Prunus persica has been proposed as a genomic model for deciduous trees and the Rosaceae family. Optimized protocols for RNA isolation are necessary to further advance studies in this model species such that functional genomics analyses may be performed. Here we present an optimized protocol to rapidly and efficiently purify high quality total RNA from peach fruits (Prunus persica). Isolating high-quality RNA from fruit tissue is often difficult due to large quantities of polysaccharides and polyphenolic compounds that accumulate in this tissue and co-purify with the RNA. Here we demonstrate that a modified version of the method used to isolate RNA from pine trees and the woody plant Cinnamomun tenuipilum is ideal for isolating high quality RNA from the...

Aspectos institucionais e organizacionais da agroindústria de doces de pêssego de Pelotas - RS AgEcon
Begnis, Heron Sergio Moreira; Zerbielli, Jerusa.
The production of desserts and fruit jellies is a traditional agribusiness activity in the south of Brazil, especially in Pelotas, Rio Grande do Sul. However, the Brazilian commercial opening that was processed in the beginning of the 1990’ years, it influenced negatively this segment, mainly in relation to the peach. This resulted in a new roll of conditions that culminated in the need of a revaluation of the transactional behavior among the agents involved in this segment. In this aspect, it is necessary to consider that the agribusiness system capacity of organization depends on the contractual arrangements that are established among them. In spite of these contractual relationships that causes an influence on the system efficiency it depends, not only...

Chemical evaluation and effect of bagging new peach varieties introduced in southern Minas Gerais - Brazil Ciênc. Tecnol. Aliment.
Lima,Annete de Jesus Boari; Alvarenga,Ângelo Alberico; Malta,Marcelo Ribeiro; Gebert,Deyse; Lima,Elisa Boari de.
Brazilian peach production is insufficient for domestic supply. Aiming to increase production in the state of Minas Gerais, 16 new peach tree varieties were introduced in the Serra da Mantiqueira region and their fruit were analyzed for sugar, total phenol and total carotenoid contents by colorimetric methods, and for organic acid content by high-performance liquid chromatography. In addition, we examined the effect of fruit bagging on the levels of such constituents. To do so, the fruit of half the trees of each variety were bagged. The levels of sugars, phenols, carotenoids and organic acids are genotypic characteristics and significantly differed among varieties. Despite being a good form of fruit protection, providing better aspect and reducing the...

Evaluation of a Non-Destructive Impact Sensor to Determine On-Line Fruit Firmness Chilean J. Agric. Res.
Homer,Ian; García-Ramos,Francisco Javier; Ortiz-Cañavate,Jaime; Ruiz-Altisent,Margarita.
A non-destructive impact sensor to measure on-line fruit firmness was evaluated. This sensor is an adaptation of a static model used in the laboratory to measure fruit quality and was installed in an experimental fruit packing line with a commercial sizer chain. The firmness index is related to the acceleration-time curve supplied by an accelerometer attached to an impacting arm. The main objective of this study was to evaluate sensor performance and sources of variation. We made classification trials on three fruits: peaches (Prunus persica (L.) Batsch), apples (Malus domestica Borkh.), and pears (Pyrus communis L.), as well as working trials, such as placing the fruit, orientation, and others. The sensor works correctly at a speed of 7 fruits s-1 (0.63 m...

Factors influencing the adoption of improved cultivars: a case of peach farmers in Pakistan Ciência Rural
Ullah,Arif; Khan,Dilawar; Zheng,Shaofeng; Ali,Uzair.
ABSTRACT: This study aimed to investigate factors influencing the adoption of improved cultivars (ICs) in peach production in Khyber Pakhtunkhwa province of Pakistan. A total of 270 respondents were randomly selected from the three different cultivated areas of Khyber Pakhtunkhwa, namely, Peshawar, Nowshera and Swat. Binary choice model was used in this study to categorise the ICs of peach farmers into adoption and non-adoption. The study identifies that socio-economic, institutional farm resources, and climatic factors are influencing the adoption of ICs of peach production. Results of the estimated model reveal that farmer’s age, education, household size, membership, cell phone, farm size, extension services and the role of the non-government...

GIBBERELLIC ACID REDUCES FLOWERING AND TIME OF MANUAL THINNING IN ‘MACIEL’ PEACH TREES Rev. Bras. Frutic.
GIOVANAZ,MARCOS ANTÔNIO; FACHINELLO,JOSÉ CARLOS; SPAGNOL,DANIEL; WEBER,DIEGO; CARRA,BRUNO.
ABSTRACT The thinning peach fruits is to reduce the load of the plant with the aim to improve the size, flavor and color of the remaining fruits. However, the practice of thinning requires high demand of manpower and become more expensive the production costs. The objective of this study was to evaluate the reduction of flowering buds and the time of hand thinning in ‘Maciel’ peach after gibberellic acid (GA3) application at different times and concentrations. The experiments were performed in the harvests of 2012 and 2013 in a commercial peach orchard located in the county of Morro Redondo, RS. The plants of cultivar Maciel, in 2012, were sprayed with four doses of GA3 0, 25, 75 and 125 mg L-1, at different times 20, 40 and 60 days after full bloom...

Polyphenoloxidase activity in peaches after vibration REA
Dantas,Tiago B. H.; Ferraz,Antônio C. de O.; Honório,Sylvio L.; Lima,Giuseppina P. P..
The vibration during transportation is one of the main causes of fruit damage. The actual methodologies used for damage evaluation in fruits seem to be subjective, since most of them are based on visual evaluation. In this study the effect of vibration, for one and two hours, on polyphenoloxidase (PPO) activity in peach pulp was investigated. The relation among PPO activity variation, postharvest time and resting time were also considered. Results showed that vibration affects PPO activity. However, it was not possible to correlate PPO activity variation and vibration.
