Alongamento e número de brotações de orquídeas cultivadas in vitro sob diferentes meios de cultura e concentrações. Repositório Alice

A família orquidácea corresponde a maior dentre as angiospermas e podem se desenvolver nos mais diferentes ambientes. Um fator que contribui para a dificuldade da propagação das orquídeas em condições naturais é a baixa ou a nula germinação de suas sementes na ausência de micorrizas. Objetivou-se com esse trabalho avaliar a multiplicação e alongamento in vitro de orquídeas CW 3709 (C. Walkeriana 12 & C. Walkeriana Lobomau).

Determination of endogenous IAA and carbohydrates during the induction and development of protocorm-like bodies of Cattleya tigrina A. Richard Biological Sciences

 Tissue culture techniques have been employed for orchid mass propagation by means of the morphogenetic route of protocorm like-bodies (PLBs). This study aimed to analyze and compare Indole-acetic acid (IAA) and sugar endogenous levels in protocorm-like bodies (PLBs) induction and development in Cattleya tigrina, in order to better understand this process and to optimize micropropagation procedure protocols. Leaves grown on MS (Murashige and Skoog) culture medium, suplemented with 9 μM TDZ for PLBs induction and development were collected after 0, 2, 7, 14, 30, 60 and 100 days of cultivation, for further analysis. Genetic analysis of species in the Genus Catasetum (ORCHIDACEAE) using RAPD Markers BABT

In this work, RAPD molecular markers were used to access the genetic variability and to study the inter and intraespecifc relationship in a group of 37 species, including 56 individuals. A total of 15 RAPD primers were selected for DNA amplification. From a total of 221 bands analyzed, 209 (95%) were polymorphics. The level of interespecifc genetic similarity ranged from 37% between Catasetum complanatum and Catasetum laminatum to 83% between Catasetum triodon and Catasetum uncatum. The intraspecifc genetic similarity varied 88% for the individuals of Catasetum triodon to 93% between the individuals of Catasetum atratum and Catasetum macrocarpum. A família orquidácea corresponde a maior dentre as angiospermas e podem se desenvolver nos mais diferentes ambientes. Os meios de cultura utilizados na micropropagação fornecem as substâncias essenciais para o crescimento e desenvolvimento in vitro de células, tecidos e órgãos vegetais e se baseiam nas exigências das plantas quanto aos nutrientes minerais. Com o presente trabalho objetivou-se avaliar o efeito do meio MS e Knudson na germinação in vitro de Aspasia variegata.

Na natureza, geralmente, não é possível obter a germinação de sementes de orquídeas sem a presença de fungos micorrízicos, sendo que apenas 5% dessas sementes chegam a formar novas plantas. No entanto, com o cultivo in vitro é possível a germinação assimbiótica de até 100% das sementes. Neste contexto, objetivou-se com o presente trabalho avaliar o efeito de diferentes meios de cultura sobre a germinação de sementes e crescimento inicial de plântulas de Sobralia Rondoni in vitro.

In vitro propagation of Anathallis adenochila (Loefgr.) F. Barros (Orchidaceae), a species endemic to southern and southeastern Brazil Acta Botanica

Anathallis adenochila is a small epiphytic orchid, endemic to the Atlantic Forest in the southern and southeastern regions of Brazil. Information on the species is scarce and limited to distribution and occurrence data, with no reports about its development. In the present study, plantlets were propagated in vitro, and the influence of different concentrations of macronutrient salts and sucrose on the survival and development of the species was assessed. Longevity of Epidendrum ibaguense inflorescences influenced by the storage temperature and ethylene inhibitor BABT

This work had the goal to evaluate the influence of temperature and dry storage on the postharvest longevity of Epidendrum ibaguense flowers previously treated with silver thiosulfate (STS). The flowers were harvested and pulsed for 30 minutes with 2 mM STS. Then both, the STS treated and untreated flowers were packed in perforated low density polyethylene bags, packed in cardboard boxes and stored at 5 and 10°C, where they remained for four and eight days. After this period, the flowers were kept in vase with distilled water at 25ºC, and analyzed at every 24 h. Radicular anatomy of twelve representatives of the Catasetinae subtribe (Orchidaceae: Cymbidieae) Anais da ABC (AABC)

Considering that the root structure of the Brazilian genera belonging to the Catasetinae subtribe is poorly known, we describe the roots of twelve representatives from this subtribe. For anatomical analysis, the roots were fixed in FAA 50, preserved in ethanol 70% and sectioned at its medium region using razor blades. The sections were stained with 0.05% astra blue and safranin and mounted in glycerin. For the identification of starch we used Lugol's solution; for lignin, floroglucin chloridric; for lipids, Sudan III, and for flavanoids, potassium hydroxide. Response of cattleya hybrids for Fusarium oxysporum f. sp. cattleyae Foster BABT

The Cattleya genus has a great importance in the flower agro-business market. Fusarium wilts, caused by Fusarium oxysporum f. sp. cattleyae, is considered one of the main factors of decline and death of plants of this genus. Using seven hybrids (intra and intergenerics) of Cattleya, tests of resistance and susceptibility to F. oxysporum were performed in conditions of greenhouse for 12 months, using, as evaluation criterion, a scale of the disease severity ranging from one (resistant) to eight (highly susceptible). Somatic embryos from Phalaenopsis classic Spotted Pink (Orchidaceae) protocorms Ciência Rural

ABSTRACT: The aim of the present study was to induce the formation of somatic embryos in protocorms from Phalaenopsis Classic Spotted Pink hybrids at two physiological maturation stages, namely: 80 and 120 days after seed inoculation (DASI). Protocorms were inoculated in ½ MS medium supplemented with 0.1 mg L-1 ANA and 3 mg L-1 TDZ. Protocorms were inoculated 120 days after sowing were more developed at the 15th cultivation day due to the formation of pro-embryogenic structures. It was possible seeing the formation of globular- and torpedo-stage somatic embryos at the 30th day of cultivation in somatic embryogenesis (SE) induction medium. In nature, orchid seeds obtain the nutrients necessary for germination by degrading intracellular fungal structures formed after colonization of the embryo by mycorrhizal fungi. Protocols for asymbiotic germination of orchid seeds typically use media with high concentrations of soluble carbohydrate and minerals. However, when reintroduced into the field, seedlings obtained via asymbiotic germination have lower survival rates than do seedlings obtained via symbiotic germination. The ground orchids ‘Pecteilis sp.’ have great potentials for development to commercial scale. This orchid group rest in dry season and flowering in rainy season, less taking care; so, it can be developed to pot plant similar to tulip. This research aims to develop new Thai orchid product for commercial scale by research and development. This work was done at Sisaket Horticulture Research Center during 2006-2011. It was collection of, the orchids ‘Pecteilis sp.’ from natural in north-east Thailand and market, 810 orchid plants included 500 plants of Pecteilis sagarikii and 310 plants of Pecteilis susannae. Moreover, four good parent lines were selected, and a pair of intergeneric hybrid between Pcteilis x Habenaria was bred for commercial scale.
