Angiogenesis and Tumor Progression Inhibition of Cyclooxygenase-2 Selective Inhibitor Celecoxib Associated with Poly (lactic-co-glycolic acid) in Tumor Cell Line Resistant to Chemotherapy International Journal of Morphology
Roa,Ignacio; Cantín,Mario; Vilos,Cristian; Rosas,Carlos; Lemus,David.
Although, antineoplastic therapies have now been developed reduction of tumor progression, itis necessarytofind new therapeutic alternatives to suppress angiogenesis. Thus celecoxib (Cx) has been used for its antiangiogenic action in combination with certain polymeric compounds such as poly (lactic co-glycolic acid) (PLGA) acid, which help to improve the bioavailability and avoid effects of long drug administrations. For this purpose we used a murine tumor modelinduced by mammary adenocarcinoma cells resistant to chemotherapy (TA3-MTXR). CX/PLGA inhibits the microvascular density, VEGF expression and cell proliferationinaddition to increased apoptosis (P <0.0001). Cx reduces tumor progression in a concentration of 1000 ppm associated with PLGA, reducing...

Development of VEGF-loaded PLGA matrices in association with mesenchymal stem cells for tissue engineering BJMBR
Rosa,A.R.; Steffens,D.; Santi,B.; Quintiliano,K.; Steffen,N.; Pilger,D.A.; Pranke,P..
The association of bioactive molecules, such as vascular endothelial growth factor (VEGF), with nanofibers facilitates their controlled release, which could contribute to cellular migration and differentiation in tissue regeneration. In this research, the influence of their incorporation on a polylactic-co-glycolic acid (PLGA) scaffold produced by electrospinning on cell adhesion and viability and cytotoxicity was carried out in three groups: 1) PLGA/BSA/VEGF; 2) PLGA/BSA, and 3) PLGA. Morphology, fiber diameter, contact angle, loading efficiency and controlled release of VEGF of the biomaterials, among others, were measured. The nanofibers showed smooth surfaces without beads and with interconnected pores. PLGA/BSA/VEGF showed the smallest water contact...

ABSTRACT: This study examined the efficacy of an intravitreal dexamethasone-loaded device for the control of postoperative ocular inflammation in dogs following phacoemulsification. Twenty dogs with bilateral mature senile cataracts were prepared for surgery using routine protocols. A biodegradable poly (lactic-co-glycolic acid) copolymer device was inserted through the pars plana into the vitreous chamber immediately before phacoemulsification (device group [DG], n=20). Following surgery, a conventional group (CG) received local and systemic steroids, mydriatics, and antibiotic therapy. The same treatment protocol was adopted in DG, except for steroids. All eyes were examined before surgery and at various times after phacoemulsification. Ultrasonography...

Pietra,Renata Celi Carvalho de Souza; Cruz,Rosana Carvalho; Melo,Carla Nunes; Rodrigues,Lívia Bomfim; Santos,Patrícia Campi; Bretz,Gabriel Pissolati Matos; Soares,Betânia Maria; Sousa,Gerdal Roberto de; Ferreira,Marcus Vinícius Lucas; Cisalpino,Patrícia Silva; Magalhães,Paula Prazeres; Farias,Luiz de Macêdo; Pinotti,Marcos.
ABSTRACT Antimicrobial photodynamic therapy (aPDT) involves the association of a photosensitizing agent with a light source with the goal of causing apoptosis or microbial lysing. The use of compounds with natural active principles is gaining prominence throughout the world. Several studies from groups that are linked to the development of innovations in the pharmaceutical market have used natural dyes, such as curcumin, the efficacy of which has been demonstrated in aPDT trials. Difficulties related to physicochemical stability, solubility and cell penetration are some of the challenges associated with this field. The present work aimed to prepare, investigate the characteristics and improve the photodynamic activity of PLGA-based nanoparticles loaded...

Acuña,Luis; Suazo Galdames,Iván; Zavando,Daniela; Elgueta,Samanta; Velásquez,Luis; Vilos,Cristian; Cantín,Mario.
The administration of microencapsulated drug in a matrix acid poly(lactic-co-glycolic acid) (PLGA) by intramuscular (IM) in humans has been approved by the FDA for various applications though it is not clear what effect they have on the morphological parameters of muscle tissue. The aim of this study was to analyze the morphological changes in the skeletal muscle tissue with their use. We used 12 adult female Sprague Dawley rats (Rattus novergicus) that were injected into their right gastrocnemius muscle belly with: sterile vehicle solution (G1, n = 4), 0.5 mg PLGA microparticle (G2, n = 4) and 0.75 mg PLGA microparticle (G3, n = 4), both dissolved in a sterile vehicle solution. At 14 days post injection the number and diameter of muscle fibers, the level...

Preparation and physicochemical characterization of prazosin conjugated PLGA nanoparticles for drug delivery of flutamide BJPS
Fattahi,Ali; Ghiasi,Mansoureh; Mohammadi,Pardis; Hosseinzadeh,Leila; Adibkia,Khosro; Mohammadi,Ghobad.
In the current work, a sustained drug delivery system of flutamide (FLT) was developed using Poly(D,L-lactide-co-glycolide) (PLGA) decorated bypoly(ethylene glycol) (PEG) grafted prazosin (PLGA-PEG-Praz) as a targeting moiety. In a multi-step reaction, PLGA was linked to PEG and prazosin. The structure of the synthesized polymers was confirmed by FTIR and 1H-NMR. Flutamide-loaded nanoparticles were prepared by quasi-emulsion solvent diffusion technique. The nanoparticles were evaluated for size, zeta potential, polydispersity index, drug crystallinity, loading efficiency, and release properties. Also, the physicochemical properties of the nanoparticles were analyzed using Scanning Electron Microscopy (SEM), Differential Scanning Calorimetry, and Powder...

Tadalafil-loaded PLGA microspheres for pulmonary administration: preparation and evaluation BJPS
Yang,Zhenlei; Wang,Longmei; Tian,Liu; Zhang,Xiuping; Huang,Guihua.
Tadalafil, a long-acting PED-5 inhibitor, is commonly used for the treatment of pulmonary arterial hypertension (PAH). However, its efficacy and clinical application are severely limited by the poor water solubility, low bioavailability and a series adverse effects (e.g. headaches, indigestion). In this study, tadalafil was prepared and loaded into biodegradable PLGA (poly(lactic-co-glycolic acid)) microspheres (TDF-PLGA-MS) via emulsification-solvent evaporation. The resulting microspheres were processed into pulmonary inhalant by freeze drying. The TDF-PLGA-MS was spherical and uniform, with an average particle diameter ~10.29 µm. The encapsulation efficiency and drug loading yield of TDF-PLGA-MS were 81.68% and 8.52%, respectively. The investigation of...
