A conservative region of the mercuric reductase gene (merA) as a molecular marker of bacterial mercury resistance BJM
Sotero-Martins,Adriana; Jesus,Michele Silva de; Lacerda,Michele; Moreira,Josino Costa; Filgueiras,Ana Luzia Lauria; Barrocas,Paulo Rubens Guimarães.
The most common bacterial mercury resistance mechanism is based on the reduction of Hg(II) to Hg0, which is dependent of the mercuric reductase enzyme (MerA) activity. The use of a 431 bp fragment of a conservative region of the mercuric reductase (merA) gene was applied as a molecular marker of this mechanism, allowing the identification of mercury resistant bacterial strains.

An overview on molecular markers for detection of ochratoxigenic fungi in coffee neans BABT
Fungaro,Maria Helena Pelegrinelli; Sartori,Daniele.
Ochratoxin A (OTA) is one of the most abundant food-contaminating mycotoxins. Its occurrence in several agricultural commodities has been considered a worldwide problem. This toxin is mainly produced by Aspergillus species. OTA has nephrotoxic, immunosuppressive, and carcinogenic effects and consequently the contamination with this toxin represents a high risk for human and animal health. In the last 5 years, several investigators have applied molecular methods in order to develop PCR assays for identifying and quantifying OTA-producing fungi in coffee beans samples. The main objective is to allow the detection of microorganisms capable of producing OTA, preferentially prior to ochratoxin production and accumulation. In this contribution several of these...

Asadiar,Leila Sadat; Rahmani,Fatemeh; Siami,Abbas.
Elaeagnus is a Eurasian tree with 77 species worldwide. In this study, ISSR markers were used to establish the level of genetic relationships and polymorphism across nine genotypes of Elaeagnus angustifolia collected from 9 different regions of West Azarbaijan province. The ISSR analysis with 11 anchored primers also generated 116 scorable loci, of which 92 were polymorphic (79.3%). The estimated Jaccard similarity coefficient ranged from 0.44 to 0.76 for the ISSR markers. Cluster analysis was carried out, based on the Unweighted Pair Group Method with Arithmetic Averages (UPGMA) and the dendrogram drawn with the help of the NTSYSpc 2.02 software. The analysis revealed 5 main clusters for the ISSR data. According to our results, there is a relatively high...

Association between MUC1 gene polymorphism and expected progeny differences in Nelore cattle (Bos primigenius indicus) Genet. Mol. Biol.
Souza,Fabio Ricardo Pablos de; Vozzi,Pedro Alejandro; Vila,Reginaldo Aparecido; Boligon,Arione Augusti; Galerani,Marli Aparecida Vani; Lobo,Raysildo Barbosa; Martelli,Lucia Regina.
MUC1 is a heavily glycosylated mammalian transmembrane protein expressed by mucosal secretory tissues for both protection against microbial infection and lubrication. An important characteristic of MUC1 is its variable number of tandem repeats (VNTR) containing several sites for O-glycosylation. VNTR length has been associated with many human diseases and with certain economically important traits in domestic ruminants. The aim of the present study was to correlate the length of MUC1 gene VNTR with expected progeny differences (EPDs) obtained for growth, fertility and carcass traits. Five alleles were identified, with alleles containing short VNTRs being more frequent than those with long, thereby demonstrating that Brazilian Nelore cattle are...

Boty-II, a novel LTR retrotransposon in Botrytis cinerea B05.10 revealed by genomic sequence Electron. J. Biotechnol.
Zhao,Ming; Zhou,Jin yan; Li,Zhi dong; Song,Wei wei; Tan,You jiu; Tan,Hong.
Botrytis cinerea is a necrotrophic pathogen causing pre- and post-harvest diseases in at least 235 plant species. It manifests extraordinary genotype and phenotype variation. One of the causes of this variation is transposable elements. Two transposable elements have been discovered in this fungus, the retrotransposon (Boty), and the transposon (Flipper). In this work, two complete (Boty-II-76 and Boty-II-103) and two partial (Boty-II-95 and Boty-II-141) long terminal repeat (LTR) retrotransposons were identified by an in silico genomic sequence analysis. Boty-II-76 and Boty-II-103 contain 6439 bp nucleotides with a pair of LTRs at both ends, and an internal deduced pol gene encoding a polyprotein with reverse transcriptase and DDE integrase domains. They...

