Determination of ochratoxin. A content in wheat bread samples collected from the Algarve and Bragança regions, Portugal: Winter 2007 IPB - Escola Superior Agrária
Bento, J.; Pena, A.; Lino, C.M.; Pereira, J.A..
Ochratoxin A (OTA) is a mycotoxin produced by fungi such as Penicillium verrucosum and Aspergillium spp. and has been found to have a variety of potentially deadly toxic effects. The favoured substrate for fungal growth and OTA production appears to be cereals and flour-based products, including bread. Due to the dietary relevance of bread for the Portuguese population, it is imperative that its OTA content remains well within safe quantities. As such, bread samples collected from commercial surfaces across the Algarve region and from the city of Bragança during the winter of 2007 were tested for OTA through extraction with immunoaffinity columns and quantification by liquid chromatography coupled with fluorescence detection. Although OTA content was found...

Effects of ochratoxin a on broiler leukocytes Rev. Bras. Ciênc. Avic.
Moura,MA; Machado,CH; Porfírio,LC; Freire,RB.
This study evaluated alterations in the qualitative cellular profile of leukocytes caused by the administration of low doses of ochratoxin-A (OTA) in poultry. Sixty chicks were separated in three experimental groups: control, PBS-treated and OTA-treated. Blood smears from all birds were analyzed three and six hours post-treatment. Differential leukocyte counting demonstrated that OTA reduced the percentage of lymphocytes and eosinophils and significantly increased the number of heterophils and monocytes.

Exposure to ochratoxin A through consumption of grape juices produced by steam distillation method and intended for school meals Ciência Rural
Dachery,Bruna; Manfroi,Vitor; Welke,Juliane Elisa.
ABSTRACT: The grape juice was included in meals of public schools in the states of Rio Grande do Sul and Santa Catarina, Brazil. The juices intended for schools have been produced by small wineries through the steam distillation method. This method presents a particularity in relation to other juice production methods, since the grapes are not macerated to extract the juice. In literature is well established that maceration of grapes is the main critical point associated with the ochratoxin A (OTA) contamination in juices. OTA is a mycotoxin with nephrotoxic and immunosuppressives effects. The aim of this study was to verify the OTA occurrence in juices produced by steam distillation method, as grape juice may be a source of OTA exposure for children. No...

Fungicidal effect of silver nanoparticles on toxigenic fungi in cocoa PAB
Villamizar-Gallardo,Raquel; Cruz,Johann Faccelo Osma; Ortíz-Rodriguez,Oscar Orlando.
Abstract: The objective of this work was to evaluate the microbicidal effect of silver nanoparticles (AgNPs) on potentially toxigenic fungi affecting cocoa (Theobroma cacao) crops. These fungi, isolated from diseased cocoa pods, were characterized phenotypically and genotypically. The microbicidal effect was assessed by measuring radial mycelial growth, in synthetic culture media, and at different AgNP concentrations in plant tissues. The inhibition effect was monitored in Petri dishes, and changes in fungal structures were observed through scanning electron microscopy. Two potentially toxigenic fungi were highly prevalent: Aspergillus flavus and Fusarium solani. The inhibition assays, performed in liquid and solid synthetic culture media, showed that...

Genetic relationships among strains of the Aspergillus niger aggregate BABT
Ferracin,Lara Munique; Frisvad,Jens Christian; Taniwaki,Marta Hiromi; Iamanaka,Beatriz Thie; Sartori,Daniele; Schapovaloff,Maria Elena; Fungaro,Maria Helena Pelegrinelli.
We analyzed the genetic relationships between 51 fungal isolates previously identified as A. niger aggregate, obtained from dried fruit samples from worldwide origin and 7 A. tubingensis obtained from Brazilian coffee beans samples. Greater fungal diversity was found in black sultanas. Aspergillus niger sensu stricto was the most prevalent species. It was found in all fruit substrates of all geographical origins. Based on Random Amplification of Polymorphic DNA (RAPD) and β-tubulin sequences data two groups of A. niger were found. In spite of the small number of isolates from Group IV an association between extrolite patterns and molecular clustering is speculated. A. tubingensis were the second most frequent species and this species were clearly...

Influencing factors on bread-derived exposure to ochratoxin A: type, origin and composition IPB - Escola Superior Agrária
Duarte, S.; Bento, J.; Pena, A.; Lino, C.M.; Delerue-Matos, C.; Oliveira, M.B.P.P.; Alves, M.R.; Pereira, J.A..
The nearly ubiquitous consumption of cereals all over the world renders them an important position in international nutrition, but concurrently allocates exposure to possible contained contaminants. Mycotoxins are natural food contaminants, difficult to predict, evade, and reduce, so it is important to establish the real contribution of each contaminated food product, with the aim to evaluate mycotoxin exposure. This was the key objective of this survey and analysis for ochratoxin A content on 274 samples of commercialized bread in the Portuguese market, during the winter 2007. Different bread products were analyzed through an HPLC-FD method, including traditional types, novel segments, and different grain based bread products. A wide-ranging low level...

Monitoring of ochratoxin a exposure of the Portuguese population through a nationwide urine survey – Winter 2007 IPB - Escola Superior Agrária
Duarte, S.; Bento, J.; Pena, A.; Lino, C.M.; Delerue-Matos, C.; Oliva-Teles, T.; Morais, S.; Correia, M.; Oliveira, M.B.P.P.; Alves, M.R.; Pereira, J.A..
Ochratoxin A (OTA) is a mycotoxin produced by a variety of fungi, such as Penicillium verrucosum and Aspergillium spp., which has been found to have a wide number of potentially deadly toxic effects, and can enter the human organism through a variety of means. It then finds its way into the bloodstream and, after a lengthy process, is eventually excreted through the urine. It can thus be detected in its original form not only in blood samples but also in this biological medium. As such, and in an attempt to evaluate the exposure of the Portuguese population to this mycotoxin, morning urine samples were collected during the Winter of 2007, from each of five geographically distinct Portuguese locations — Bragança, Porto, Coimbra, Alentejo, and Algarve — and...
