Analysis of the SHP2 enhancer for the use of tissue specific activation tagging in Arabidopsis thaliana Genet. Mol. Biol.

Activation tagging is a powerful tool to identify new mutants and to obtain information about possible biological functions of the overexpressed genes. The quadruple cauliflower mosaic virus (CaMV) 35S enhancer fragment is a strong enhancer, which is most commonly used for this purpose. However, the constitutive nature of this enhancer may generate lethal mutations or aberrations in different plant organs by the same overexpressed gene. A tissue-specific activation tagging approach may overcome these drawbacks and may also lead more efficiently to the desired phenotype. Current genomic resources in melon and other cucurbits National Institute of Agronomic Research

In recent years there has been an effort towards the development of genomic resources in several cucurbit species, mainly in melon, cucumber and watermelon. The number of available sequences for these species in public databases has increased recently. This has allowed the design of microarrays for transcriptome analysis and the discovery of SNP markers for genotyping. Other genomic tools are also available in melon as BAC libraries, mutant populations and saturated genetic maps. Some international initiatives have emerged that will probably lead to the sequencing of the complete genomes of some cucurbit species during the next years.

DNA polymorphism and total protein in mutants of Metarhizium anisopliae var. Anisopliae (Metsch.) Sorokin strain E9 BJM

Five mutants (MaE10, MaE27, MaE24, MaE41 e MaE49) of Metarhizium anisopliae wild strain E9 were analysed for DNA profile through the RAPD technique and for changes in total protein content by spectrophotometry, polyacrylamide gel electrophoresis and densitometry. The pattern of RAPD markers showed genetic polymorphism among the strains: out of twenty primers seven were selected, producing 113 bands. Forty seven bands were present in all strains (41.6% of monomorphic bands) and 66 showed polymorphism (58.4%). The mean coefficient of similarity among all strains was 0.75 (75%). Abstract Urease catalyzes the hydrolysis of urea to ammonia and carbon dioxide. The ammonia (nitrogen (N) product of urease activity) is incorporated into organic compounds. Thus, urease is involved in N remobilization, as well as in primary N assimilation. Two urease isoforms have been described for soybean: the embryo-specific, encoded by the Eu1 gene, and the ubiquitous urease, encoded by Eu4. A third urease-encoding gene was recently identified, designated Eu5, which encodes the putative protein product SBU-III. Filtration enrichment method for isolation of auxotrophic mutants of Trichoderma harzianum Rifai. Repositório Alice

The isolation of genetic markers, like drug resistance and auxotrophy, is a laborious but important step in genetic research. The isolation of auxotrophic mutants of Trichoderma harzianum using the filtration enrichment technique was more effective than using the total isolation technique. Most of 12 auxotrophic mutants exhibited similar growth rate and higher sporulation when compared with the wild type, but only two mutants (TWS-410 and TW5-523) could grow in 500ug/L of benomyl.

Improvement of xylanase production by a parasexual cross between Aspergillus niger strains BABT

A diploid strain (D4) isolated via parasexual recombination between two Aspergillus niger xylanase overproducing mutants was characterised in terms of enzyme production and catabolite repression by glucose. This strain increased xylanase production (607 nkat/ml), which was nearly 100% higher than titers achieved by the wild type strain (305 nkat/ml) and 28% higher than the best mutant used to induce parasexual cycle. Diploid D4 was also less sensitive to carbon catabolite repression by glucose, since xylanolytic activity was detected under conditions normally repressing production by the wild type strain. No decrease in maximal xylanase levels was observed in the presence of glucose for diploid D4.

Objetivou-se adaptar e utilizar o metodo de inducao de mutacao por irradiacao no melhoramento de crisantemo, para a obtencao de mutantes na colaracao de petalas da cultivar Repin Rosa. Mudas obtidas a partir de estacas enraizadas foram submetidas a irradiacao com 20 Gy de raios-gama aplicando-se a seguir o metodo de podas repetidas para avanco das geracoes, antes da selecao dos mutantes. Observou-se uma frequencia de 5,80% de mutantes para coloracao de petalas. Antagonism of Trichoderma spp. against other microrganisms in general operates by exolysis, breaking down the walls of fungal pathogens. Trichoderma species seem to be the best source of extracellular cellulase that can solubilize highly ordered cellulose. Cell walls of sclerotia of slerotiorum usually contain chitin and b-glucans. Such sclerotia may be vulnerable to attack by many microrganisms. The objective of this research was the isolation of improved strains for b-glucosidase production. The dark coloration of insects eyes is attributed to the accumulation of the brown pigment insectorubin, a mixture of ommochromes, xanthommatin and several ommins, biosynthesized from tryptophan.
