A recessive mutant cell line, B7, which is partially responsive to both interferon (IFN)- a and IFN-g is described. B7 was FACS sorted from a cellular pool, which was obtained from the parental cell line 2C4, after several rounds of mutagenesis. The partial responsiveness to IFN was observed both in terms of expression of cell surface markers (CD2, class I and II HLAs) and mRNA expression of IFN-stimulated genes (9-27; 6-16; 2'-5' OAS; GBP and HLA-DRa). A genetic cross with the U4 mutant (JAK1-, a member of the Janus family of nonreceptor tyrosine kinase) did not restore full IFN-responsiveness to B7, and JAK1 cDNA transfection into B7 restored the wild phenotype of the cell... |

Plant breeding aims to develop cultivars with good agronomic traits through gene recombination and elite genotype selection. To support Coffea arabica breeding programs and assist parent selection, molecular characterization, genetic diversity (GD) analyses, and circulating diallel studies were strategically integrated to develop new cultivars. Molecular markers were used to assess the GD of 76 candidate parents and verify the crossing of potential F1 hybrids. Based on the complementary agronomic traits and genetic distance, eight elite parents were selected for circulating diallel analysis. The parents and 12 hybrids were evaluated based on 10 morpho-agronomic traits. For each trait, the effects of general and specific combining abilities, as well as the... |

The Infectious Bursal Disease Virus (IBDV) causes immunosuppression in young chickens. Advances in molecular virology and vaccines for IBDV have been achieved by viral reverse genetics (VRG). VRG for IBDV has undergone changes over time, however all strategies used to generate particles of IBDV involves multiple rounds of amplification and need of in vitro ligation and restriction sites. The aim of this research was to build the world?s first VRG for IBDV by yeast-based homologous recombination; a more efficient, robust and simple process than cloning by in vitro ligation. The wild type IBDV (Wt-IBDV-Br) was isolated in Brazil and had its genome cloned in pJG-CMV-HDR vector by yeast-based homologous recombination. The clones were transfected into chicken... |

Common beans are a key source of protein and are consumed daily by most of the Brazilian population. More than 70% of what is consumed in this country is classified as carioca beans, based on seed qualities and appearance. We evaluated progenies of carioca common beans of the Brazilian agriculture research agency (Embrapa) recurrent selection program. This recurrent selection program is based on resistance to common bean bacterial blight (CBB), seed yield, and other important agronomic traits. Selection also considers the genetic representativeness of the parents and the genetic diversity among phenotypically selected progenies. Initially, 60 superior progenies were selected based on resistance to CBB and on seed type, in two locations (Santo Antônio de... |

The aim of this work was to study the genetic variability among the wild boars, crossbred animals and pigs using microsatellite markers. Five genetic groups were studied. The fragments of three microsatellites developed for Sus scrofa domestica - IGF1, ACTG2 and TNFB - were amplified through PCR technique to evaluate the expected intra populacion variability (He) and observed (Ho) heterozygosity, and endogamy coefficient (F IS ) within each population and inter population variability F IS , testing relationship among five genetic groups to establish the genetic distance among them. The high level of observed heterozygosity values varied between 0.537 and 0.7871. Generally, F IS was low, suggesting that the endogamy did not exist between the tested animals. |

Rice has extensive collections of germplasm, but a small fraction of genotypes are used in breeding programs worldwide. This aimed to assess the heterosis and the combining ability for higher grain yield and earliness, and their correlations to genetic distance among accessions belonging to the Embrapa ́s Rice Core Collection. Crosses were made in complete diallel, without the reciprocals, being phenotyped in generations F2 and F7 for grain yield (GY) and days to flowering (DTF). It were estimated the varietal effect, mean heterosis, varietal heterosis, specific combining abilityand general combining ability (GCA) of the parents in each generation. For the selection of new lines, the combinations involving the genitors with the highest magnitudes... |

MicroRNAs (miRNAs, miRs) encompass a class of small non-coding RNAs that often negatively regulate gene expression. miRNAs play an essential role in skeletal muscle, determining the proper development and maintenance of this tissue. In comparison to other organs and tissues, the full set of muscle miRNAs and its expression patterns are still poorly understood. In this report, a chicken skeletal muscle miRNA library was constructed, and the expression of selected miRNAs was further characterized during muscle development in chicken lines with distinct muscling phenotypes. Clone library sequence analysis revealed 40 small RNAs with similarities to previously described chicken miRNAs, seven miRNAs that were never identified before in chicken, and some... |

Mutant cell lines B3 and B10, which are unresponsive to both interferon (IFN)-alpha and IFN-gamma, and line B9, which does not respond to IFN-gamma stimulation, are described. The mutants were submitted to fluorescence-activated cell sorting (FACS) from a cellular pool, which was obtained from the parental cell line 2C4 after several rounds of mutagenesis. The unresponsiveness to IFN stimulation was observed both in terms of expression of cell surface markers (CD2, class I and II HLAs) and mRNA expression of IFN-stimulated genes (2'-5' oligoadenylate synthetase (OAS), 9-27, and guanylate binding protein (GBP)). Genetic crossing of B3, B9 and B10 with U3 (STAT1-), gamma2a (JAK2-) and U4 (JAK1-) mutants, respectively, did not restore IFN responsiveness to... |
