Abstract The acerola tree (Malpighia emarginata Sessé &Moc. ex DC.) is a fruit species that has prospered in the Brazilian tropical regions, especially in the Northeast, thanks to the high content of vitamin C in the fruits, among other characteristics that make it attractive to the producer, such as early production. Although there is increasing demand for the product, the production of certified seedlings of good commercial varieties is still a barrier to further expansion of the crop. Over the last few years, considerable effort has been spent by Embrapa in the construction of a plant disease database representative enough for the development of effective methods for automatic plant disease detection and recognition. In October of 2016, this database, called PDDB, had 2326 images of 171 diseases and other disorders affecting 21 plant species. PDDB size, although considerable, is not enough to allow the use of powerful techniques such as deep learning. In order to increase its size, each image was subdivided according to certain criteria, increasing the number of images to 46,513. Macrophomina is a genus belonging to Botryosphaeriaceae that comprises well-known necrotrophic pathogens related to hundreds of plant hosts around the world. Historically, M. phaseolina is the causal agent of charcoal rot in several crops, mainly in tropical and subtropical areas around the world. However, after a recent genetic diversity study using morphological and molecular approaches, which resulted in the epitypification of M. phaseolina, and the description of a new Macrophomina species associated with charcoal rot disease, the hypothesis that other cryptic species could be present under the name M. phaseolina was raised. Cucurbits have great economic, nutritional and social importance in the Brazilian semiarid region. In this region, many factors can result in reduced productivity of these crops, especially fungal and viral diseases. Therefore, knowledge of cucurbits is crucial for proper disease management. The objective of this work was to identify the major diseases of cucurbits grown in some municipalities in the Hinterland of the state of Pernambuco. Thus, plant samples were collected with symptoms in crops in the municipalities of Salgueiro, Serra Talhada, Floresta, Petrolândia, Ibimirim, Custódia and Inajá. To assess the diversity of Phytophthora species affecting cacao in Brazil, 40 new isolates were obtained from cacao pods exhibiting symptoms of black pod disease collected in different smallholder farms in 2017. Further, ten cacao-infecting isolates morphologically identified as P. citrophthora and P. palmivora were molecularly characterized. The genomic regions beta-tubulin, elongation factor 1 alpha, heat shock protein 90, and internal transcribed spacer, and the mitochondrially encoded cytochrome c oxidase I and II genes were PCR-amplified and Sanger-sequenced from the cacao-infecting Phytophthora isolates. |
