AFLP and SRAP markers linked to the mj gene for root-knot nematode resistance in cucumber Scientia Agricola

Root-knot nematodes (Meloidogyne spp.) are an important worldwide pest of cucumber (Cucumis sativus L.). Molecular markers linked to the Javanese root-knot nematode (M. javanica) resistance gene mj in cucumber may aid marker assisted selection. One-hundred AFLP (EcoRI-MseI) and 112 SRAP were used to screen resistant and susceptible parents for polymorphisms to develop molecular markers linked to the mj gene. Of the 100 AFLP primers, 92 produced bands and two yielded candidate markers (E-ATT/M-CAA and E-AAC/M-CTG). These two bands were cut off from polyacrylamide gel, cloned and sequenced. An RNA extraction method with high integrity and purity as well as the selection of adequate reference genes are prerequisites for gene expression analysis. For common bean seeds, there is no well-defined protocol that can be used in a laboratory routine for gene expression analysis. In this study, an extraction protocol for RNA from common bean seeds, which produced material with good integrity for qPCR (RIN ≥ 6.5), was optimized. In addition, 10 reference genes were evaluated under qPCR standard conditions using different tissue samples of common beans. Desenvolvimento de marcadores associados à resistência do bicho-mineiro: seleção e validação de SNPs. Repositório Alice

O objetivo deste estudo é o desenvolvimento de marcadores do tipo SNPs associados com a resistência ao bicho-mineiro em café. A estratégia principal utilizou as análises genômicas de microarranjo, para caracterização de expressão diferencial de genes-candidatos em plantas resistentes infestadas com o bicho-mineiro. Inicialmente, análises in silico de 2137 destes genes identificaram aqueles potencialmente relacionados com a especificidade da resposta de defesa da planta. Uma vez selecionados, o perfil de expressão de 22 genes foi confirmado por qRT-PCR em experimentos utilizando plantas resistentes e suscetíveis, infectadas pelo bicho-mineiro. [[abstract]]Viral diseases of ornamental plants cause major losses in productivity and quality. Host pl1ant resistance offers an effective means of controlling plant diseases caused by viruses. It minimizes the necessity for the application of pesticides. However, there are many ornamentals in which no natural disease resistance is available. Genetic engineering allows the introduction of specific, in some instances broad spectrum , disease resistance derived from other species , or even from the pathogen it self into plant genotypes that have been selected for desirable horticultural properties.

Rice has extensive collections of germplasm, but a small fraction of genotypes are used in breeding programs worldwide. This aimed to assess the heterosis and the combining ability for higher grain yield and earliness, and their correlations to genetic distance among accessions belonging to the Embrapa ́s Rice Core Collection. Crosses were made in complete diallel, without the reciprocals, being phenotyped in generations F2 and F7 for grain yield (GY) and days to flowering (DTF). RESUMO: A identificação de perfis de expressão gênica diferenciais e redes metabólicas é uma ferramenta valiosa para a caracterização genética de caracteres agronômicos. Neste estudo, utilizamos análises de expressão em larga-escala para identificar processos biológicos alterados em plantas com teores reduzidos de cafeína. A expressão diferencial de genes selecionados observada in silico foi então validada em experimentos in vivo. O primeiro passo foi o sequenciamento em larga-escala de RNA extraído de tecidos jovens e em desenvolvimento, de plantas sem cafeína e de plantas com concentrações normais do composto.