Characterization and genetic relationships among Brazilian biotypes of Schizaphis graminum (Rondani) (Hemiptera: Aphididae) using RAPD markers Neotropical Entomology
Lopes-da-Silva,Marcelo; Tonet,Gabriela E.L.; Vieira,Luiz G.E..
The greenbug Schizaphis graminum (Rondani) is one of the most important cereals pests in the world. Within populations of this species, several biotypes, which are clones that share same virulence relationships with cultivated plants, can be distinguished. Molecular markers have been used to genetically characterize insect populations because they are fast and cost effective. In order to find RAPD markers to identify Brazilian S. graminum biotypes, nineteen clonal populations of three biotypes (B, C and E) from Brazil and three clonal populations from the U.S. were examined. Eighteen primers were used to analyze the material, but only six primers revealed polymorphisms and among those, three produced diagnostic band profiles that allowed biotype...

Comparative proteomic analysis of indica and japonica rice varieties Genet. Mol. Biol.
Yang,Yanhua; Zhu,Keming; Xia,Hengchuan; Chen,Liang; Chen,Keping.
Indica and japonica are two main subspecies of Asian cultivated rice (Oryza sativa L.) that differ clearly in morphological and agronomic traits, in physiological and biochemical characteristics and in their genomic structure. However, the proteins and genes responsible for these differences remain poorly characterized. In this study, proteomic tools, including two-dimensional electrophoresis and mass spectrometry, were used to globally identify proteins that differed between two sequenced rice varieties (93-11 and Nipponbare). In all, 47 proteins that differed significantly between 93-11 and Nipponbare were identified using mass spectrometry and database searches. Interestingly, seven proteins were expressed only in Nipponbare and one protein was...

Cucumber mosaic virus resistance and reproductive biology of brazilian melon accessions Rev. Bras. Frutic.
Sanabria-Verón,Nadia Carolina; Melo,Cláusio Antônio Ferreira de; Pereira,Jadergudson; Nunes,Glauber Henrique de Sousa; Oliveira,Ohana Luiza Santos de; Corrêa,Ronan Xavier.
Abstract In melon (Cucumis melo) the resistance to Cucumber Mosaic Virus (CMV) and the knowledge about reproductive characteristics are important for selection of genitors for breeding population. So, the objective was to investigate if different accessions of melon from a Brazilian germplasm bank have variability in resistance to CMV (FNY strain) and polymorphism in resistance loci markers, as well as whether there is genetic diversity on resistance and floral morphology descriptors and reproductive biology, aiming at genetic improvement. The symptoms induced by CMV were evaluated in 52 melon genotypes which were genotyped with three markers of single nucleotide polymorphisms related to CMV resistance. A sample of 24 genotypes was characterized for...

Development and characterization of microsatellite loci for Campomanesia xanthocarpa (Myrtaceae) and cross amplification in related species Biological Sciences
Góes, Bruna Delgado; Ruas, Eduardo Augusto; Benício, Lucas Milanez; Cassiano, Daniele; Souza, Felipe Pinheiro de; Ruas, Paulo Maurício.
Campomanesia xanthocarpa is a native tree, of common occurrence in almost all Brazilian Forest formations, which has its fruits and timber with high commercial value. Using an enriched genomic library we isolated and characterized microsatellite loci for C. xanthocarpa (Myrtaceae), in order to estimate genetic diversity parameters for this and related species. Twenty-eight microsatellite loci were identified and ten of them successfully amplified and showed polymorphism in a sample of 96 individuals, from four natural populations. The number of alleles per locus ranged from two to eight, and the observed and expected heterozygosities varied from 0.042 to 1.000 and from 0.294 to 0.855, respectively. These markers were tested and validated in two related...

Development of microsatellite loci and population genetics of the catfish Pimelodus yuma (Siluriformes: Pimelodidae) Neotropical Ichthyology
Joya,Cristhian Danilo; Landínez-García,Ricardo Marcel; Márquez,Edna Judith.
ABSTRACT Pimelodus yuma (formerly Pimelodus blochii) is a freshwater fish, endemic to the Colombian Magdalena-Cauca and Caribbean basins that experiences habitat disturbances resulting from anthropogenic activities. Due to the lack of information about the population genetics of this species, this study developed 14 species-specific microsatellite loci to assess the genetic diversity and population structure of samples from the lower section of the Cauca River. The studied species showed genetic diversity levels higher than the average values reported for Neotropical Siluriformes and significant inbreeding levels as was described for some congeners. Furthermore, P. yuma comprises two coexisting genetic groups that exhibit gene flow along the lower section...
