Cloning of superior coffee plants by somatic embryogenesis can assist breeding programs on reducing the cost and time for launch of new cultivars. This study aimed to evaluate the efficiency of this methodology for cloning coffee trees with high heterozygosity, and to gather evidence that clonal progenies are faithful copies of mother plants. Selected plants IAC1 and IAC 2 from Coffea arabica breeding populations, resistant to leaf rust and leaf miner, respectively, were cloned via indirect somatic embryogenesis. The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. Nanism is one of the most frequent type of mutant in micropropagated banana plants from the Cavendish subgroup. The present study aimed at studying some of the hormone factors involved in this type of mutation. Rhizomes from normal and dwarf plants from the cultivar Grand Naine were incubated for 5 d in the presence of [³H]-L-tryptophan, [³H]-indole-3-acetic acid and gibberellin, to quantify the endogenous levels of indole-3-acetic acid-ester, indole-3-acetic acid-amide, free indole-3-acetic acid, and cytokinins. A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Cell cultures of Mandevilla velutina have proved to be an interesting production system for biomass and secondary metabolites able to inhibit the hypotensive activity of bradykinin, a nonapeptide generated in plasma during tissue trauma. The crude ethyl acetate extract of cultured cells contains about 31- to 79-fold more potent anti-bradykinin compounds (e.g., velutinol A) than that obtained with equivalent extracts of tubers. Somaclonal variation may be an explanation for the wide range of inhibitor activity found in the cell cultures.
