The activities of laccase, Mn-peroxidase, phosphatase and dehydrogenase were studied in a Chilean Andisol (Temuco Serie) contaminated with pentachlorophenol (PCP). Experiments were conducted on the top of 0-20 cm soil, adding different concentrations of PCP (0.41, 4.1 and 41 mg kg-1). Enzymatic activities and residual concentrations of PCP were evaluated in soil incubated at 25 ± 1 ºC for 112 days. Enzyme activities of PCP-treated soil varied according to the PCP concentration added. For instance, an inhibitory effect on Mn-peroxidase activity was not observed during incubation at the three levels of PCP, whereas acid phosphatase activity was only affected by the 0.41 mg kg-1 PCP treatment. The objective of this work was to study the effect of grazing systems: rotational, continuous and the absence of cattle, in pastures of Brachiaria brizantha on microbial activity (respiratory, potential nitrification, solubilizing, and enzymatic) and on the chemical attributes (contents of organic carbon, total nitrogen, organic phosphorus, and moisture) of Eutrustox soil over one year. Eight Canchim breed animals were used. The samples were collected in the four seasons from 2005 to 2006. An Aspergillus niger UFV-1 phytase was characterized and made available for industrial application. The enzyme was purified via ultrafiltration followed by acid precipitation, ion exchange and gel filtration chromatography. This protein exhibited a molecular mass of 161 kDa in gel filtration and 81 kDa in sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE), indicating that it may be a dimer. It presented an optimum temperature of 60 °C and optimum pH of 2.0. The KM for sodium phytate hydrolysis was 30.9 mM, while the kcat and kcat/KM were 1.46 ×105 s−1 and 4.7 × 106s−1.M−1, respectively. ABSTRACT Mg2+-ATPase activity was detected in the three salivary glands of adult triatomines, males and females, of Triatoma infestans (Klug, 1834) and Panstrongylus megistus (Burmeister, 1835) (Heteroptera, Triatominae). A predominance of binucleated cells in D1 and D2 and mononucleated in D3 was observed, with bulky and polyploidy nuclei. ATPase activity was detected in the nuclei, possibly in euchromatin and nucleolus, where this enzyme probably acts in the transcription process. ATPase reaction was also evidenced in the nuclear membrane, which is probably associated with nuclear-cytoplasmatic transport. The number of colony forming units (CFU) of different groups of bacteria and fungi in samples stored at temperatures of 5 to -12oC for 0-32 weeks was evaluated. The number of CFU obtained after the different periods of storage of red latossol soil was compared with the number of colonies obtained immediately after removal of soil samples (time zero). The number of total bacteria and actinomycetes in the samples remained practically unchanged throughout the storage period. The number of Gram-negative bacteria decreased by as much as 69% compared to control, while the number of Bacillus spp and of fungi increased 1.9 to 4.9 times starting from the 12th week in samples stored at 5oC. At present, reforestation has focused on native forests with anthropogenic intervention and eroded soils. There is interest in producing Nothofagus seedlings which can overcome adverse conditions encountered on reforestation sites. It is necessary to find new fungi that can be utilized as mycorrhizal inoculants and that enable the seedlings to increase their tolerance to adverse conditions. Two ectomycorrhizal strains of the fungus Descolea antarctica (D1 and D2) were cultured at different temperatures, pH levels and the activities of amylases, cellulases, and phosphatases were determined. Many enzymes produced by fungi have relevant biotechnological applications in several industrial areas. The purpose of this study was to collect and isolate filamentous fungi from soil and humus, plants and sugar cane bagasse of different regions of the São Paulo state. Forty isolates were examined for their ability to produce xylanase, glucose-oxidase, alkaline phosphatase, acid phosphatase, phytase, pectinase and amylase. Among these, twenty three isolates exhibited enzymatic potential. The xylanases produced by two of these isolates (Aspergillus caespitosus and A. phoenicis) showed good potential for pulp bleaching. Some plant species can change soil phosphorus (P) availability and this may be an important tool in managing tropical high fixing phosphorus soils. An experiment was conducted to evaluate phosphorus transformations in the soil and phosphatase activity during periods of Congo grass (Brachiaria ruziziensis, Germain et Evrard) growth in two tropical soils receiving 20, 40, 80, 160 mg dm-3 of inorganic P. Ultrastructural cytochemical techniques and electron microscopy were used for localization of acid phosphatase activity during spermiogenesis in Eriocheir sinernsis. The results showed that: Acid phosphatase was synthesized in the endoplasmic reticulum in the early spermatids. The acid phosphatase was found gradually in nucleus, the membrane of acrosomal vesicle, the cytoplasmic region and the acrosomal tubule. And then the reaction product particles became thicker during the spermiogenesis. In the mature sperm, acid phosphatase was localized in the percutor organ slightly, but it was massive and compact in the acrosomal tubule. |

The purpose of this study was to evaluate the microbiological activity, inorganic phosphorus solubilization (Pi) and acid phosphatase in the cultivated soil, in a combination of soybean (Glycine max) plants and weeds.
