Ability of Allium cepa L. root tips and Tradescantia pallida var. purpurea in N-nitrosodiethylamine genotoxicity and mutagenicity evaluation Anais da ABC (AABC)
Rainho,Claudia R. de; Kaezer,Andréa; Aiub,Claudia A.F.; Felzenszwalb,Israel.
N-nitroso compounds, such as N-nitrosodiethylamine (NDEA), can be formed by the reaction of secundary amines with nitrosating agents, and are suspected to be involved in tumors in humans. NDEA has been considered a weak carcinogen in genotoxic assays probably due to the inefficient nitrosamine activation system that is used and/or to the efficient repair system. In this work, we evaluated the sensibility of Allium cepa L. root tips and Tradescantia stamen hair mutation assay (Trad-SH) using Tradescantia pallida var. purpurea for NDEA (0.1; 0.5; 5 and 25mM) genotoxicity and mutagenicity induction. Allium cepa L. was treated with different NDEA concentrations for 3h, for 3 consecutive days, including negative control (distilled water) and positive control...

Absence of mutagenicity in somatic and germ cells of mice submitted to subchronic treatment with an extract of Croton cajucara Benth. (Euphorbiaceae) Genet. Mol. Biol.
Santos,Fabio V.; Mesquita,Suzana F.P.; Faria,Maria José S.S.; Poersh,Aline; Maciel,Maria Aparecida M.; Pinto,Angelo C.; Morimoto,Helena K.; Cólus,Ilce Mara S..
The plant Croton cajucara Benth. (Euphorbiaceae) is a medicinal plant from the Brazilian Amazon where it is commonly known as sacaca. The principal compound isolated from C. cajucara stem-bark extracts is the clerodane-type diterpene trans-dehydrocrotonin (DCTN) which presents several biological activities, including antiulcerogenic, anti-inflammatory, hypoglycemic, antimutagenic and antitumoral activity. However, few studies have been carried out to evaluate the therapeutic potential of raw C. cajucara extracts. We studied mutagenicity and antimutagenicity effects of C. cajucara methanol extract using the micronucleus assay in bone marrow cells and the dominant lethal assay in mice submitted to subchronic treatments. The blood testosterone levels of the...

Alterations in the mitotic index of Allium cepa induced by infusions of Pluchea sagittalis submitted to three different cultivation systems Anais da ABC (AABC)
Rossato,Liana V.; Tedesco,Solange B.; Laughinghouse IV,Haywood D.; Farias,Júlia G.; Nicoloso,Fernando T..
We evaluated the antiproliferative effect of infusions from Pluchea sagittalis using the Allium cepa test. Infusions in three concentrations (2.5, 5, and 25 g dm-3) of leaves cultivated in three environments (in vitro, acclimatized growth chamber, and field) were used. Six onion bulbs were used for each of the eight treatments, and the mitotic index was obtained from 6000 cells per treatment. In conclusion, leaf infusions of P. sagittalis cultivated in the field have a high antiproliferative activity, as well as the cultivation system influences the antiproliferative potential.

Assessment of the genetic risks of a metallic alloy used in medical implants Genet. Mol. Biol.
Gomes,Cristiano C.; Moreira,Leonardo M.; Santos,Vanessa J.S.V.; Ramos,Alfeu S.; Lyon,Juliana P.; Soares,Cristina P.; Santos,Fabio V..
The use of artificial implants provides a palliative or permanent solution for individuals who have lost some bodily function through disease, an accident or natural wear. This functional loss can be compensated for by the use of medical devices produced from special biomaterials. Titanium alloy (Ti-6Al-4V) is a well-established primary metallic biomaterial for orthopedic implants, but the toxicity of the chemical components of this alloy has become an issue of concern. In this work, we used the MTT assay and micronucleus assay to examine the cytotoxicity and genotoxicity, respectively, of an extract obtained from this alloy. The MTT assay indicated that the mitochondrial activity and cell viability of CHO-K1 cells were unaffected by exposure to the...

Abstract It is understood that drugs regardless of their order of administration can exhibit drug interactions. Established on the fact that treatment of hypertension may last for decades and prolong usage of multiple drug regimen may induce substantial pathophysiological changes. Hence, This study was designed to evaluate the possible synergistic toxic effects of anti-hypertensive (carvedilol), and anti-inflammatory drug (celecoxib) alone and in combinations. Well-established MTT assay, Single Cell Gel Electrophoresis (SCGE) and Ames assay were employed to evaluate the toxicity at cellular level. Results from MTT assay on Vero cell line revealed that drug combinations have more pronounced anti-proliferative activity with combine IC50 value of 13.7:47.8...

Effects of Pterocaulon polystachyum DC. (Asteraceae) on onion (Allium cepa) root-tip cells Genet. Mol. Biol.
Knoll,Michelle Frainer; Silva,Antonio Carlos Ferreira da; Canto-Dorow,Thais Scotti do; Tedesco,Solange Bosio.
In Brazil leaf infusions of the plant Pterocaulon polystachyum DC (Asteraceae) are used to treat digestive problems. We used plants from six P. polystachyum populations to prepare fresh aqueous leaf infusions at the concentration normally used in Brazil (2.5 g L-1) and at twice (5 g L-1) and four times (10 g L-1) this concentration. We rooted onion bulbs in a water control or one of the infusions for 24 h and used the in vivo onion (Allium cepa) root-tip cell test to investigate the potential mutagenicity of the infusions by calculating the mitotic index for the control and the experimental treatments and comparing them using the Chi-squared test (chi2, p = 0.05). There was a highly significant decrease in the mitotic index of root-tip cells treated with...

Carbonari,Karina; Gonçalves,Luciane; Roth,Daniela; Moreira,Patrick; Fernández,Ricardo; Martino-Roth,Maria da Graça.
It is well recognized that electromagnetic fields can affect the biological functions of living organisms at both cellular and molecular level. The potential damaging effects of electromagnetic fields and very low frequency and extremely low frequency radiation emitted by computer cathode ray tube video display monitors (VDMs) has become a concern within the scientific community. We studied the effects of occupational exposure to VDMs in 10 males and 10 females occupationally exposed to VDMs and 20 unexposed control subjects matched for age and sex. Genetic damage was assessed by examining the frequency of micronuclei in exfoliated buccal cells and the frequency of other nuclear abnormalities such as binucleated and broken egg cells. Although there were no...

Ortho-nitrobenzyl derivatives as potential anti-schistosomal agents BJPS
Lopes,Marcela Silva; Suzuki,Brian Michio; Pereira,Glaécia Aparecida do Nascimento; Probst,Alexandra Christina; Ferreira,Rafaela Salgado; Oliveira,Júlia Teixeira de; Tecchio,Kimberly Brito; Santos,Fabio Vieira dos; Caffrey,Conor Robert; Oliveira,Renata Barbosa de.
ABSTRACT In the search for new anti-schistosomal agents, a series of fifteen ortho-nitrobenzyl derivatives was assayed in vitro against both the schistosomulum (somule) and adult forms of Schistosoma mansoni. Compounds 8 and 12 showed significant activity against somules at low micromolar concentrations, but none was active against adults. The SAR demonstrated that the compounds most active against the parasite were mutagenic to the human cell line RKO-AS45-1 only at concentrations 10- to 40-fold higher than the worm-killing dose. Given their electrophilicity, compounds were also screened as inhibitors of the S. mansoni cysteine protease (cathepsin B1) in vitro. Amides 5 and 15 exhibited a modest inhibition activity with values of 55.7 and 50.6 % at 100...
