Action of cobra venom on the renal cortical tissues: electron microscopic studies J. Venom. Anim. Toxins
RAHMY,T. R..
The effect of intramuscular (IM) injection of a sub-lethal dose of Naja haje cobra venom (0.015 mug/gm body weight) on the ultrastructure of renal cortical tissues of rabbits was studied at different time intervals after envenomation. Succinic dehydrogenase (SDH) enzyme activity was also detected in the renal cortical tubules. Three hours after venom injection, slight changes were seen in visceral cells and glomerular endothelia. The cortical tubular epithelia revealed an increase of lysosomal structures, cytoplasmic vacuolization, and nuclear irregularity. Severe ultrastructural changes were recorded 6 hours after envenomation, including hypertrophied parietal cells, blebbed visceral epithelial cells, and foot process disorganization. Dilated glomerular...

PAIVA, P. P. de; DELCORSO, M. C.; MATHEUS, V. A.; QUEIROZ, S. C. do N. de; COLLARES-BUZATO, C. B.; ARANA, S..
The aim of this work was to evaluate the sensitivity of Pacu fingerlings (Piaractus mesopotamicus) by measuring the effects of median lethal concentration (LC50) of atrazine (ATZ - 28.58 mg/L) after acute exposure (up to 96 h). The fish were exposed to the LC50 of ATZ for 96 h (28.58 mg/L) in a static system. During the experiment, the fingerlings were randomly distributed in four glass tanks (50 L) containing dechlorinated water. Four glass tanks were for the control group, and four were for the ATZ-exposed group (n=4 per glass tank), given a total number of 16 animals tested per group. The genotoxicity was evaluated by micronucleus (MN) test in erythrocytes from peripheral blood. Qualitative and semi-quantitative histopathological analyses, and also...

Amphotericin B-related nephrotoxicity in low-risk patients BJID
Berdichevski,Roberto Herz; Luis,Luciano Billodre; Crestana,Letícia; Manfro,Roberto Ceratti.
INTRODUCTION: Amphotericin B (AmphoB) is the drug of choice for treatment of severe fungal illnesses; however, it is very nephrotoxic. Modified (less toxic) amphotericins are very expensive. In low-risk patients, saline loading would be enough to prevent significant loss of renal function. MATERIAL AND METHODS: Patients with normal renal function and within the first 24 hours of treatment with AmphoB were prospectively enrolled in the study. Patients in intensive care units or who were using vasoactive drugs were excluded. Saline loads were infused before and after the AmphoB treatment. Blood and urine analyses were made at the beginning and at the end of the treatment. Serum creatinine was repeated 30 days after the end of the AmphoB treatment. RESULTS:...

Effects of sirolimus alone or in combination with cyclosporine A on renal ischemia/reperfusion injury BJMBR
Pereira,B.J.; Castro,I.; Burdmann,E.A.; Malheiros,D.M.A.; Yu,L..
Calcineurin inhibitors exacerbate ischemic injury in transplanted kidneys, but it is not known if sirolimus protects or exacerbates the transplanted kidney from ischemic injury. We determined the effects of sirolimus alone or in combination with cyclosporin A (CsA) on oxygenated and hypoxic/reoxygenated rat proximal tubules in the following in vitro groups containing 6-9 rats per group: sirolimus (10, 50, 100, 250, 500, and 1000 ηg/mL); CsA (100 µg/mL); sirolimus (50 and 250 ηg/mL) + CsA (100 µg/mL); control; vehicle (20% ethanol). For in vivo studies, 3-week-old Wistar rats (150-250 g) were submitted to left nephrectomy and 30-min renal artery clamping. Renal function and histological evaluation were performed 24 h and 7 days after ischemia (I) in five...

de Souza,Vanessa Barboza; Oliveira,Rodrigo Fagundes L. de; Lucena,Hévio Freitas de; Ferreira,Aurigena A. A; Guerra,Gerlane Coelho Bernardo; Freitas,Maria de Lourdes; Queiroz,Karla Cristiane de Souza; de Araújo Júnior,Raimundo F.
Due to its prominent role in major excretory pathways, the kidney is particularly sensitive especially to toxicity for antimicrobials drugs. Storage of these drugs in the renal cortex, their effect on renal cells, have consequences on the renal function, and then reabsorbed by renal tubules induce nephrotixicity. Our objective was to show the renal morphopatological alterations induced by gentamicin through the histochemical methods of routine periodic acid de Schiff (PAS) staining and imunohistochemical staining for the expression of the protein P53, which is considered as a marker for cellular apoptosis. This allows the early detection of tubular lesions. The renal morphopathologic findings were cell apoptosis, basal membrane interruption, mesangial...

Abstract Acute renal failure (ARF) is an extremely important public health issue in need of novel therapies. The present study aimed to evaluate the capacity of mesenchymal stem cell (MSC) therapy to promote the improvement and recovery of renal function in a preclinical model. Wistar rats were used as the experimental model, and our results show that cisplatin (5mg/kg) can efficiently induce ARF, as measured by changes in biochemical (urea and creatinine) and histological parameters. MSC therapy performed 24h after the administration of chemotherapy resulted in normalized plasma urea and creatinine levels 30 and 45d after the onset of kidney disease. Furthermore, MSC therapy significantly reduced histological changes (intratubular cast formation in...

Abstract Background: Envenoming by kraits (genus Bungarus) is a medically significant issue in South Asia and Southeast Asia. Malayan krait (Bungarus candidus) venom is known to contain highly potent neurotoxins. In recent years, there have been reports on the non-neurotoxic activities of krait venom that include myotoxicity and nephrotoxicity. However, research on such non-neurotoxicity activities of Malayan krait venom is extremely limited. Thus, the aim of the present study was to determine the myotoxic, cytotoxic and nephrotoxic activities of B. candidus venoms from northeastern (BC-NE) and southern (BC-S) Thailand in experimentally envenomed rats. Methods: Rats were administered Malayan krait (BC-NE or BC-S) venom (50 μg/kg, i.m.) or 0.9% NaCl...

Protection against cephalosporin-induced lipid peroxidation and nephrotoxicity by (+)-cyanidanol-3 and vitamin E BJMBR
Cojocel,C.; Tolle,K.-L.; El-Hajj,H.; Baumann,K..
The ability of the clinically used cephalosporins: cephalothin, cefotaxime and cefotiam to induce lipid peroxidation (LPO) and renal damage was compared to that of nephrotoxic cephaloridine under in vivo conditions. Glutathione was measured in rat liver or in renal cortex as non-protein sulfhydryls. LPO was measured in plasma, renal cortex and liver by the generation of malondialdehyde or as the increase in renal cortical concentration of conjugated dienes. Impairment of renal function was measured as the decrease in renal cortical accumulation of the organic anion p-aminohippurate (PAH). Administration of cephalosporins to rats as a single dose (2000 mg/kg, ip) induced a significant glutathione-depletion in the renal cortex with cephaloridine, and in the...
