Improving the genetic transformation efficiency of Cucumis melo subsp. melo ‘Piel de Sapo’ via Agrobacterium National Institute of Agronomic Research

The effects of five Agrobacterium strains, two selection agents, and co-culture conditions were evaluated on the transformation efficiency of three melon lines (‘T111’, ‘PS8’, ‘PNY’) belonging to the cultivar Cucumis melo subsp. melo ‘Piel de Sapo’, the C. melo subsp. melo ‘Védrantais’ line, and the accession C. melo subsp. agrestis ‘PI 161375’. Transformation efficiencies for accession ‘PI 161375’ (10.6 %) were higher than those obtained for ‘Piel de Sapo’ (0.1 %) or ‘Védrantais’ (3.1 %). Among the ‘Piel de Sapo’ lines, the highest rates of transformation were observed with ‘T111’. Selection with pmi solved the problem of escapes encountered with nptII on ‘Piel de Sapo’. This study analyzes the relationship between the changes in the energy balance components and the muskmelon stomatal resistance under plastic mulch conditions. This experiment was conducted in a muskmelon crop with and without plastic mulch, from April to July, 1998. To measure the energy balance components (net radiation (Rn), latent heat flux LE, sensible heat flux H and surface soil flux G). The effect of soil solarization during 30 days and the fertilize with goat manure to the soil at the rate of 0,20 and 40 t ha-1 was studied; the objective of this experiment was to increase soil temperature in order to study its effect on weeds population and on muskmelon yield. Solarization produced maximum soil temperatures of 55°C and 44°C at the depth of 1.5 and 10 cm respectively, which in turn clearly reduced the emergence and growth of weeds in solarized plots. Goat manure only increased soil temperature by 1.5 to 2.5°C, but it had an apparent antagonist effect on weeds density, however, this effect was not clear on solarized plots.
