Coconut (Cocos nucífera L.) production has been declining in México and other countries due to deadly diseases; therefore, massive production of palms resistant to diseases is needed for replanting. Production of somatic embryos and micropropagation could be a very useful technique to produce free disease coconut population. Explants (embryogenic-structures) of Pacific Tall 2 ecotype were cultured in an induction medium defined as I (semisolid Y3 medium containing 0.65 mM 2,4-dichlorophenoxyacetic acid) and treated with the cytokinin benzyladenine (BA) at 0, 25, 100 or 200 ,µM to evaluate its effect on embryogenic callus formation. The BA treatments did not affect the percentage of embryogenic callus formation. In order to evaluate the effect of the BA... |

Abstract Papaya (Carica papaya L.) is a fruit tree of great economic and food interest worldwide. Its propagation can occur both sexually and asexually. Traditionally, it is propagated by seeds; however, vegetative propagation has been often used with the use of in vitro and ex vitro culture techniques. In vegetative propagation, the cutting method has been extensively explored. Research has also been developed using grafting. In recent years, papaya cultivation has also become a model for other fruit plants in the use of biotechnological techniques aiming applications ranging from germplasm conservation to genetic transformation of plants. Since the beginning of the 1970s, the culture of papaya tree tissues has been intensely studied, allowing greater... |

The aim of this review is to critically analyze the role of silver nitrate (AgNO3) in modulating plant growth and development. In recent years, basic studies on ethylene regulation opened new vistas for applied research in the area of micro-propagation, somatic embryogenesis, in vitro flowering, growth promotion, fruit ripening, and sex expression. Silver nitrate has proved to be a very potent inhibitor of ethylene action and is widely used in plant tissue culture. Few properties of silver nitrate such as easy availability, solubility in water, specificity and stability make it very useful for various applications in exploiting plant growth regulation and morphogenesis in vivo and in vitro. Silver ion mediated responses seem to be involved in polyamines,... |

In coffee, bioreactors are the most promising way for scaling-up micropropagation processes, particularly somatic embryogenesis. The availability of an efficient somatic embryogenesis process would allow the rapid mass production of heterozygous materials such as selected Coffea canephora clones and F1 Arabica hybrid varieties. For the last fifteen years, bioreactors (mechanically or pneumatically agitated bioreactors, temporary immersion bioreactors) have mostly been used on coffee to optimize the mass regeneration of somatic embryos from embryogenic tissues. This review presents the main results, obtained with several bioreactor models, concerning the different steps of the micropropagation process : i) the multiplication of embryogenic tissues, ii) the... |

The objective of this study was to establish cell suspension culture and plant regeneration via somatic embryogenesis of a Brazilian plantain, cultivar Terra Maranhão, AAB. Immature male flowers were used as explant source for generating highly embryogenic cultures 45 days after inoculation, which were used for establishment of cell suspension culture and multiplication of secondary somatic embryos. Five semisolid culture media were tested for differentiation, maturation, somatic embryos germination and for plant regeneration. An average of 558 plants per one milliliter of 5% SCV (settled cell volume) were regenerated in the MS medium, with 11.4 µM indolacetic acid and 2.2 µM 6-benzylaminopurine. Regenerated plants showed a normal development, and no... |

Two somatic embryogenesis receptor-like kinase genes (identified as AcSERK1 and AcSERK2) have previously been characterized from pineapple (Ananas comosus). In this work, we describe the characterization of a third gene (AcSERK3) in this family. AcSERK3 had all the characteristic domains and shared extensive sequence homology with other plant SERKs. AcSERK3 expression was studied by in situ hybridization and quantitative real-time PCR to analyze its function. Intense in situ hybridization signals were observed only in single competent cells and competent cell clusters; no hybridization signal was detected in the subsequent stages of somatic embryogenesis. AcSERK3 was highly expressed in embryogenic callus compared to other organs, e.g., 20-80 fold more... |

