Callus induction and plant regeneration of the endemic Astragalus nezaketae in Turkey Electron. J. Biotechnol.

A callus induction and plant regeneration protocol was developed from leaf and petiole explants of the endemic Astragalus nezaketae. Explants were cultured on Murashige and Skoog medium (MS) supplemented with different plant growth regulators (PGRs) [?-naphthaleneacetic acid (NAA), benzyladenine (BA), 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin (Kin), thidiazuron (TDZ)]. The combinations and concentrations of PGRs were shown significant variations for the frequency of callus formation, appearence of callus and the potential of callus differentiation. Callus induction and plant regeneration of Ulex europaeus Electron. J. Biotechnol.

A callus induction and plant regeneration protocol was developed from leaf and thorn explants for the plant Ulex europaeus. Explants were incubated on 2% sucrose half-strength Murashige and Skoog Medium (MS) with various combinations of plant growth regulators and antioxidants. The best frequency of callus and shoot formation was obtained with 2,4-dichlorophenoxyacetic acid (2,4-D) 1 mg/l x kinetin (Kin) 0.2 mg/l (DK Medium; callus induction) and zeatin (Z) 1 mg/l (DK medium; shoot induction). Both media were supplemented with ascorbic acid 200 mg/l to prevent browning and death of the explants. Endogenous auxin and cytokinin contents associated with shoot formation in leaves of pineapple cultured in vitro Braz. J. Plant Physiol.

The in vitro culture of pineapple leaves on a shoot induction medium (SIM) results in the formation of protuberances and further development in shoots, and plantlets. The contents of endogenous indoleacetic acid (IAA) and five cytokinins (Cks), N6(2-isopentenyl)adenine (iP), N6(2-isopentenyl)adenosine (iPR), zeatin (Z), zeatin riboside (ZR) and N6-benzyladenine (BA), present in the basal portion of those leaves, were correlated to the organogenic response that occurs over 15 days of culture. The endogenous auxin/cytokinins ratio was lowest on the 3rd day, mainly due to a strong increase in the iP level. Oil palm (Elaeis guineensis Jacq.) is the highest yielding oil-bearing crop. However, being a perennial crop, genetic improvement of oil palm is extremely slow. Indeed, compared to other annual oil crops such as soybean and rapeseed, genetic manipulations remained less important. Therefore, to remain competitive, oil palm growers and breeders need new and novel approaches. Celosia argentea (Var.) cristata (Amaranthaceae) is a widely cultivated ornamental plant, which has antibacterial, astringent, haemostatic, hypertensive, ophthalmic, and parasitic significance. This study describes a protocol for in vitro callus induction and plant regeneration from leaf and stem explants of C. argentea using Murashige and Skoog (MS) medium. ABSTRACT The present study reports a shoot organogenesis-based system for in vitro regeneration of Passiflora miniata, an Amazonia passion fruit species. Root segments were cultured in Murashige and Skoog (MS) medium supplemented with different concentrations (range 2-9 µM) of 6-benzyladenine (BA); thidiazuron (TDZ) or kinetin (KIN). Plant growth regulators were not added to the control treatment. Root explants have showed a high regenerative potential. After 30 days of in vitro culture, the root explants showed several shoots formed direct and indirectly. Influence of genotype and explant source on indirect organogenesis by in vitro culture of leaves of Melia azedarach L. Biocell

In vitro regeneration of shoots from leaf explants of the Paradise tree (Melia azedarach L.) was studied. Three different portions (proximal portion, distal portion and rachis of the leaflets) of three developmental stages (folded, young still expanding and completely expanded) of leaves of 10 - 15 years old plants of seven genotypes were cultured on Murashige and Skoog (1962) medium (MS) supplemented with 1mg.l-1 benzylaminopurine (BAP) + 0.1mg.l-1 kinetin (KIN) + 3 mg.l-1 adenine sulphate (ADS). Callus cultures from cotyledon and leaf explants of a Spanish cultivar of melon (Amarillo Oro) were tested for growth and morphogenic capacity on several culture media with different concentrations of IAA (indole-3-acetic acid) in combination with 1.0 mg.L-1 BA (6-benzylaminopurine) or 6.0 mg.L-1 KIN (kinetin). The best results were achieved with cotyledon explants. The leaf explants presented low bud formation capacity. High salt concentrations in soils negatively affect maize growth. Techniques such as remote hybridization and in vitro selection have been extensively used to accelerate breeding processes. In order to determine the usefulness of Tripsacum to improve salt tolerance in maize, the effects of NaCl, in vitro and in vivo, were evaluated in an intergeneric hybrid (MT) obtained from crossing Zea mays with Tripsacum dactyloides. The purposes of this work were to obtain embryogenic cell suspensions (ECS) from scalps and to regenerate plants of the banana CIEN-BTA-03. Shoot apexes were grown in the scalp-induction medium of Murashige and Skoog plus BA and IAA, following four diverse treatments. The first two, ME22 and ME25, were solid media supplemented with (mg L-1) 22.7 BA plus 0.192 IAA, and 25 BA plus 0.217 IAA, respectively, all containing 1.8 g L-1 of phytagel, and subcultures were performed monthly and bimonthly over 16 months. ABSTRACT Shoot regeneration, callus growth, and biosynthesis of shikonin in callus cultures of Onosma sericeum were examined. Plant tissue culture was used as an alternative method for increasing the production of shikonin, a secondary metabolite. The isolated cultures were subjected to abiotic factors such as light, plant growth regulators, and nutritional factors. Identification was carried out by High- Performance Liquid Chromatography (HPLC) after 10th subculture. Plant regeneration of natural tetraploid Trifolium  Hum pratense L Biol. Res.

The regeneration of natural tetraploid T. pratense, originated from Erzurum-Turkey, is reported in this study. This plant has low seed setting and hard seed problems due to polyploidy. Hypocotyl, cotyledon, apical meristems, epicotyl and young primary leaves were inoculated on MS and PC-L2 media containing different concentrations of BAP and NAA as growth regulators. The best shoot formation has been observed on explants initiated from apical meristem placed on PC-L2 medium that includes 2 mg dm-3 BAP and 1 mg dm-3 NAA. 94.4% of the shoots originated from calli were rooted on PC-L2 medium with 1 mg dm-3 NAA.
