ABSTRACT Date palm (Phoenix dactylifera L.) is a fruit tree resilient to adverse climatic conditions predominating in hot arid regions of the Middle East and North Africa. The date fruit contains numerous chemical components that possess high nutritional and medicinal values. Traditional propagation by offshoots is inefficient to satisfy current demands for date palm trees. Alternatively, micropropagation provides an efficient means for large-scale propagation of date palm cultivars. Development of blast resistant somaclones of the upland rice cultivar araguaia PAB

The degree of blast resistance of upland rice (Oryza sativa L.) cultivar Araguaia has decreased over time causing significant yield losses. The major objective of this study was to obtain blast (Pyricularia grisea) resistant somaclones, adapting greenhouse and field selection procedures. Rice blast resistance and agronomic traits were assessed in R2 to R6 generations derived from regenerant plants (R1) from immature panicles of Araguaia. The evaluation and selection procedures include testing of early segregating populations and fixed lines in the advanced generations, under natural field conditions, and artificial inoculations in the greenhouse, with prevalent races IB-1 and IB-9 of P. grisea. Cloning of superior coffee plants by somatic embryogenesis can assist breeding programs on reducing the cost and time for launch of new cultivars. This study aimed to evaluate the efficiency of this methodology for cloning coffee trees with high heterozygosity, and to gather evidence that clonal progenies are faithful copies of mother plants. Genetic changes in grapevine genomes after stress induced by in vitro cultivation, thermotherapy and virus infection, as revealed by AFLP Genet. Mol. Biol.

The Amplification Fragment Length Polymorphism (AFLP) technique was employed to study genetic variations which can be induced in vines by the stress occurring during different aspects of viticulture (in vitro cultivation, in vitro thermotherapy and virus infection). Analysis of AFLP banding patterns, generated by using 15 primer combinations, pointed to negligible genetic variation among plants exposed to individual stress. The average of similarity coefficients between differently stressed plants of the cultivars Müller Thurgau and Riesling were 0.984 and 0.991, respectively, as revealed by AFLP analysis. Genetic instability of sugarcane plants derived from meristem cultures Genet. Mol. Biol.

The RADP (Random amplified polymorphic DNA) technique was used to detect tissue-culture-induced variations in sugarcane. Plants of the Brazilian variety RB83-5486 propagated via rhizomes and via meristem cultures were studied. The polymorphism rate for 98 RAPD loci was 6.93% when the plants derived from meristems. Besides, in order to evaluate the influence of the number of subcultures on the generation of somaclonal variation, field-grown RB83-5486 plants derived from 10 meristems were studied after five subcultivations. Identification of rapd marker linked to blast resistance gene in a somaclone of rice cultivar Araguaia Trop. Plant Pathol.

The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis. Initially, the two parental DNAs from the resistant donor SC09 and 'Araguaia' were analyzed using random primers. Of the 240 primers tested, 203 produced amplification products. A large number of micropropagated plantlets of banana, Musa acuminata var. Nanjanagudu Rasabale (NR), that were developed from axillary shoot bud explants over 10 years ago were screened for genetic variation, if any, using RAPD (Random Amplified Polymorphic DNA) and ISSR (Inter-Simple Sequence Repeats) markers. Of the 4000 in vitro plantlets, 11 were used for screening that involved shoot cultures with distinct variation in morphological characteristics (morphotypes). Genetic and cytogenetic changes are frequently observed in plants regenerated from tissue culture. This phenomenon, termed somaclonal variation, is undesirable in genetic transformation processes. DNA-based molecular markers have been used to characterize and evaluate genetic variability among individuals. The RAPD technique was used to evaluate somaclonal variation in maize plants derived from tissue culture from the maize inbred line L48 (derived from Suwan). Cell cultures of Mandevilla velutina have proved to be an interesting production system for biomass and secondary metabolites able to inhibit the hypotensive activity of bradykinin, a nonapeptide generated in plasma during tissue trauma. The crude ethyl acetate extract of cultured cells contains about 31- to 79-fold more potent anti-bradykinin compounds (e.g., velutinol A) than that obtained with equivalent extracts of tubers. Somaclonal variation may be an explanation for the wide range of inhibitor activity found in the cell cultures. Somaclonal variation in Cereus peruvianus Mill. (Cactaceae): its potential to generate new varieties and broaden the species's genetic basis JBAG

In current study Random Amplified DNA-Polymorphism markers were used to detect genetic variations in C. peruvianus somaclones and to examine relationships among somaclones and plants grown from seeds in cultivated populations. The possible association of DNA fragments with morphological traits of somaclones was also investigated. Polymorphism in somaclones (62.24%) was higher than that detected in cultivated plants (42.57%). The casual grouping of somaclones presented atypical and typical shoot phenotypes showed that the random amplifi ed DNA-fragment polymorphisms detected in present study were not associated with shoot morphologies. A variação somaclonal em arroz para resistência à brusone foi investigada nas gerações R2, R3, e R4 das plantas regeneradas (Rl) derivadas de panículas imaturas da cultivar suscetível IAC- 47. A reação de brusone nas folhas foi avaliada nas populações R2 e linhas R3 através de inoculações artificiais com a raça virulenta IB-1 de Pyricularia grisea, sob condições controladas em casa de vegetação. As plantas da população R2 segregaram para resistência completa e para diversos graus de suscetibilidade. A resistência parcial das plantas R2 que apresentaram reação moderadamente suscetível foi determinada nas linhas R3.