ABSTRACT: Plants of Coffea arabica Icatu IAC 925 germplasm show resistance to Meloidogyne paranaensis under field conditions, but with variable rates of segregation. One mother plant, IAC 3, of Icatu IAC 925 was evaluated under controlled conditions through the reaction of its clones, obtained by somatic embryogenesis. Reproduction of nematodes in clones were compared with an open-pollinated progeny from the same mother plant, C. arabica cultivar Mundo Novo IAC 515-20, susceptible to nematodes, and Clone IAC 1, also susceptible and obtained by somatic embryogenesis. Open-pollinated progeny showed segregation to resistance and was classified as susceptible according to the reproduction factor. The average number of nematodes (eggs and J2) per gram of roots... |

ABSTRACT An efficient regeneration system is a pre-requisite for the application of genetic transformation and functional genomics study of important plants. In this study, the effect of different factors (plant growth regulators, casein hydrolysate, aspartic acid and ascorbic acid) on in vitro embryogenesis and regeneration of Arta, Bahar and Zagros cultivars from mature and immature explants were investigated. Immature and mature embryos were dissected from disinfected seeds 20-25 days after pollination and imbedded mature seeds, respectively, and cultured on MS (Murashige and Skoog) medium supplemented with different compounds. The results showed that immature embryos expose high capacity of embryogenesis and regeneration in comparison with mature... |

Campomanesia adamantium is a native plant species of Brazilian Cerrado with diverse economic potential and great medicinal importance. Its sexual propagation is impaired by the recalcitrance of its seeds, which prevents effective and profitable propagation. With the purpose of establishing commercial crops and minimizing the extractive use of vegetal resources, the aim of the present study was to induce embryogenic calli in nodal segments of gabirobeira, and to determine and characterize their embryogenic phase through the establishment of a growth curve based on cellular characteristics. Calli were induced using nodal segments inoculated in WPM culture medium without the addition of hormones (control) and with different concentrations of 2,4-D, IAA, IBA,... |

Somatic and zygotic embryos from mature seeds of rauli-beech, Nothofagus alpina (Poepp. & Endl.) Oerst., were encapsulated in different artificial endosperms in order to generate a cover that fulfills the function of nourishment and protection of the embryos, facilitating their later germination. The content of sodium alginate varied by 4%, 3%, and 2%, as did the immersion time in calcium chloride (CaCl2), which acts as complexing agent. The artificial endosperm components of the Murashige and Skoog medium (MS) were added, supplemented with 0.5 mg L-1 indolacetic acid (IAA), 0.5 mg L-1 naphthaleneacetic acid (NAA), 2 mg L-1 6-benzylaminopurine (BAP) and 30 g L-1 sucrose. The germinative behaviors of encapsulated somatic and zygotic embryos were... |

Abscisic acid (ABA) and indole-3-acetic acid (IAA) participate in the propagation of plants by somatic embryogenesis, causing polar structural differentiation of the embryo. The goal of the assay was to compare endogenous levels of ABA and IAA between somatic embryos (SE) and zygotic embryos (ZE) of Nothofagus alpina (Poepp. & Endl.) Oerst. In this study, a somatic embryo maturation assay involving the addition of varying concentrations of exogenous ABA was performed on cotyledonary-stage of N. alpina. Furthermore, the endogenous levels of ABA and IAA were quantified in the immature ZE, the mature ZE, and the embryonic axis of a mature embryo of N. alpina. The current study utilized high performance liquid chromatography (HPLC) for quantification.... |

The first step in plant transformation using somatic embryogenesis or organogenesis for plant regeneration in vitro has been the identification of genotypes with the best totipotency. Thus, the objective of this experiment was to evaluate the totipotency of ten cultivars and one soybean line from the Coodetec Gene Bank. The somatic embryos developed from immature cotyledons measuring from 4 to 6 mm in length, and the organogenesis was evaluated from mature soybean seeds. The cultivars CD 201, CD 216 and CD 220 were the most promising genotypes to produce somatic embryos, whereas the cultivar CD 217 had the highest quantity of differentiated explants, which represents the best regeneration efficiency for participating in organogenic systems. In the... |
