There is currently great interest in the cultivation of the Jatropha curcas L., known locally as physic nut, for the production of biodiesel, leading to an increase in the demand for seeds and precise information as to their physiological quality, especially vigour. The objective was to evaluate the efficiency of the accelerated ageing test, with variations, in verifying the vigour of Jatropha seeds. Four seed lots were initially characterized as to their physiological quality by germination and vigour tests. For accelerated ageing test, temperatures of 42 and 45 ºC were tested during 48, 72 and 96 hours. The experiment was carried out in a completely randomized design and the averages of the treatments compared by Tukey test (p<0,05). The Pearson... |

Accelerated aging is one of the most useful tests used for the evaluation of seed vigor but it is seldomly used to test melon (Cucumis melo L.) seeds. The objective this research was to compare different procedures of the accelerated aging test to evaluate the physiological quality of melon seeds and the efficiency of using saturated salt solution for the control of water uptake by seeds. Five seed lots each of the hybrids AF-646 and AF-682 were tested for germination, seedling emergence, traditional accelerated aging (periods 0f 48, 72 and 96 hours, at 38 or 41°C) and salt saturated accelerated aging. The accelerated aging test (traditional procedure and with salt solution) for 72h and 96h, at 38 or 41°C was sensitive to detect differences in the... |

ABSTRACT In carrot seed production, harvesting of the umbels is generally staggered, or sometimes the whole plant is harvested with the umbels at different stages of maturation; in that case, there is a risk of harvesting immature seeds and of losses due to natural dehiscent, or to physiological deterioration as a result of unfavourable environmental factors. The present study aimed to evaluate the physiological and biochemical changes related to the maturation of 'Planalto' carrot seeds from diferent umbels. The samples were collected 120 days after anthesis, with the primary, secondary and tertiary umbels being harvested separately. Seeds were analysed for germination, first count and germination rate, at temperatures of 25 ºC (optimum) to 35 ºC... |

ABSTRACT Seed companies increasingly need to obtain rapid and reliable results on seed quality. In this context, this study aimed at verifying the efficiency of CO2 measurement for classifying West Indian gherkin (Cucumis anguria L.) seed vigor, using three seed lots of cv. “Do Norte” and three of cv. “Nordestino”. In order to assess seed physiological quality, the following tests were performed: germination, seedling emergence, emergence speed index, shoot length, total dry matter, electrical conductivity and respiratory activity. The experimental design was completely randomized, with four replicates, and the means were compared by Tukey test at 5% probability level. Pearson’s simple correlation was performed between the traditional vigor tests and the... |

Coffee is one of the main agricultural commodities in the country, and it is important to conservation of plant material for breeding programs. Cryopreservation is a promising alternative for preserving in the long-term the germplasm of species considered recalcitrant. However, studies should be performed to achieve maximum survival of seedlings after immersion in liquid nitrogen. The objective of this work was to find a cryopreservation protocol for storing seeds of Coffea canephora, studying two methods of cryopreservation, slow and fast cooling. Seeds were subjected to drying in silica gel up to the water content of 0.25 g g-1. In the first experiment, dried seeds were subjected to treatments of slow cooling at speeds of -1 ºC min-1,-3 ºC min-1 and -5... |

Maturity stage and drying method are the factors that most influence coffee seed quality. The objective of this study was to assess the physiological quality and investigate the electrophoretic patterns of catalase and endo-ß-mannanaseenzymes and heat resistant proteins in coffee seeds harvested at different phenological stages and dried under different conditions. Physiological quality was assessed when the seeds had developed the green, greenish-yellow, cherry, overripe and dry stages after three treatments: no drying, conventional drying and fast drying. After each treatment, the physiological quality of the seeds was assessed using the germination test and electrophoretic patterns of heat resistant proteins and the activity of catalase and... |

ABSTRACT The electrical conductivity test enables the evaluation of seed vigor for a maximum period of 24 hours, but the need to count the seeds to obtain a sample for analysis makes it difficult to apply this test to small seeds, such as forage grasses. The objective of this study was to establish a practical methodology for testing the electrical conductivity of Brachiaria brizantha (Hochst. Ex A. Rich.) Stapf ‘Marandú’ seeds and to estimate its effect on seedling emergence in the field. The water content, germination, first germination count, first emergency count, percentage, and rate of seedling emergence speed in the laboratory, seedling emergence in the field, and electrical conductivity of eight batches of seeds was analyzed. In the last test, the... |

ABSTRACT This study aimed at assessing the quality of the physiology and health of the coriander seeds processed in a gravity table. This was investigated at two settings namely, I with angle X 3.5° and angle Y 3.1° and II with 3.9° and 2.7° angles, with five gravity table discharges (high, high intermediate, intermediate, low intermediate and low) besides feeding. The experiment adopted the 2x6 factorial scheme, in a completely randomized design, with four replications. Two coriander seed batches of cultivar Verdão were used. The gravity table was adjusted to have 2.9 m s-1 air speed on the platform. The seed fractions discharged in the lower portion of the gravity table revealed poorer physiological performance than that of the others. Regulation I... |
