Citrulline modulates muscle protein metabolism in old malnourished rats Inra

Protein energy malnutrition is common in the elderly, especially in hospitalized patients. The development of strategies designed to correct such malnutrition is essential. Our working hypothesis was that poor response to nutrition with advancing age might be related to splanchnic sequestration of amino acids, which implies that fewer amino acids reach the systemic circulation. Administration of citrulline, which is not taken up by the liver, can offer a means of increasing whole body nitrogen availability and, hence, improve nutritional status. Thirty old (19 mo) rats were submitted to dietary restriction (50% of food intake) for 12 wk. Cytotoxicity of fumonisin B1 : implication of lipid peroxidation and inhibition of protein and DNA syntheses Inra

The effects of fumonisin B1 (FB1) from Fusarium moniliforme on lipid peroxidation and protein and DNA syntheses were studied in monkey kidney cells (Vero cells). FB1 was found to be a potent inducer of malondialdehyde (MDA), one of the secondary products formed during lipid peroxidation. At 0.14 μM (0.1 μg/ml), FB1 induced 0.496 ± 0.1 nmoles of MDA/mg protein, compared to the control level 0.134 ± 0.01 nmoles of MDA/mg protein (P < 0.005). No inhibition of protein or DNA synthesis was observed at this concentration of FB1. Inhibition of protein and DNA syntheses was observed at FB1 concentrations >14 μM (10 μg/ml) with an IC50 of 33 μM for both protein synthesis and DNA synthesis. Advances in quantitative proteomics have facilitated the measurement of large-scale protein quantification, which represents net changes in protein synthesis and breakdown. However, measuring the rate of protein synthesis is the only way to determine the translational rate of gene transcripts. In plant mitochondria, some of the tRNAs are encoded by the mitochondrial genome and resemble their prokaryotic counterparts, whereas the remaining tRNAs are encoded by the nuclear genome and imported from the cytosol. Generally, mitochondrial isoacceptor tRNAs all have the same genetic origin. One known exception to this rule is the group of tRNAGly isoacceptors in dicotyledonous plants. A mitochondrion-encoded tRNAGly and at least one nucleus-encoded tRNAGly coexist in the mitochondria of these plants, and both are required to allow translation of all four GGN glycine codons.
