O objetivo deste trabalho foi estudar os efeitos da nutricao nitrica e amoniacal na distribuicao de nitrato e na atividade in vitro da redutase do nitrato em plantas de capim coloniao ( Panicum maximum Jacq.). as plantas receberam 7 mM de nitrogenio nas forma N- NH4 +, N-NO3- ou combinadas nas seguintes proporcoes 0/100, 25/75, 50/50, 75/25 e 100/0. A extracao enzigmatica foi feita em Tris-HCI 50mM pH 7,5, EDTA 0,5 mM e b-mercaptoetanol 10mM, e para o ensaio utilizou-se 200 mmoles de Tris-HCI pH 7,5, 20 mmoles KNO3, 0,1 mmol NADH e 0,1 mL de Triton X-100 1% no volume final de 2,0 mL. A concentracao de nitrato foi levemente superior na parte aerea quando as plantas receberam ambas as formas de nitrogenio, e na raiz seta decresceu com o aumento da proporcao... |

The Achillea millefolium L. is a perennial herb with important antibacterial, antifungal, anti-inflammatory, antitumoral, and antioxidant properties. This research aimed to investigate the effect of shading (75%; black net) and nitrogen fertilization (0, 75 and 150 kg urea ha-1) on the nitrogen metabolism, essential oil yield and antimicrobial activity of A. millefolium at vegetative- and reproductive-stage. Two experiments were carried out to evaluate the effect of supplementation with different nitrogenous compounds on the activities of carboxymethil cellulase (CMCase) and glutamate dehydrogenase (GDH). In the first experiment, four treatments were evaluated in vitro: cellulose, cellulose with casein, cellulose with urea, and cellulose with casamino acids. After 6, 12 and 24 hours of incubation, CMCase and GDH activity, pH, and concentrations of ammonia nitrogen (AN) and microbial protein were measured. We are using molecular, biochemical, and genetic approaches to study the structural and regulatory genes controlling the assimilation of inorganic nitrogen into the amino acids glutamine, glutamate, aspartate and asparagine. These amino acids serve as the principal nitrogen-transport amino acids in most crop and higher plants including Arabidopsis thaliana. We have begun to investigate the regulatory mechanisms controlling nitrogen assimilation into these amino acids in plants using molecular and genetic approaches in Arabidopsis. Assimilation of nitrate and ammonium are vital procedures for plant development and growth. From these primary paths of inorganic nitrogen assimilation, this metabolism integrates diverse paths for biosynthesis of macromolecules, such as amino acids and nucleotides, and the central intermediate metabolism, like carbon metabolism and photorespiration. This paper reports research performed in the CitEST (Citrus Expressed Sequence Tag) database for the main genes involved in nitrogen metabolism and those previously described in other organisms.
