ANALYSES OF GENETIC VARIABILITY IN LENTINULA EDODES THROUGH MYCELIA RESPONSES TO DIFFERENT ABIOTIC CONDITIONS AND RAPD MOLECULAR MARKERS BJM

The growth of thirty-four Lentinula edodes strains submitted to different mycelial cultivation conditions (pH and temperature) was evaluated and strain variability was assessed by RAPD molecular markers. The growth at three pH values (5, 6 and 7) and four different temperatures (16, 25, 28 and 37ºC) was measured using the in vitro mycelial development rate and water retention as parameters. Mycelial cultivation was successful at all pH tested, while the ideal temperature for mycelial cultivation ranged between 25 and 28ºC. The water content was lower in strains grown at 37ºC. Abstract The antagonistic effectiveness of native strains of Trichoderma spp. on Fusarium oxysporum f. sp. pisi. in vitro, greenhouse and field conditions, were evaluated. in vitro conditions, the antagonistic capacity of 12 strains of Trichoderma spp., C2, C7, C12 and C21 strains, exhibited a better behavior measured by the following variables: inhibition halo and mycelial growth. In greenhouse conditions, the four strains, which showed the best in vitro antagonistic behavior, were evaluated using a DIA experimental design with factorial arrangement for three factors, which corresponded to strain, concentration and dose. O presente trabalho teve como objetivo identificar 11 isolados de Guignardia citricarpa, agente causal da mancha preta dos citros (MPC), obtidos de frutas cítricas sintomáticas de diferentes regiões geográficas, por meio da PCR e caracterização morfofisiológica das estruturas propagativas, esporulação e crescimento micelial em diferentes meios de cultura, temperaturas e regimes de luz, nas condições de laboratório. Cultivation techniques for Agaricus brasiliensis (composting method) and substratum formulations are the same ones used for Agaricus bisporus. Most of the carbon-to-nitrogen (C:N) ratios reported for A. brasiliensis are similar to those used for A. bisporus on the composting method and there are few studies about the variation of C:N ratios for A. brasilienses on the axenic method. Comparison of radial growth rate of the mutualistic fungus of Atta sexdens rubropilosa forel in two culture media BJM

In vitro culture of the mutualistic fungus of leaf-cutting ants is troublesome due to its low growth rate, which leads to storage problems and contaminants accumulation. This paper aims at comparing the radial growth rate of the mutualistic fungus of Atta sexdens rubropilosa Forel in two different culture media (Pagnocca B and MEA LP). Although total MEA LP radial growth was greater all along the bioassay, no significant difference was detected between growth efficiencies of the two media. Cultivation of Flammulina velutipesmushroom using various agro-residues as a fruiting substrate PAB

The objective of this work was to evaluate the feasibility of different agro-residues as a carbon source in the fruiting substrates of Flammulina velutipesmushroom and the effect of supplementation with the nitrogen sources spent brewer's yeast and rice bran. The following fruiting substrates were evaluated: rubber wood sawdust (SD), paddy straw (PS), palm empty fruit bunches (EFB), and palm-pressed fiber (PPF). Cultivation was done on each agro-residue, based on formulations consisting of two substrates at the ratios of 3:1, 1:1, and 1:3. Mycelial growth rate and basidiocarp yield were evaluated. Difference between isolates from brown eye spot and black spot lesions in coffee plants PAB

Abstract: The objective of this work was to evaluate the difference between the isolates from two cercospora leaf spot symptoms (brown eye spot and black spot) in relation to conidial morphology, mycelial growth, cercosporin production, and reproduction of symptoms in coffee leaves collected in three municipalities in the state of Minas Gerais, Brazil. There was a difference between the isolates for the studied variables, but without any relation to the type of symptom. The differences found were not associated with the origin of the isolates. The symptoms characterized as black spot were associated with different host conditions during the infection process.

