Assessing paternity in japanese quails (Coturnix japonica) using microsatellite markers - inferences for its mating system and reproductive success Rev. Bras. Ciênc. Avic.

Microsatellite markers were analyzed in Japanese quails, Coturnix japonica, using different methodologies (PAGE and automated genotyping), in order to evaluate their use in paternity testing. Ten animal triplets composed by a female and two males were used to mate and generate an offspring. Paternity was determined in five-day-old embryos, and the data generated by fluorescent labeled and tailored primers in PCR and further automated genotyping were robust. Three microsatellite markers were polymorphic (Na = 5-8, H E = 0.75) and no loci were found to deviate significantly from Hardy-Weinberg equilibrium or showed any evidence of linkage disequilibrium (p > 0.05). Os ácidos clorogênicos (CGAs) são compostos químicos importantes de Coffea spp. para a qualidade da bebida, pois eles interferem na adstringência e podem alterar o aroma e sabor da bebida. Aproximadamente 310.000 ESTs de Coffea estão disponíveis e possibilitam o acesso à variabilidade nucleotídica da planta e o desenvolvimento de marcadores moleculares ligados à qualidade da bebida para as principais enzimas da via de biossíntese dos CGAs: PAL, C4H, 4CL, CQT e C3?H. Neste trabalho foram detectados polimorfismos dos tipos SNP, INDEL ou SSR dentro das sequências nucleotídidicas disponíveis no Projeto Genoma Café e no NCBI. Genetic diversity of 60 Hevea genotypes, consisting of Asiatic, Amazonian, African and IAC clones, and pertaining to the genetic breeding program of the Agronomic Institute (IAC), Brazil, was estimated. Analyses were based on phenotypic multivariate parameters and microsatellites. Five agronomic descriptors were employed in multivariate procedures, such as Standard Euclidian Distance, Tocher clustering and principal component analysis. Genetic diversity of Spanish melons (Cucumis melo) of the Madrid provenance National Institute of Agronomic Research

The genetic diversity of five winter melon landraces having a historic presence in the town of Villaconejos, Spain (near Madrid) and four reference accessions including one accession from snakemelon, was assessed using the allelic variation at 19 SSR loci. Seventy-two polymorphic bands were scored which provided for adequate discrimination among the accessions examined. Cluster analysis (UPGMA) employing SSR-based genetic similarity (GS, Jaccard similarity coefficient) data resulted in a dendrogram with two major clades. Abstract Angular leaf spot (ALS) and powdery mildew (PWM) are two important fungi diseases causing significant yield losses in common beans. In this study, a new genetic linkage map was constructed using single sequence repeats (SSRs) and single nucleotide polymorphisms (SNPs), in a segregating population derived from the AND 277 x SEA 5 cross, with 105 recombinant inbred lines. Phenotypic evaluations were performed in the greenhouse to identify quantitative trait loci (QTLs) associated with resistance by means of the composite interval mapping analysis. Obtaining interspecific hybrids, and molecular analysis by microsatellite markers in grapevine PAB

The objective of this work was to assess the potential of interspecific hybridization of Vitis labruscana and Muscadinia rotundifolia by using artificial cross-pollinations. Microsatellite markers were used to confirm interspecific hybridizations and the identity of the parental genotypes. In crosses in which M. rotundifolia was used as the female parent, no true hybrids were obtained. In the reciprocal crosses, 114 seedlings were identified as true V. labruscana x M. rotundifolia hybrids. Self pollination occurred in direct and in reciprocal crosses. Transferability and characterization of microsatellite markers in two Neotropical Ficus species Genet. Mol. Biol.

Microsatellite markers were transferred and characterized for two Neotropical fig tree species, Ficus citrifolia and Ficus eximia. Our study demonstrated that microsatellite markers developed from different subgenera of Ficus can be transferred to related species. In the present case, 12 of the 15 primer pairs tested (80%) were successfully transferred to both of the above species. Eleven loci were polymorphic when tested across 60 F. citrifolia and 60 F. eximia individuals. For F. citrifolia, there were 4 to 15 alleles per locus, whereas expected heterozygosities ranged from 0.31 to 0.91. In the case of F. eximia, this was 2 to 12 alleles per locus and expected heterozygosities from 0.42 to 0.87.
