Mycoplasma gallisepticum (MS) and Mycoplasma synoviae (MS) are important avian pathogens and cause economic losses to the poultry industry. Molecular biology techniques are currently used for a rapid detection of these pathogens and the adoption of control measures of the diseases. The aim of this study was to develop and validate a technique for simultaneous detection of MG and MS by multiplex real time polymerase chain reaction (PCR). The complete assay (Multiplex MGMS) was designed with primers and probes specific for each pathogen and developed to be carried out in a single tube reaction. Vaccines, MG and MS isolates and DNA from other Mycoplasma species were used for the development and validation of the method. Further, 78 pooled clinical samples...

Santos,FF; Li,J; Brandão,MDM; Machado,LS; Soares,NM; Nascimento,ER; Khan,MI; Pereira,VLA.
ABSTRACT The objective of this study was to identify the species and characterize the genetic relationships among mycoplasma isolates from commercial layer hen flocks using 16S-23S rDNA intergenic spacer region (IGSR) sequencing. Twenty-one isolates were obtained from samples collected from commercial layer flocks in four Brazilian states: São Paulo, Minas Gerais, Rio de Janeiro and Espírito Santo. The isolates were recovered from the São Paulo, Rio de Janeiro and Espírito Santo states. Eleven isolates were originated from tracheal swabs, five from shell gland swabs and five from ovary fragment collection. The 16S-23S rDNA IGSR of isolates were amplified by PCR, and the obtained products were subsequently sequenced. The consensus of each isolate was...

Agüero,J.A; Hotzel,H; Aguilar,Lisandra; Lima,Claudia; Sachse,K; Martínez,Siomara.

Influence of culture medium components on Mycoplasma gallisepticum growth Rev Salud Anim.
Pérez Bueno,Tania; Sánchez Miranda,Lilian; Rosado Ruiz-Apodaca,Ileana; Díaz Bravo,Tito; Rodríguez Edreira,Mercedes; Martínez Marrero,Siomara.
The composition of a culture medium produced in Cuba to grow Mycoplasma gallisepticum was evaluated using a response surface methodology. Initially, the influence of Mycoplasma base, horse serum and yeast extract on biomass production was assessed in a 2³ factorial experimental design. Then, a central composite design was applied. Mycoplasma base and horse serum factors were selected for this study. In 7 L and 35 L bioreactors, the variations carried out in the culture medium composition were estimated. The experimental results were fitted to a second order polynomial model equation. Finally, the optimal growth conditions established were at concentrations of 1,88% Mycoplasma base and 18,16% horse serum (v/v) with 0,6% of yeast extract. For the conditions...

Molecular cloning and expression of a computationally predicted surface antigen of Mycoplasma gallisepticum Rev Salud Anim.
Agüero,José Antonio; Aguilar-Bultet,Lisandra; Rodríguez,Ariadna.
Mycoplasmas, the simplest self-replicating organisms known, are distinguished phenotypically from other bacteria by their minute size and total lack of cell wall. The poultry industry is affected by several species of mycoplasmas, but Mycoplasma gallisepticum (MG) is the most economically significant one. The attachment of mycoplasmas to host respiratory epithelial cells constitutes a critical step in the pathway leading to infection and disease and is achieved by lipoproteins localized on the bacterial surface. In a recent in silico study, it was predicted a set of MG putative surface proteins with potential antigenic properties that could be used as candidates for exploring new vaccines, and for diagnostic tests as well. One of those potential candidates...

Phenotypic and antigenic variation of Mycoplasma Gallisepticum vaccine strains BJM
Ferraz,Priscila N.; Danelli,Maria das Graças M..
Phenotypic and antigenic variations among Mycoplasma gallisepticum vaccine F and Ts-11 strains were investigated by SDS-PAGE and serological methods (hemagglutination-inhibition and immunoblot assays). The SDS-PAGE system followed by densitometer analysis showed weak phenotypic variability between the strains, being the major difference close to the 75 kDa level where a prominent peptide band was detected only in the F vaccine strain. Polyclonal antibodies to the M. gallisepticum antigens produced in chickens were used in serological tests for this antigenic variability study. There were strong cross-reactions between the strains and homologous/ heterologous antibodies. The most evident characteristic was the specific response of the vaccine-type F...

Protein profile analysis by SDS-PAGE of Mycoplasma Gallisepticum strains S6(208) and F-K810 grown in Hayflick's and Frey's media BJM
Ferraz,Priscila N.; Menezes,Claudio C. P.; Danelli,Maria das Graças M.; Lizeu,Jackeline de O. P.; Nascimento,Elmiro R. do; Lucchesi,Daniela M..
The Mycoplasma gallisepticum strains [wild-type S6 (208) and a vaccine-type F-K810] grown in Frey´s and Hayflick´s media were analyzed by SDS-PAGE. No visual changes in the protein profiles of these strains were observed regardless of the media composition used, although the polyacrylamide gel electrophoretograms showed minor differences do exist when densitometer traces of the gel are compared. Both strains were easily differentiated on SDS-PAGE analysis by a peptide band p75, that is specific for MG F-K810 strain, used as vaccine.

SURVEILLANCE FOR NEWCASTLE DISEASE VIRUS, AVIAN INFLUENZA VIRUS AND MYCOPLASMA GALLISEPTICUM IN WILD BIRDS NEAR COMMERCIAL POULTRY FARMS SURROUNDED BY ATLANTIC RAINFOREST REMNANTS, SOUTHEASTERN BRAZIL Rev. Bras. Ciênc. Avic.
Guimarães,MB; Hurtado,R; Bello,CP; Vanstreels,RET; Ferreira,AJP.
ABSTRACT The geographic overlap between areas of Atlantic rainforest and human activities allows interactions to occur between humans and wild and domestic animals. Despite the great importance of the domestic animal-wildlife-human interface that occurs at poultry farms in terms of public health, economic production and wildlife conservation, there are few studies in Brazil examining the distribution and health of wild birds that interact with poultry farms. From January to December 2010, mist nets were used to capture 166 free-ranging birds that were within close proximity to three poultry farms in Atlantic rainforest remnants in south-eastern Brazil. The species composition was examined, and molecular methods were used to test for avian influenza virus,...