Efeito de benomyl no crescimento micelial de isolados de Trichoderma spp. in vitro. Repositório Alice

Estudou-se o efeito de benomyl no crescimento micelial de Trichoderma spp., quando observou-se que de 402 isolamentos de caule, colo e raizes de plantas de maracuja em dois meios de cultura: (1) BDA + Ampicilina (50 ppm) + Clorafenicol (20 ppm) + Benomyl (20 ppm) e, (2) BDA + Tetraciclina (100 ppm), Trichoderma spp. surgiu em 42 dos isolamentos, sempre no meio de cultura. 1. Effect of surfactants and temperature on germination and vegetative growth of Beauveria bassiana BJM

Three non-ionic surfactants: Tween20, Tween80 and Breakthru® were screened for their effects on spore germination and mycelial growth rates and for their influence on three isolates of Beauveria bassianaspore germination at various temperatures. Tween20 and Tween80 were compatible with all the B. bassiana isolates in the germination studies, but inhibited germination at higher surfactant concentrations, irrespective of the conidial concentrations. Breakthru® had an inhibitory effect on germination even at the lowest concentration of 0.1% on all the B. bassiana isolates. O objetivo deste trabalho foi avaliar a fungitoxicidade de produtos pertencentes aos grupos dos benzimidazóis, triazóis, estrobilurinas, isoftalonitrilas e ditiocarbamatos sobre a germinação conidial e o crescimento micelial in vitro de isolados de Myrothecium roridum e, in vivo, sobre a severidade da mancha-de-mirotécio em plantas de algodoeiro. Bipolaris euphorbiae Muchovej & Carvalho can be combined with herbicide in order to control a large spectrum of weed species, being a strong candidate for the biocontrol of Euphorbia heterophylla L. Growth and antibacterial activity of Lentinula edodes in liquid media supplemented with agricultural wastes Electron. J. Biotechnol.

Antibacterial activity of Lentinula edodes against Bacillus subtilis was evaluated in cell-free filtrates obtained after growth in 14 different culture media. The highest B. subtilis growth inhibition was promoted by filtrates of growth media supplemented with rice bran, vermiculite or molasses. L. edodes dry mycelial biomass in liquid culture with 0.5% added rice bran was 3.2 mg/ml, after growth for 30 days at 25ºC without shaking, and 4.3 mg/ml under orbital shaking (150 rpm). However, antibacterial activity, detected between 20 and 24 days of incubation of stationary cultures, was absent in filtrates of aerated cultures. Growth of symbiont fungi of some higher attine ants in mineral medium Ciência Rural

Bioassays were conducted to verify the possibility of culturing the symbiont fungus of some higher attine in mineral medium and finding out the optimum pH value for their satisfactory mycelial growth. Three organic media and one mineral medium were inoculated with isolates from Atta sexdens piriventris and Acromyrmex heyeri. In mineral medium different values of pH (4.0, 5.0, 6.0 and 7.0) were tested with isolates from A. laevigata and A. laticeps. The behavior of isolates (colony diameter) was different in the mineral medium. However, even the one which grew the least of all provided enough mycelial for RAPD analysis. Morphology and mycelial growth rate of Pleurotus spp. strains from the Mexican mixtec region BJM

Two native Pleurotus spp. strains (white LB-050 and pale pink LB-051) were isolated from rotten tree trunks of cazahuate (Ipomoea murucoides) from the Mexican Mixtec Region. Both strains were chemically dedikaryotized to obtain their symmetrical monokaryotic components (neohaplonts). This was achieved employing homogenization time periods from 60 to 65 s, and 3 day incubation at 28 °C in a peptone-glucose solution (PGS). Herein the order of effects of nutrient components on mycelial growth was tryptone > Ca2+ > soluble starch > folacin, corresponding to the following optimal concentrations: 1.58% Soluble Starch, 3.16% Tryptone, 0.2 mmol l-1 Ca2+ and 0.005% Folacin. O álamo (Populus deltoides) é uma espécie florestal que possui diversos problemas fitossanitários, como a ocorrência de manchas foliares, ferrugens e podridões radiculares. Desde 2010 tem-se verificado a morte de árvores em plantios comerciais no estado do Paraná, Brasil, ocasionados pelo fungo Rosellinia bunodes. Devido a carência de informações sobre este patossistema, objetivou-se com esse trabalho, realizar a caracterização fisiológica de isolados de R. bunodes e sintomatológica da roseliniose em álamo.
