Extracellular ATP in the lymphohematopoietic system: P2Z purinoceptors and membrane permeabilization BJMBR
Persechini,P.M.; Bisaggio,R.C.; Alves-Neto,J.L.; Coutinho-Silva,R..
The effects of extracellular nucleosides and nucleotides on many organs and systems have been recognized for almost 50 years. The effects of extracellular ATP (ATPo), UTPo, ADPo, and other agonists are mediated by P2 purinoceptors. One of the most dramatic effects of ATPo is the permeabilization of plasma membranes to low molecular mass solutes of up to 900 Da. This effect is evident in several cells of the lymphohematopoietic system and is supposed to be mediated by P2Z, an ATP4--activated purinoceptor. Here, we review some basic information concerning P2 purinoceptors and focus our attention on P2Z-associated phenomena displayed by macrophages. Using fluorescent dye uptake, measurement of free intracellular Ca2+ concentration and electrophysiological...

Statistical investigation of Kluyveromyces lactis cells permeabilization with ethanol by response surface methodology BJM
Faria,Janaína T. de; Rocha,Pollyana F.; Converti,Attilio; Passos,Flávia M.L.; Minim,Luis A.; Sampaio,Fábio C..
The aim of our study was to select the optimal operating conditions to permeabilize Kluyveromyces lactis cells using ethanol as a solvent as an alternative to cell disruption and extraction. Cell permeabilization was carried out by a non-mechanical method consisting of chemical treatment with ethanol, and the results were expressed as β-galactosidase activity. Experiments were conducted under different conditions of ethanol concentration, treatment time and temperature according to a central composite rotatable design (CCRD), and the collected results were then worked out by response surface methodology (RSM). Cell permeabilization was improved by an increase in ethanol concentration and simultaneous decreases in the incubation temperature and treatment...

The production of sorbitol by permeabilized and immobilized cells of Z. mobilis in sucrose BABT
Vignoli,Josiane Alessandra; Celligoi,Maria Antonia Colabone; Silva,Rui Sérgio Ferreira da; Barros,Márcio de.
The production of sorbitol by permeabilized and immobilized cells of Zymomonas mobilis in Luffa cylindrica was investigated in sucrose medium. A full 2³ factorial design was used to verify the influence of each factor and its interactions. The cell permeabilization showed a significant and negative effect upon the production of sorbitol, while the time of cultivation and the immobilization process were significant and positive. The results demonstrated that the cell immobilization and the time of cultivation of 36 h presented higher production of sorbitol.
