Angiotensin-(1-7) potentiates the coronary vasodilatatory effect of bradykinin in the isolated rat heart BJMBR
Almeida,A.P.; Frábregas,B.C.; Madureira,M.M.; Santos,R.J.S.; Campagnole-Santos,M.J.; Santos,R.A.S..
It has been shown that angiotensin-(1-7) (Ang-(1-7)) infusion potentiates the bradykinin (BK)-induced hypotensive response in conscious rats. The present study was conducted to identify Ang-(1-7)-BK interactions in the isolated rat heart perfused according to the Langendorff technique. Hearts were excised and perfused through the aortic stump under a constant flow with Krebs-Ringer solution and the changes in perfusion pressure and heart contractile force were recorded. Bolus injections of BK (2.5, 5, 10 and 20 ng) produced a dose-dependent hypotensive effect. Ang-(1-7) added to the perfusion solution (2 ng/ml) did not change the perfusion pressure or the contractile force but doubled the hypotensive effect of the lower doses of BK. The BK-potentiating...

Blastocyst-endometrium interaction: intertwining a cytokine network BJMBR
Castro-Rendón,W.A.; Castro-Álvarez,J.F.; Guzmán-Martinez,C.; Bueno-Sanchez,J.C..
The successful implantation of the blastocyst depends on adequate interactions between the embryo and the uterus. The development of the embryo begins with the fertilized ovum, a single totipotent cell which undergoes mitosis and gives rise to a multicellular structure named blastocyst. At the same time, increasing concentrations of ovarian steroid hormones initiate a complex signaling cascade that stimulates the differentiation of endometrial stromal cells to decidual cells, preparing the uterus to lodge the embryo. Studies in humans and in other mammals have shown that cytokines and growth factors are produced by the pre-implantation embryo and cells of the reproductive tract; however, the interactions between these factors that converge for successful...

Chagas disease: Present status of pathogenic mechanisms and chemotherapy Biol. Res.
Maya,Juan Diego; Orellana,Myriam; Ferreira,Jorge; Kemmerling,Ulrike; López-Muñoz,Rodrigo; Morello,Antonio.
There are approximately 7.8 million people in Latin America, including Chile, who suffer from Chagas disease and another 28 million who are at risk of contracting it. Chagas is caused by the flagellate protozoan Trypanosoma cruzi. It is a chronic disease, where 20%-30% of infected individuals develop severe cardiopathy, with heart failure and potentially fatal arrhythmias. Currently, Chagas disease treatment is more effective in the acute phase, but does not always produce complete parasite eradication during indeterminate and chronic phases. At present, only nifurtimox or benznidazole have been proven to be superior to new drugs being tested. Therefore, it is necessary to find alternative approaches to treatment of chronic Chagas. The current treatment...

Effects of two d-cloprostenol administrations given at different intervals on estrus and ovulation in cyclic dairy goats. Repositório Alice
BONATO, G. C.; CÔRTES, L. R.; ARRAIS, A. M.; FIGUEIRA, L. M.; OLIVEIRA, T. de A.; MACHADO, V. de O.; MAIA, A. L. R. e S.; SOUZA-FABJAN, J. M. G.; FONSECA, J. F. da.
Abstract: Goats are seasonal breeders depending on breed and latitude. Among the hormonal methods used in controlled reproduction programs, there are those that mimic the activity of the corpus luteum and those that inhibit it. Prostaglandin (PGF2?) fits the latter, with the advantage of having a better cost-benefit, since it is cheaper and more natural method, as it has fewer side effects than progestin devices. In addition, the literature is incipient regarding follicular and ovulatory data in goats subjected to PGF2? or its analogues. This study aimed to check in dairy goats the efficiency of estrus synchronization treatment using two doses of d-cloprostenol administered in different intervals, during the natural breeding season. The results quoted...

Eicosanoids in Insect Biology Neotropical Entomology
STANLEY,DAVID W.; NOR ALIZA,A.R.; TUNAZ,HASAN; PUTNAM,SEAN M.; PARK,YOUNGJIN; BEDICK,JON C..
Prostaglandins and related eicosanoids are oxygenated metabolites of certain C20 polyunsaturated fatty acids. Eicosanoids are best understood in the context of their clinical significance in human medicine. We suggest a new and broader view of eicosanoids, which we have been calling a 'biological paradigm'. Under this view, we note that eicosanoids were taken into roles as cellular signal moieties long before the origins of the Metazoa. During the evolutionary diversification of animals, eicosanoids have been recruited into an array of biological roles, some of which occur only in insects and other invertebrates. These multiple actions endow eicosanoids with unusual explanatory power in understanding biological phenomena. We review the roles of eicosanoids...

