Clinicopathological and immunohistochemical evaluation of oesophageal leiomyosarcoma in a dog Ciência Rural
Alves,Renata Cintra Ciccone; Batista,Tatiana Linardi; Laufer-Amorim,Renée; Elias,Fabiana; Calazans,Sabryna Gouveia; Fonseca-Alves,Carlos Eduardo.
Oesophageal leiomyosarcoma is a malignant neoplasm of smooth muscle fibres that is infrequently reported in the literature. Because of the importance of definitive diagnosis and the low incidence of this tumour in dogs, it was reported a case of oesophageal leiomyosarcoma in a dog. A 13-year-old intact male Rottweiler presented regurgitation, hyporexia, and chronic weight loss. After an imaging examination, tumour in the thoracic oesophagus was suspected and a biopsy was sugested for definitive diagnosis. However, the owner did not permit the procedure. Six months after the clinical diagnosis, the patient was brought for evaluation of anorexia, apathy, and severe dyspnoea. Because of the patient's clinical signs and lack of response to emergency treatment,...

Functionomics: the analysis of a postgenomic concept on the basis of pregenomic pharmacological studies in smooth muscle Anais da ABC (AABC)
Jurkiewicz,Aron; Caricati-Neto,Afonso; Jurkiewicz,Neide H..
The term functionomics (Amin 2003, Neumann et al. 2004) refers to a postgenomic integrated Systems Biology (Attur et al. 2002) using a multidimensional approach for cells, tissues and organs. It considers current or future involvement among genomics, proteomics or metabolomics, including the main factors that cause biological responses and modulation under different conditions. Our objective in the present review is to summarize the contemporary understanding of functionomics of smooth muscle pharmacology, based on the results obtained on the pregenomic era during several years in our laboratory. The present approach is based on the knowledge of the dynamics of the receptor system, which comprises a cascade of phenomena, leading from the drug...

Intensity of swimming exercise influences aortic reactivity in rats BJMBR
Brito,A.F.; Silva,A.S.; Souza,I.L.L.; Pereira,J.C.; Silva,B.A. da.
Exercise is known to cause a vasodilatory response; however, the correlation between the vasorelaxant response and different training intensities has not been investigated. Therefore, this study evaluated the vascular reactivity and lipid peroxidation after different intensities of swimming exercise in rats. Male Wistar rats (aged 8 weeks; 250-300 g) underwent forced swimming for 1 h whilst tied to loads of 3, 4, 5, 6, and 8% of their body weight, respectively (groups G3, G4, G5, G6 and G8, respectively; n=5 each). Immediately after the test, the aorta was removed and suspended in an organ bath. Cumulative relaxation in response to acetylcholine (10−12-10−4 M) and contraction in response to phenylephrine (10−12-10−5 M) were measured. Oxidative stress was...

Interlinkages in the Tunica Media of Goat Aorta International Journal of Morphology
Ogeng'o,Julius; Malek,Adel Abdel; Kiama,Stephen.
The arrangement and interconnections between various components of the aortic wall influence its physicomechanical properties and functional alterations that occur in disease and ageing. The goat is a suitable model for studying cardiovascular disease, but details of the intrinsic organization of its aorta are unknown. This study therefore investigated the histomorphology of aortic tunica media in the goat by transmission electron microscopy. Sixteen healthy juvenile and adult domestic male goats (capra hircus) purchased from livestock farms in the outskirts of Nairobi were used in the study. The animals were euthanized with overdose of sodium pentabarbitone 20mg/kg, and fixed with 3% phosphate buffered glutaraldehyde solution by gravimetric perfusion....

Regulation of capacitative and non-capacitative Ca2+ entry in A7r5 vascular smooth muscle cells Biol. Res.
TAYLOR,COLIN W; MONEER,ZAHID.
A capacitative Ca2+ entry (CCE) pathway, activated by depletion of intracellular Ca2+ stores, is thought to mediate much of the Ca2+ entry evoked by receptors that stimulate phospholipase C (PLC). However, in A7r5 vascular smooth muscle cells, vasopressin, which stimulates PLC, empties intracellular Ca2+ stores but simultaneously inhibits their ability to activate CCE. The diacylglycerol produced with the IP3 that empties the stores is metabolized to arachidonic and this leads to activation of nitric oxide (NO) synthase, production of NO and cyclic GMP, and consequent activation of protein kinase G. The latter inhibits CCE. In parallel, NO directly activates a non-capacitative Ca2+ entry (NCCE) pathway, which is entirely responsible for the Ca2+ entry that...

