.58% and 89.40% , respectively. ABSTRACT An efficient in vitro regeneration protocol for medicinally important herb Swertia chirayita was developed and the genetic fidelity was assessed using RAPD and ISSR markers. The best shoot regeneration was observed on MS basal supplemented with 1.0 mg/L Benzyl amino purine (BAP) in combination with Indole-3-acetic acid (IAA) (0.5 mg/L) that resulted in the increase by multiplication rate (7.65) with an average of 33.33 numbers of shoots and average shoot length of 2.70 cm. Currently, this collection includes 32 high quality clones, selected for their good bearing and healthy state. Most of them belong to different discrete local populations. The objective of this work was to characterize the biochemically systemic bacterial isolated from banana plants, to evaluate the bacterial sensitivity to antibiotics, and to determine the phytotoxicity of banana shoots during in vitro proliferation. Systemic bacteria belonging to the Klebsiella and Aeromonas genera were isolated from the “Maravilha” (FHIA 01 AAAB), “Preciosa” (PV 4285 AAAB) and “Thap Maeo” (AAB) varieties and were then characterized. Laelia halbingeriana pertenece a la familia Orchidaceae y debido a su descubrimiento relativamente reciente, no se ha caracterizado morfológica, cromosómica ni molecularmente. Con la finalidad de conocer la diversidad de esta especie y su posible relación con L. anceps, L. superbiens y L. autumnalis, la caracterización molecular mediante el análisis de RAPDs se utilizaron 16 iniciadores, cuatro iniciadores mostraron productos de amplificación aceptable. Se observó 60.97 % de polimorfismo que denota gran variabilidad entre las especies de Laelia. Por otra parte se observó que el número cromosómico más común es de 2n=36. Coriander is commonly used for medicinal purposes, food applications, cosmetics and perfumes. Herein, the production of antioxidants in vegetative parts (leaves and stems) of in vivo and in vitro grown samples was compared. In vitro samples were clone A- with notorious purple pigmentation in stems and leaves and clone B- green. Seeds were also studied as they are used to obtain in vivo and in vitro vegetative parts. Lipophilic (tocopherols, carotenoids and chlorophylls) and hydrophilic (sugars, ascorbic acid, phenolics, flavonols and anthocyanins) compounds were quantified. ABSTRACT Nidularium procerum and Nidularium innocentii (Bromeliaceae) were cultivated in vitro on media supplemented with different sources and levels of GA3 (gibberellic acid). These sources were the commercial powder (analytical degree) and fermented extract obtained by Fusarium moniliforme via solid state fermentation. The in vitro elongation and rooting of these plants were evaluated after 50 days of cultivation. The GA3 present in the fermented extract possess the same effect of purified GA3 (analytical degree) for the increase of the height of aerial part of shoots of N. innocentii, but not for the N. procerum being the GA3 fermented extract in a lesser degree. Para el establecimiento in vitro de Stevia rebaudiana se aplicaron cinco tratamientos de desinfestación y se obtuvo al usar Cloro® (1.8 % cloro activo), Ridomil y Fungimycin previo a la siembra in vitro y al agregar al medio Cefotaxima (500 mgL-1) se obtuvo un porcentaje de contaminación de 50 % permitiendo una supervivencia de 47 %.
