Abstract: We have established how natural compounds from green propolis collected by the species Apis mellifera act against the growth of Pythium aphanidermatum. On the basis of mass spectrometry (Q-ToF MS), we determined that Artepillin C, the major constituent of green propolis, underlies the effect and displays activity against P. aphanidermatum at a minimal inhibitory concentration of 750 ?g.ml-1. Biophysical studies based on model membranes showed that this inhibitory effect may be linked with a membrane-related phenomenon: Artepillin C increases the permeability of membranes with relatively high fluidity in their lateral structure, a feature that is in line with the lipid composition reported for the cytoplasmic membrane of P. aphanidermatum.... |

Different concentrations (0,10, 20, 30, 40 and 50% v/v) of manure (pH 7.90; P%=0.525; N%=1.44; C%=3.52; K%=1.57; Ca%=1.09; Mg%=3.2) were added to a substrate consisting of perlite:vermiculite:peat:sand (2:1:2:1), with or without 10 g of NPK fertiliser (12:12:12) in 500 ml pots. These pots were randomized distributed in a growth chamber with five replications for each manure and fertilizer concentration. The substrates were infested with 10 g/l of a Pythium inoculum (broken corn-sand medium) 15 days before sowing 10 cucumber seeds (cv. Mezzolungo-Marketer). Foram coletadas amostras de água e solo em áreas de cerrado no Estado de São Paulo, Brasil, de fevereiro/1999 a fevereiro/2001. Estas amostras foram trazidas ao laboratório e tratadas segundo a técnica de iscagem múltipla. Dentre os fungos pitiáceos isolados, nove espécies pertencem ao gênero Pythium Pringsheim: Pythium echinulatum Matthews, P. graminicolum Subramanian, P. irregulare Buisman, P. rostratum Butler, P. spinosum Sawada, P. torulosum Coker & Patterson, P. ultimum Trow var. ultimum, P. undulatum Petersen e P. vexans de Bary. En raíces de plantas adultas de tomate se observó una lesión castaña de aspecto húmedo, que se extendía 2-4 cm por encima del suelo. Las plantas atacadas se marchitaron y murieron. El objetivo del trabajo fue aislar e identificar al organismo causal de esta sintomatología. Se usaron medios de cultivos generales y selectivos. A las 24 horas desarrollaron colonias con micelio blanco algodonoso. Se evaluó la morfología de la colonia y la producción, morfología y dimensiones de estructuras reproductivas. A solarização durante dois meses, de um solo naturalmente infestado com Pythium, promoveu o controle do patógeno, de modo semelhante a aplicação de metalaxyl + mancozeb, na cultura de crisântemo. A incidencia media de plantas mortas foi de 2,3% nas parcelas solarizadas: 1,0% no solo solarizado que recebeu metade da dose dos fungicidas: 9.0% com a aplicacao da dose completa dos fungicidas ( 1g de matalaxyl + 4,8g de mancozeb/m2) e 38,9% na testemunha nao tratada. A solarização também promoveu o controle de plantas daninhas, avaliada através de peso de matéria seca das plantas emergentes aos 21 dias após o transplante das mudas, semelhantemente ao herbicida oxidiazon. |

The genus Pythium (Pythiaceae (Peronosporomycetes - ex Oomycetes), Kingdom Straminipila) includes important pathogens, affecting a wide range of hosts of economic value, causing damping-off and decline of herbaceous and wooded plants due to rootlets rot. Three new species of Pythium, namely, P. oopapillum, P. emineosum and P. camurandrum are presented in this paper based on morphological descriptions and molecular phylogenetic characterisation. These new species were isolated from various ecological regions in Canada. They have unique morphological features in the genus Pythium, and form distinct clades in maximum parsimony analyses, which are also supported by maximum likelihood phylogeny using general time reversible model (GTR), and Bayesian inference (BI) phylogeny using Markov Chain Monte Carlo (MCMC) analysis methods. A comparative study of the new species with closely related taxa, their clade positions, and morphological features are described in this paper. |
