Rodrigues, K.A..
The reproductive biology of Ramnogaster arcuata inhabiting waters of the Rio de la Plata estuary (Argentina - Uruguay) was studied by using macroscopic and histological analysis of ovaries. R. Arcuata specimens were collected from two research cruises of the National Institute of Fisheries Research and Development (INIDEP) carried out in June 1998 and June 1999. During these surveys, bottom salinity and temperature data were also registered. In addition, information about annual catches of this species in the Rio de la Plata, obtained from different INIDEP cruises performed during the last ten years, was analyzed. During winter, a prevalence of R. arcuata individuals was observed in the Rio de la Plata estuary. In summer, this species apparently moves...

Goudeau, M; Lachaise, F; Kubisz, P; Goudeau, H.
Electrophysiological measurements and related cytological controls were done on immature and submature oocytes of Homarus gammarus . By chromatography (HPLC) and radio-immunoassays (RIA) it was established that submature oocytes displayed different ecdysteroids. Furthermore, the structure of the stalk (funiculus) for the laid eggs attachment was analyzed. it consisted of the two superimposed vitelline envelopes which were likely modified by the cortical reaction exudate during the fertilization process. In addition, the successive secretory phases, regarding to the embryonic envelopes were evidenced. Finally, the ecdysteroid titers of embryonic stages were measured.

Effect of two ram sperm capacitating media on acrosome reaction and zona-free hamster oocyte penetration test Braz. J. Vet. Res. Anim. Sci.
FERRARI,Silvia; BARNABE,Valquíria Hyppolito; ZÜGE,Roberta Mara; ZOGNO,Maria Amélia.
An in vitro zona-free hamster oocyte penetration test was utilized in 24 trials in order to evaluate the capacitation media used for ram sperm. A pool of fresh semen was collected from three crossed breed rams. Two semen drops were washed by centrifugation and incubated in high ionic strength treatment (HIS) or in a defined medium with HEPES, on heat ewe serum and heparin. After the incubation to promote capacitation, simplified triple-stain technique was used to evaluate the spontaneous acrosome reaction of the capacitated sperm. Superovulation in 96 golden hamsters was induced by PMSG and hCG. The oocytes were treated with hyaluronidase and trypsin to remove, respectively, the cumulus cells and the zona pellucida. Oocytes and capacitated sperm were...

SARAIVA, N. Z.; PERECIN, F.; MÉO, S. C.; FERREIRA, C. R.; TETZNER, T. A. D.; OLIVEIRA, J. C.; GARCIA, J. M..
The developmental competence of enucleated oocytes is a key factor that determines the overall success of animal cloning. Enucleation is an invasive procedure in traditional nuclear transfer (NT). The objective of this work was to evaluate the effects of demecolcine, a microtubule-depolymerizing agent, on metaphase II (MII) bovine oocytes and to verify the capacity of embryonic development after NT using chemically assisted enucleation. In the first experiment, oocytes after 21 h of IVM were exposed for 2 h to several concentrations of demecolcine: 0 (control), 0.025, 0.05, 0.2, and 0.4 µg mL?1, and evaluated in relation to membrane protrusion formation. After the best concentration of demecolcine was determined, the nuclear and microtubular dynamics of...

We aimed to elucidate the effects of PGE2 and PGF2a on the in vitro maturation (IVM) of bovine oocytes. First, cumulus-oocyte complexes were matured in the media supplemented with or without PGE2, PGF2a, or PGE2 plus PGF2a for the final 24, 12, or 6 h of culture. Then, the cumulus-oocyte complexes were matured in the absence or presence of a PG endoperoxide synthase 2 (PTGS2) enzyme inhibitor (NS398) supplemented with PGE2, PGF2a, or PGE2 plus PGF2a. Finally, the expression of genes associated with PGs activity in cumulus cells (PTGS2, PG E-synthase-1 [PTGES1], and aldo-keto reductase 1 [AKR1B1]) or oocytes (receptors for PGE2 [PTGER2] and PGF2a [PTGFR]) of different competencies was quantified. Supplementation of the IVM medium with PGs did not improve in...

Histological characterization of cellular types during Scinax fuscovarius oogenesis (Lutz) (Anura, Hylidae) Zoologia
Oliveira,Classius de; Santos,Lia Raquel de Souza.
This paper describes morphological characteristics of the ovarian germinative cells of the Scinax fuscovarius (Lutz, 1925). The ovary is organized in primordial germinative cells (oogonia) and follicular structures (ovarian follicle) - oocytes surrounded by follicular cells. Oogonia: their nests are peripherically localized, containing cells with large and oval nucleus. Oocytes I: basophilic cytoplasm; the spherical nucleus presents few nucleoli, one or two; the follicular cells (one tenuous layer) surround these previtellogenic cells. Oocytes II: the cell is larger and the cytoplasm becomes more basophilic; the nucleus presents few nucleoli and contains many chromosomes in the periphery (beginning the perinucleolar stage). Oocytes III: the cytoplasm...

Influence of climatic conditions on in vitro production of bovine embryos Arq. Bras. Med. Vet. Zootec.
Pessoa,G.A; Navarro,R.B; Rauber,L.P; Brass,K.E; Silva,C.A.M; Rubin,M.I.B.
Temperature and rainfall were analyzed daily during six years to evaluate their influence on in vitro production of bovine embryos. Weekly replications (n=480) were performed on 14,778 ovaries collected at slaughterhouses. Cumulus oocyte complexes (n=19,180) were fertilized with a pool of Bos taurus taurus semen in one incubator with 5% CO2. Presumable zygotes were cultured in gasified plastic bags with 5% CO2, 5% O2, and 90% N2. In the first year, cleavage and embryo yield were 60.3% and 15.6%, respectively, being lower (P<0.05) than in the following years. Average cleavage rates were always lower in winter (P<0.0001), thus producing less embryos. Winter climatic conditions had a negative influence on in vitro production, when cleavage and embryo...

Molecular Characterization, Electrophysiological and Contraceptive Effect of Chilean Latrodectus Venom International Journal of Morphology
Navarrete Gómez,Patricia; Ormeño,David; Miranda,Antonio; Sánchez Gutiérrez,Raúl; Romero Mejía,Fernando; Parodi Rivera,Jorge.
Since the 1970s, There have been studies of the venom of Latrodectus sp. spiders, in particular the latrotoxin (LTX) of Latrodectus mactans. Many of the studies were aimed at understanding the action of the venom on the muscular system. Now accepted that LTX is able to generate a calcium-permeable membrane pore and modulate the release of synaptic vesicles that activate a receptor and induce cellular changes. Interestingly, when work began with venom obtained from the Latrodectus sp present in Chile, it generated clinical indications similar to the bite of this spider in another country, with some differences in intensity. The purpose of the first studies was to understand the systemic mechanisms of this venom, and other active compounds were studied for...
