Genetic transformation allows the release of improved cultivars with desirable characteristics in a shorter period of time and therefore may be useful in citrus breeding programs. The objective of this research was to establish a protocol for genetic transformation of Valencia and Natal sweet oranges (Citrus sinensis L. Osbeck) and Rangpur lime (Citrus limonia L. Osbeck). In vitro regeneration of common bean (Phaseolus vulgaris L.) is a requirement for genetic transformation which involves induction and development to the whole plant. Success in regeneration of common bean from tissue organ culture has been achieved to some extent in the last years. However, genotype effects in regeneration response as well as efficiency and reproducibility have been a limiting factor. Avaliacao da capacidade de regeneracao in vitro em tomateiro industrial. Repositório Alice

Este trabalho teve por objetivo avaliar a capacidade de regeneracao das cultivares de tomateiro industrial (Lycopersicon esculentum Mill) IPA-5 e IPA-6, utilizando quatro composicoes de meio de cultura descritos na literatura e cinco variacoes de inoculacao. Foi testada uma nova variacao de inoculacao, denominada cotiledone fendido. A maior frequencia de formacao de gemas vegetativas foi 100% no caso de IPA-5, e 65% no caso de IPA-6. Para induzir o alongamento de brotos, foram necessarios tres subcultivos dos explantes apresentando gemas. Bauhinia forficata link shoot regeneration: histological analysis of organogenesis pathway BABT

Plant regeneration was achieved from cells of callus induced from hypocotyl segments of Bauhinia forficata on half strength Murashige and Skoog culture medium supplemented with several concentrations of BAP. Within 40 days of culture shoot buds formation was observed on callus surface. Calli were then transferred to a same composition culture medium without plant growth regulator in order to induce shoot elongation. Histological studies indicated that in vitro plant regeneration in B. forficata occurred through indirect organogenesis. A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. Different growth regulators combinations were tested on the production of anther callus in tomato cultivar IPA 5. Calli were induced on media supplemented with 1.0mgL-1 gibberellic acid (GA3), 0.05mgL-1 alpha-naphthaleneacetic acid (NAA) plus 0.1mgL-1 6-benzylaminopurine (BAP), or with 1.0mgL-1 BAP plus 1.0mgL-1 NAA. The medium containing 1.0mgL-1 BAP and 1.0mgL-1 NAA produced the highest calli frequency, and promoted plant regeneration by indirect organogenesis, when calli were transferred to 0.01mgL-1 BAP and 0.001mgL-1 NAA. The formation of the limbs, the digestive tract, the nervous system, the sensory and hormonal structures begins in this prawn during the embryonic stage C, according to Richard's definition (1974) and is performed at the stage H. The digestive tract differentiates with a caudo-cephalic gradient. The digestive gland is formed at least partially by vitellophages. The C.N.S. appears with a cephalo-caudal gradient and the regions of the eyestalk are quickly differentiated. The organ of Bellonci and the sensory pore appear independently during the stage F. The first neurosecretory cells are detected in the brain during the stage D.

The early stages of organogenesis in metazoans differ drastically between higher order taxa such as phyla and classes. The segmented germ band stage in insects, the nauplius stage of crustaceans, and the neurula/pharyngula stage in vertebrates are examples of this diversification. In striking contrast with this divergence, is the similarity of these stages within these taxa, i.e., within insects, crustaceans, and vertebrates. The early stages of organogenesis, or phylotypic stages, have, thus, remained very similar in most species since the evolutionary origin of the taxa. It is possible to determine the optimum time for permanence of vegetative propagules (mini-cuttings) inside a greenhouse for rooting, and this value can be used to optimize the structure of the nursery. The aim of this study was to determine the dynamics of adventitious rooting in mini-cuttings of three clones of Eucalyptus benthamii x Eucalyptus dunnii. Sprouts of H12, H19 and H20 clones were collected from mini-stumps that were planted in gutters containing sand and grown in a semi-hydroponic system. A organogênese é uma técnica pouco estudada na micropropagação de erva- mate. Este trabalho objetivou avaliar diferentes meios de cultura na calogênese in vitro e organogênese de Ilex paraguariensis St. Hil. Foram coletadas folhas em plantas em casa-de-vegetação. Segmentos foliares foram colocados em meios de cultura 1⁄4 MS, WPM ou JADS, contendo zeatina e 2,4-D. O meio MS foi mais eficiente na indução de calos. No meio WPM foi observada rizogênese. Nos meios testados não houve formação de brotações adventícias.

Growth curve and development of the internal calli structure of Eucalyptus camaldulensis Dehn BABT

The objective of this work was to elucidate the growth curve of Eucalyptus camaldulensis Dehn. calli analyzing their anatomical modifications. A sigmoid aspect of the growth curve of the calli fresh matter was observed, with five different phases (lag, exponential, linear, deceleration and decline). In the lag phase, the highest growth percentage 87%, was observed, which reduced during the evaluation period to 17% in the linear phase. As for the anatomical analyses, cellular multiplications was observed during the lag and exponential phases and increase in cell size during the linear phase, promoting the calli volume growth and the establishment of the globular conformation.

The initiation and development of organogenic buds and somatic embryos in HBsAg-transgenic cherry tomato (Lycopersicum esculentum var. cerasiforme) mutant were studied histologically. The leaf explants of the mutant were cultured on Murashige & Skoog (MS) basal medium supplemented with 6-BA 1.0 mg/L and IAA 0.05 mg/L for callus induction. The Y organ appears during the embryonic development of the prawn (P. serratus ) at a stage approximately C, according to Richard's definition (1974). 3 weeks separate this stage from hatching at a temperature of 20 degree C. The organ formation is realized over 48 hours and is marked by an histological modification and a small mitotic crisis in the epidermal tissue from which it differentiates. Its metameric localization is mainly maxillular.
