After a serious injury or sudden death, epididymis cauda sperm recovery and cryopreservation may present as the last opportunity to obtain genetic material from a valuable stallion. This study evaluated the viability of cooled equine sperm collected by three different methods: sperm of ejaculated (G1), sperm recovered from the epididymal cauda immediately after orchiectomy (G2) and sperm recovered from the epididymal cauda after storage for 24 hours at 5°C (G3). To obtain G1 sperm, two ejaculates were collected. After 1 week, all stallions underwent a bilateral orchiectomy, and one of the removed epididymides was flushed to obtain G2 sperm. The aim of this study was to identify heparin-binding proteins (HBPs) in seminal plasma of Nellore (Bos taurus indicus) bulls. Bulls (n=4), 30-36 months old, 500-550kg with satisfactory seminal quality were selected. After the centrifugation, samples of the seminal plasma were pooled and the HBPs were isolated by heparin-affinity chromatography. The recovered HBPs fractions were pooled. One-dimensional sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDSPAGE) 12.5% was performed in vertical minigels. Eight bands with molecular weights ranging from 15 to 63kDa were observed. Two proteins were identified (22 and 25kDa), similar to those previously described in Bos taurus taurus bulls. ABSTRACT: Studies have been performed to identify the proteins present in canine seminal plasma (SP) and relate them to sperm quality as well as to discover molecular markers of reproductive tract diseases. There is evidence that heparin-binding proteins, zinc-binding proteins, and lactoferrin as well as the matrix metalloproteinase, superoxide dismutase, catalase, and glutathione peroxidase enzymes are associated with canine sperm quality. This study aimed to determine the effects of different concentrations of botulinum toxin type A (BT-A) on semen parameters, and seminal plasma biochemical and protein profiles of dogs with benign prostatic hyperplasia (BPH). Eighteen sexually intact male dogs with BPH were randomly divided in three groups, and received an intraprostatic injection of saline solution (control group - CG), 250UI (GI) or 500UI (GII) of BT-A under transabdominal ultrasound guidance. With the increasing use of artificial insemination techniques, little importance has been given to seminal plasma, since it is substituted by other diluters. However, natural sexual intercourse would not work without seminal plasma as diluter for spermatozoa in the epididimus. The study of seminal plasma brings important information for the research on diluters, which are made to substitute the forme. Thus, if the seminal plasma composition were known, it would be easier to preserve semen samples, specially those of species that present cryopreservation problems. Beside, knowledge on "fertility markers" that may be present in seminal plasma would be of great importance in the identification of animals of high reproductive value. |
