We report the antinociceptive activity, determined by the writhing, formalin and hot-plate tests in mice, of crude (F0/60), lectin and carbohydrate fractions isolated by ammonium sulfate precipitation (0 to 60%) from Bryothamnion seaforthii and B. triquetrum, species of red algae. Not only fraction F0/60 but also lectins from both species significantly inhibited acetic acid-induced abdominal contractions after intraperitoneal or oral administrations. In the formalin test, lectins (1 and 5 mg/kg, ip, and 5 to 20 mg/kg, po) inhibited the 1st and 2nd phases (5 and 20 min, respectively), but the effect occurred predominantly on the 2nd phase. The antinociceptive effects of a lectin (LEC) isolated from the marine alga Amansia multifida were determined in Swiss mice. The LEC (1, 5, and 10 mg/kg) inhibited acetic acid-induced abdominal writhings in a dose-dependent manner after intraperitoneal or oral administration. A partial but significant inhibition of writhings was observed after the combination of LEC (10 mg/kg) with avidin (1 mg/kg), a potent inhibitor of the hemmaglutinant activity of the lectin. However, total writhing inhibition was demonstrable in the group of mice treated with LEC plus mannose (1 mg/kg), as compared to LEC alone or to control groups. Se evaluó el efecto del tratamiento alcalino con NaOH sobre el rendimiento en la extracción y en las propiedades físicas del agar de Gracilaria verrucosa recolectada en Santa Verónica (Atlántico, Colombia). El tratamiento alcalino se realizó a temperatura entre 80 y 85 °C, con concentraciones de solución de NaOH 3, 5, 7 y 10 % (p/v). Se determinó el rendimiento de la extracción y al agar obtenido se le midió la fuerza del gel y las temperaturas de fusión y gelificación. L'algue rouge, Gracilaria verrucosa, a été cultivée, durant le printemps 2009, en mode suspendu à différents emplacements et profondeurs représentant les différences biogéochimiques de la lagune de Bizerte au Nord de la Tunisie. La concentration et la distribution des nutriments (ammonium, phosphate) dans l'eau, ainsi que la salinité, la température, l’oxygène et la transparence de l’eau ont été déterminées. Les méthodes analytiques internationales standardisées ont été appliquées. Les effets du mode de culture, de l’emplacement et de la profondeur sur les rendements en poids, protéines, R-Phycoérythrine et sucres totaux de l’algue ont été étudiés. L'exploitation des algues rouges appartenant au genre Gracilaria pour l'extraction de l'agar a considérablement augmenté au cours des dernières décennies. En Tunisie Gracilaria verrucosa (Hudson) Papenfuss 1950 (Rhodophyta), est candidate pour la culture dans la lagune de Bizerte (Nord de la Tunisie) où ne pousse que sur une courte période de l’année. This study aims to optimize an alternative method of extraction of carrageenan without previous alkaline treatment and ethanol precipitation using Response Surface Methodology (RSM). In order to introduce an innovation in the isolation step, atomization drying was used reducing the time for obtaining dry carrageenan powder. The effects of extraction time and temperature on yield, gel strength, and viscosity were evaluated. Three freshwater Rhodophyta species (Audouinella eugenea, A. hermannii and Compsopogon coeruleus) were tested as to their responses (photosynthesis, growth and pigment concentration) to two irradiances (low light, LL, 65 µmol m-2 s-1 and high light, HL, 300 µmol m-2 s-1) and two periods (short time, ST, 4 d, and long time, LT, 28 d). Glutamate dehydrogenases (GDH, EC 1.4.1.2-4) catalyse the entry of ammonium into the organic cycle via amination of a-ketoglutarate and its release via deamination of Lglutamate. NAD-GDH facilitates NAD-/NADP-dependent deamination while NADPH-GDH facilitates NADH-/NADPH-dependent amination reactions. In the study reported here, GDHs were extracted from the red alga Gracilaria sordida and purified 10- to 180-fold before examining their amination and deamination reaction properties.