Immunoexpression of cyclooxygenase-1 and -2 in ulcerative colitis BJMBR
Paiotti,A.P.R.; Artigiani Neto,R.; Forones,N.M.; Oshima,C.T.F.; Miszputen,S.J.; Franco,M..
Ulcerative colitis (UC) is a disease of the colon and rectum characterized by a nonspecific chronic inflammation mediated by the concerted response of cellular and humoral events. Prostaglandins are synthesized by cyclooxygenase (COX)-1 and -2 and exhibit both pro- and anti-inflammatory activity. To evaluate COX-1 and COX-2 immunoexpression in 42 cases of UC and to correlate it with clinicopathological parameters, COX-1 and COX-2 expression was investigated by the immunohistochemistry method. Only patients with all pertinent clinical and evolutive data as well as with adequate biopsy material were included in the study. Fifteen samples of colorectal adenocarcinoma and 14 of large bowel with no histological changes were used for positive and negative...

Inhibition of Mayaro virus replication by prostaglandin A1 and B2 in Vero cells BJMBR
Ishimaru,D.; Marcicano,F.G.P.; Rebello,M.A..
The effect of prostaglandins (PGA1 and PGB2) on the replication of Mayaro virus was studied in Vero cells. PGA1 and PGB2 antiviral activity was found to be dose-dependent. However, while 10 µg/ml PGB2 inhibited virus yield by 60%, at the same dose PGA1 suppressed virus replication by more than 90%. SDS-PAGE analysis of [35S]-methionine-labelled proteins showed that PGA1 did not alter cellular protein synthesis. In infected cells, PGA1 slightly inhibited the synthesis of protein C, while drastically inhibiting the synthesis of glycoproteins E1 and E2.

Prostaglandin A1 inhibits the replication of bovine viral diarrhea virus BJM
Caldas,Lúcio Ayres; Freitas,Tânia Rosária Pereira; Azevedo,Renata Campos; de Souza,Wanderley.
ABSTRACT Bovine viral diarrhea virus can cause acute disease in livestock, leading to economic losses. We show that Prostaglandin A1 inhibits bovine viral diarrhea virus replication in Madin-Darby bovine kidney cells (94% inhibition using 5 µg/mL). Light and electron microscopy of infected cells shows that Prostaglandin A1 also prevents virus-induced vacuolization, but at higher concentrations (10 µg/mL).

Prostanoids modulate inflammation and alloimmune responses during graft rejection BJMBR
Rocha,P.N.; Carvalho,E.M..
Acute rejection of a transplanted organ is characterized by intense inflammation within the graft. Yet, for many years transplant researchers have overlooked the role of classic mediators of inflammation such as prostaglandins and thromboxane (prostanoids) in alloimmune responses. It has been demonstrated that local production of prostanoids within the allograft is increased during an episode of acute rejection and that these molecules are able to interfere with graft function by modulating vascular tone, capillary permeability, and platelet aggregation. Experimental data also suggest that prostanoids may participate in alloimmune responses by directly modulating T lymphocyte and antigen-presenting cell function. In the present paper, we provide a brief...

Reproductive parameters of dairy goats submitted to estrus synchronization with prostaglandin F2alfa associated or not to hCG at estrous onset. Repositório Alice
ESTEVES, L. V.; BRANDÃO, F. Z.; CRUZ, R. C.; SOUZA, J. M. G.; OBA, E.; FACO, O.; FONSECA, J. F. da.
Abstract: The objective of this study was to evaluate the efficiency of two doses of PGF associated or not to hCG on the associated reproductive parameters in dairy goats. A total of 29 goats received two doses of 30 ug d-cloprostenol latero-vulvar at a 10 day intervals (Day 1 and Day 10). The does were allocated according to body weight and body condition score into two treatments, to receive hCG (250IU) or saline at estrus onset. After the second dose of PGF, estrus was monitored and ultrasound exams were performed twice daily. All does were inseminated 16h after estrus onset. Blood collection was performed every day for progesterone assay. The use of hCG at estrus onset did not affect any studied parameter and therefore the data were pooled. Estrous...

Sincronização do estro de novilhas caprinas mestiças com PGF2a: dose e sub-dose em duas vias de aplicação. Repositório Alice
REGO, J. P. A.; SANTOS, D. O.; FARIAS, L. S.; OLIVEIRA, L. S..

The influence of dietary supplementation of arachidonic acid on prostaglandin production and oxidative stress in the Pacific oyster Crassostrea gigas ArchiMer
Seguineau, Catherine; Racotta, Ilie; Palacios, Elena; Delaporte, Maryse; Moal, Jeanne; Soudant, Philippe.
In a previous study, dietary supplementation with arachidonic acid (ARA) to oysters Crassostrea gigas increased haemocyte numbers, phagocytosis, and production of reactive oxygen species level (ROS) by haemocytes (Delaporte et al., 2006). To assess if the observed stimulation of these cellular responses resulted from changes of ARA-related prostaglandin (PG) production, we analysed prostaglandin E2 metabolite (PGEM) content on the same oysters fed three levels of ARA. Dietary supply of polyunsaturated fatty acids (PUFA) could also induce an oxidative stress that could similarly increase cellular responses; therefore, two indicators of oxidative stress were analysed: peroxidation level and antioxidant defence status. Together the observed positive...
