Different methods to quantify Listeria monocytogenesbiofilms cells showed different profile in their viability BJM
Winkelströter,Lizziane Kretli; Martinis,Elaine C.P. De.
Listeria monocytogenes is a foodborne pathogen able to adhere and to form biofilms in several materials commonly present in food processing plants. The aim of this study was to evaluate the resistance of Listeria monocytogenes attached to abiotic surface, after treatment with sanitizers, by culture method, microscopy and Quantitative Real Time Polymerase Chain Reaction (qPCR). Biofilms of L. monocytogenes were obtained in stainless steel coupons immersed in Brain Heart Infusion Broth, under agitation at 37 °C for 24 h. The methods selected for this study were based on plate count, microscopic count with the aid of viability dyes (CTC-DAPI), and qPCR. Results of culture method showed that peroxyacetic acid was efficient to kill sessile L. monocytogenes...

Evaluation of pregerminative treatments in gulupa seeds (P. edulis f. edulis Sims). Rev. Bras. Frutic.
Camargo,María Antonia Martínez; Coronado,Ana Cruz Morillo; Mora,Edilberto Cepeda; Fernández,Sandra Yaneth Mesa.
Abstract The genus Passiflora, commonly known as fruits of passion, includes numerous species, including gulupa (Passiflora edulis f. edulis), of nutracetic and economic importance, both for consumption in nature and for its agro-industrial applicability. This research was developed to evaluate the effects of various pre-germinative treatments on the germination of gulupa seeds from the municipality of Ramiriquí (Boyacá), such as: exposure to hydrogen peroxide (H2O2), hypochlorite of sodium (NaOCl), gibberellic acid (GA3), distilled water imbibition and stratification at 4 °C, in varying concentrations and exposure times (24 and 48 hours), using an experimental design completely random, with three repetitions of 50 seed each one. Statistical analysis...

Image analysis of coffee seeds submitted to the LERCAFE test Agronomy
Ribeiro, Bárbara Gomes; Abrahão, Christovão Pereira; Nery, Marcela Carlota; Nascimento, Rodrigo Marques; Rezende, Juliana Costa; Fialho, Cíntia Maria Teixeira.
The aim of this experiment was to quantify the stained areas of coffee seeds submitted to the LERCAFE test using image analysis. The seeds used were of the Catuaí Vermelho IAC 99 and Paradise cultivars. The physiological quality of the lots was assessed using germination tests, moisture content and a germination speed index. The LERCAFE test was conducted using seeds without parchment immersed in a solution of 3% sodium hypochlorite for 3 hours. Color photographs (RGB) with a resolution of 5 MPx were taken of each seed. The seeds were visually evaluated, and the functions generated from the analyses of the stained and non-stained regions were quantified by the Matlab R2009b program. Classification models were developed based on the Fisher Linear...

In vitro activity of disinfectants against Aspergillus spp BJM
Mattei,A.S.; Madrid,I.M.; Santin,R.; Schuch,L.F.D.; Meireles,M.C.A..
Fungi of the Aspergillus genus are widespread and contaminate the environment. Thousands of conidia are released from each phialide and dispersed in the air every day. These fungi are considered important mycose-causing agents in hospitals. Due to this, research to determine prevalent fungi from the Aspergillus genus in hospital environments, and an adequate disinfection program in these areas is are needed. This study evaluated the susceptibility of Aspergillus spp. isolated from a veterinary environment against four disinfectants. Successive dilutions of disinfectants (log2) were used according to CLSI M38-A2 microdilution technique adapted to chemical agents against 18 isolates of this genus. After 72 hours of incubation, the Minimum Inhibiting...

Influence of disinfestation and osmotic conditioning on the germinating behavior of australian royal palm (Archontophoenix alexandrae)seeds Rev. bras. sementes
Teixeira,Márcia Torres; Vieira,Henrique Duarte; Teixeira,Sílvio Lopes; Silva,Roberto Ferreira. da.
The technique of Osmotic Conditioning, which consists of partial and controlled hydration of the seeds, has obtained success with various species of seeds, increasing the germinating span and tolerance to the adverse conditions of the environment, and has also reduced the time elapsed between sowing and the emergence of the plants. Associated to ideal storage conditions, the treatment has increased the performance of the seeds of tropical wood species. Aiming at studying the germinating environment and the effect of osmotic conditioning on the germination of seeds of the Australian Royal Palm tree, two experiments were performed. The first one evaluated the effect of disinfestation of the seeds of the Australian Royal Palm tree with NaClO. The treatments...

Sodium hypochlorite sterilization of culture medium in micropropagation of Gerbera hybrida cv. Essandre. Repositório Alice
PAIS, A. K.; SILVA, A. P. da; SOUZA, J. C. de; TEIXEIRA, S. L.; RIBEIRO, J. M.; PEIXOTO, A. R.; PAZ, C. D. da.
Micropropagation requires controlling contamination that might compromise the success of the process. Thermal sterilization is traditionally used; however, costs deriving from equipment acquisition and maintenance render this technique costly. With the purpose of finding an alternative to thermal sterilization, this research aimed at assessing the efficiency and ideal concentration of sodium hypochlorite for sterilization of culture media and glassware used during rooting of micropropagated Gerbera hybrida cv. Essandre. Two experiments were carried out. In the first one, treatments consisted of control I (no sterilization), control II (thermal sterilization), and total active chlorine concentrations of 0.0005, 0.001, 0.002 and 0.003%. In the second...
