Acceptable microorganisms concentration in a semen sample for in vitro embryo production BJM

The aim of this study was to report that the acceptable concentration of microorganisms in a semen sample for insemination may not be safe for an in vitro fertilization procedure. It seems that the semen sample should be completely germ-free, because of the excellent microorganism proliferation condition promoted by the in vitro environment.

Los valores de referencia para el eyaculado de la rata no se han determinado quizá por la dificultad para obtenerlo directamente del macho. Assessment of viability of sperm cells of Litopenaeus vannamei on cryopreservation BABT

Aiming at assessing the cryopreservation potential of Litopenaeus vannamei sperm cells, 74 spermatophores were manually extracted from the sexually mature individuals. After the toxicity test to define the cryoprotectant concentration, suspensions of spermatic cells were cryopreserved in the groups in freezing solutions comprising different cryoprotectants such as dimethyl sulfoxide (DMSO) and ethylene glycol (EG) at 10% concentration. ABSTRACT This work aimed to investigate the relationship between echogenicity, testicular biometry, and accessory gland biometry and evaluate the echogenicity pattern of these structures in Santa Ines rams. Fifty-four healthy Santa Ines sheep were classified into three age groups: group 1 - 3 to 5 months (pre-pubertal); group 2 - 7 to 11 months (pubertal); and group 3 - 13 to 28 months (sexually mature). The averages of testicular biometry and bulbourethral gland biometry were different among age groups. ABSTRACT Bovine alphaherpesviruses 1 and 5 (BoHV-1/5) are main pathogens of respiratory, reproductive and neurological diseases in cattle. The aim of this study was to investigate the frequency of neutralizing antibodies against BoHV-1/5 in serum samples and to detect viral DNA in semen of bulls from beef cattle farms located in RS. A total of 372 serum and semen sample from bulls were collected in eighteen farms. Serum samples were submitted to virus neutralization (VN) assay, while semen samples were used to detect BoHV-1 and BoHV-5 DNA by PCR. By understanding the hormonal action behind the induction of spermiation in Leptodactylus ocellatus can help in the collection of semen for artificial insemination and cryopreservation. The experiments were conducted at the Experimental Frog Farm of the Federal University of Viçosa. Seven L. ocellatus males with the following secondary sexual characteristics were selected: black thorns, developed forelimbs, and amplexus reflex. Las características seminales de sementales Criollo Lechero Tropical (CLT) son poco conocidas y son requeridas para producir dosis de semen para inseminación artificial (IA) y sementales de alto valor genético todo el año. El objetivo del presente estudio fue caracterizar el semen de sementales CLT en dos épocas del año, fresca (enero) y calurosa (mayo) en la región central de Veracruz, México, de clima cálido subhúmedo, precipitación y temperatura media anual de 1060 mm y 26.4 oC. ABSTRACT The aim of this study was to evaluate the effects of different concentrations of (+)-catechin or (-)-epigallocatechin gallate (EGCG) on goat semen freezability. Six proven stud dogs were submitted to two seminal collections by manual stimulation. Semen was evaluated and extended in tris plus egg-yolk and glycerol according to two different dilution rates. Semen was collected from six stud dogs to compare five extenders in the semen freezing process. Each ejaculate was divided in five parts and added to tris-fructose-citric acid, glicine, lactose, skim milk and tris-fructose-citrate extenders. The semen was diluted at 37°C in extenders without glycerol, in the ratio 1:1 and cooled for 60 minutes to reach a 5°C temperature. Then, extenders with glycerol in the ratio of 2:1 were added to give the final prefreezing concentration of 4% of glycerol. Correlation between sperm characteristics and testosterone in bovine seminal plasma by direct radioimmunoassay R. Bras. Zootec.

The objectives of this study were to validate a non-extractive RIA for seminal testosterone and quantify the hormone using a solid-phase commercial kit, and study the correlation between testosterone in seminal plasma and sperm characteristics. Parallelism showed a correlation index r = 0.992 (Y = -5.47 + 1.073X; R² = 0.985), indicating that the non-extractive method presented is indicated particularly for assessment of testosterone when establishing comparisons between samples. Overall mean (±SD) of testosterone level was 0.60±0.65 ng/mL. Correlation was only found between the seminal concentrations of testosterone and pH of the semen.

Cryopreservation of semen from sex-reversed females of rainbow trout aims at rationalizing the production of stocks composed by 100% females. Semen from normal males (M) and two types of genotypic females (R and G), sex-reversed by the oral administration of 17alpha-methyltestosterone, were used. R was obtained by the fertilization of normal eggs with semen of sex-reversed females while G via gynogenetic reproduction. The aim of this study was to evaluate the use of low concentrations of natural and lyophilized low density lipoprotein (LDL) from hen's egg yolk for cryopreservation of canine semen. Different ammonium sulphate concentrations were tested to extract LDL from egg yolk. The yolk was centrifuged, and LDL was isolated using 10, 20, 40, 45, or 50% ammonium sulphate solution (ASS). The LDL-rich floating fraction was collected for chemical characterization. Dry matter content was lowest (P<0.05) in the LDL extracted with the 50% ASS. The objective of this study was to evaluate the efficacy of cleaning and decontamination procedures in liquid nitrogen tanks. We evaluated 151 canisters and 133 bottoms from 133 nitrogen tanks of companies or farms for the presence of bacteria and fungi. Samples were collected from the canisters and the bottom of tanks containing liquid nitrogen. Tanks were divided into Group 1 (G1): tanks decontaminated with 2% glutaraldehyde - Glutaron® II (n = 16 canisters in 8 tanks); Group 2 (G2): decontamination with 70% ethanol (n = 20 canisters in 10 tanks); and Group 3 (G3): decontamination with 70% ethanol (n = 115 canisters in 115 tanks). Detection of equine herpesvirus 1 DNA in a single embryo and in horse semen by polymerase chain reaction Arq. Bras. Med. Vet. Zootec.

The genome of one equine embryo and three equine semen specimens collected from a Brazilian farm were tested by polymerase chain reaction (PCR) for the presence of EHV-1 and EHV-4-specific timidine kinase (TK) sequences. The PCR detected specific EHV-1 TK gene sequences in all samples tested. The peripheral blood leukocytes (PBL) of the embryo donor mare also was amplified by EHV-1 TK primers. Infectious virus was not recovered from any specimens. The animals did not show any clinical signal of EHV-1 or EHV-4 infections. EHV-4 was not detected in the studied specimens. The results indicate that PCR was more sensitive than virus isolation in cell culture for detecting EHV-1 in semen of carrier horses.

Considering the venereal transmission of visceral leishmaniasis from dogs to bitches, the aim of this study was to verify if the penile surface and smegma from infected dogs can be the source of parasites in bitches. Twelve Leishmania infantum infected dogs had semen and smegma samples collected for submission to PCR identification of the DNA of the parasite. Semen (41.7%) and smegma (50.0%) have similar positive incidence (P>0.05; Fisher's exact test), with 58.3% of the dogs positive for semen and/or smegma samples. The proportion of positivity for both semen and smegma was 33.3%, but 8.3% was positive only for semen, and 16.7% only for smegma, revealing a moderate agreement between tests (K=0.5; Kappa index). Due to the high importance of the venereal transmission of bovine leptospirosis, this study aimed to test the ability of PCR to detect Leptospira interrogans serovar hardjo DNA in experimentally contaminated bovine semen. Employing primers directed to the 16S rRNA gene, 10 bacteria/ml of semen could be detected by PCR. Results achieved in this work show that PCR can have a great potential to detect Leptospira spp. in insemination centers.