Roles of calcium and IP3 in impaired colon contractility of rats following multiple organ dysfunction syndrome BJMBR
Zheyu,C.; Qinghui,Q.; Lunan,Y..
The purpose of the present study was to explore changes in rat colon motility, and determine the roles of calcium and inositol (1,4,5)-triphosphate (IP3) in colon dysmotility induced by multiple organ dysfunction syndrome (MODS) caused by bacteria peritonitis. The number of stools, the contractility of the muscle strips and the length of smooth muscle cells (SMC) in the colon, the concentration of calcium and IP3 in SMC, and serum nitric oxide were measured. Number of stools, fecal weight, IP3 concentration in SMC and serum nitric oxide concentration were 0.77 ± 0.52 pellets, 2.51 ± 0.39 g, 4.14 ± 2.07 pmol/tube, and 113.95 ± 37.89 µmol/L, respectively, for the MODS group (N = 11) vs 1.54 ± 0.64 pellets, 4.32 ± 0.57 g, 8.19 ± 3.11 pmol/tube, and 37.42 ±...

Sex hormone modulation of serotonin-induced coronary vasodilation in isolated heart BJMBR
Moysés,M.R.; Barker,L.A.; Cabral,A.M..
The present study was designed to evaluate the differences in the coronary vasodilator actions of serotonin (5-HT) in isolated heart obtained from naive or castrated male and female rats that were treated with either estrogen or testosterone. Hearts from 12 groups of rats were used: male and female naive animals, castrated, castrated and treated with 17ß-estradiol (0.5 µg kg-1 day-1) for 7 or 30 days, and castrated and treated with testosterone (0.5 mg kg-1 day-1) for 7 or 30 days. After treatment, the vascular reactivity of the coronary bed was evaluated. Baseline coronary perfusion pressure (CPP) was determined and dose-response curves to 5-HT were generated. Baseline CPP differed between male (70 ± 6 mmHg, N = 10) and female (115 ± 6 mmHg, N = 12) naive...

The mechanism of gentisic acid-induced relaxation of the guinea pig isolated trachea: the role of potassium channels and vasoactive intestinal peptide receptors BJMBR
Cunha,J.F.; Campestrini,F.D.; Calixto,J.B.; Scremin,A.; Paulino,N..
We examined some of the mechanisms by which the aspirin metabolite and the naturally occurring metabolite gentisic acid induced relaxation of the guinea pig trachea in vitro. In preparations with or without epithelium and contracted by histamine, gentisic acid caused concentration-dependent and reproducible relaxation, with mean EC50 values of 18 µM and Emax of 100% (N = 10) or 20 µM and Emax of 92% (N = 10), respectively. The relaxation caused by gentisic acid was of slow onset in comparison to that caused by norepinephrine, theophylline or vasoactive intestinal peptide (VIP). The relative rank order of potency was: salbutamol 7.9 > VIP 7.0 > gentisic acid 4.7 > theophylline 3.7. Gentisic acid-induced relaxation was markedly reduced (24 ± 7.0, 43...

β-Citronellol, an alcoholic monoterpene with inhibitory properties on the contractility of rat trachea BJMBR
Vasconcelos,T.B.; Ribeiro-Filho,H.V.; Lucetti,L.T.; Magalhães,P.J.C..
β-Citronellol is an alcoholic monoterpene found in essential oils such Cymbopogon citratus (a plant with antihypertensive properties). β-Citronellol can act against pathogenic microorganisms that affect airways and, in virtue of the popular use of β-citronellol-enriched essential oils in aromatherapy, we assessed its pharmacologic effects on the contractility of rat trachea. Contractions of isolated tracheal rings were recorded isometrically through a force transducer connected to a data-acquisition device. β-Citronellol relaxed sustained contractions induced by acetylcholine or high extracellular potassium, but half-maximal inhibitory concentrations (IC50) for K+-elicited stimuli were smaller than those for cholinergic contractions. It also inhibited...
