Abstract Background Sperm contains a wealth of cell surface receptors and ion channels that are required for most of its basic functions such as motility and acrosome reaction. Conversely, animal venoms are enriched in bioactive compounds that primarily target those ion channels and cell surface receptors. We hypothesized, therefore, that animal venoms should be rich enough in sperm-modulating compounds for a drug discovery program. Biological characteristics of European hake (Merluccius merluccius) sperm ArchiMer

Very little is known on European hake's reproductive biology and especially on biological characteristics of its sperm despite a growing interest in its aquaculture potential. This study reports, for the first time some hake sperm characteristics. After activation, the swimming phase lasts 3 min (8 min when activated with 50% sea water (SW) but lower initial velocity). The initial flagellar beat frequency (BF), velocity and percentage of motile cells decreased after 100 s. After 2 days at 4 degrees C, the mean Adenylate Energy Charge (AEC) level was 0.71. Sperm stored at 4 degrees C still showed motility after 9 days for 2 individuals. When cryopreserved, the motility recovery index of thawed spermatozoa ranged from 11.8% to 29.6%.

Resumo - O objetivo deste estudo foi comparar a cinética de sêmen bovino criopreservado não sexado, sexado X e sexado Y antes e depois da seleção espermática por gradiente de Percoll. Amostras criopreservadas de sêmen não sexado (grupo NS) e sexado X (grupo SX) e Y (grupo SY) por citometria de fluxo, de quatro touros, foram avaliadas quanto à motilidade e à cinética espermática com o "computer-assisted semen analysis" (CASA) e o restante da amostra de cada grupo foi submetido à seleção espermática em gradiente de Percoll (45:60%). Após a seleção, foram realizadas as mesmas avaliações que antes da passagem pelo Percoll. Nesta pesquisa avaliou-se o efeito do colesterol sobre o sêmen de garanhões da raça Nordestina sobre a qualidade espermática. Vinte ejaculados de dois aranhões foram diluídos com BotuSemen e colesterol carreado pela ciclodextrina (CCC) adicionado no sêmen: controle, 0,75mg de CCC e 1,0mg de CCC/120x106 sptz/mL, e incubado a 26°C/15min. O sêmen foi diluído 1:5 (v/v) com diluente Lactose-gema de ovo e resfriado a 5°C/2h, envasado em palhetas de 0,5mL, e acondicionado sob vapor de nitrogênio líquido, e depois imersos. Artificial insemination mastery in red drum, Sciaenops ocellatus, requires a first experimental step wOOng at getting basic biological data. Regarding males, a relation between optical density of the solution (OD) measured by spectrophotometry and sperm concentration (SC) has been established : OD O,161*SC + 0,119. Cyclic nucleotide phosphodiesterase inhibition increases tyrosine phosphorylation and hyper motility in normal and pathological human spermatozoa Biocell

Our objective was to determine the effect of phosphodiesterase (PDE) inhibition on: 1) tyrosine phosphorylation of human spermatozoa at the tail level; and 2) sperm motion parameters and hyperactivated motility. The study was conducted with normozoospermic and asthenozoospermic samples incubated under in vitro capacitating conditions. The main outcome measures were computer-assisted sperm motion analysis and fluorescent immunodetection of phosphotyrosine-containing proteins. This study evaluated the semen quality of Santa Ines sheep fed diets containing different levels of whole cottonseed (WCS). Twenty sheep were distributed into four groups that were given diets with 0; 20; 30; 40% of WCS. For this study the semen was collected by electroejaculation, and a monthly andrological examination was performed during nine months. It has been reported that mental stress causes abnormality of spermiogram parameters. We investigated the effect of psychological stress on the L-arginine-nitric oxide (NO) pathway. Semen samples were collected from 29 healthy fourth semester medical students just before (stress) and 3 months after (non-stress) the final examinations. Psychological stress was measured by the State Anxiety Inventory questionnaire. The effects of reduced doses of Ovaprim™ (GnRHa + domperidone) on sperm release of Brycon orbignyanus and Prochilodus lineatus were evaluated. Furthermore, sperm quality was compared among fresh, equilibrated and post-thaw samples. Motility characteristics of spermatozoa in cod (Gadus morhua) and hake (Merluccius merluccius) ArchiMer

As in many marine fish, motility is triggered by osmotic pressure (OP) in cod and hake spermatozoa and motility ranges several minutes with a high velocity (above 50 mu m/s) period restricted to 100 s. A decrease in the flagellar beat frequency (BF) is partly responsible of this briefness. Detailed observations of the flagella during the motility period showed: 1) sea water provokes OP damages (blebs) which impair the correct wave propagation 2) waves become restricted to the proximal flagellum and tip becomes devoid of wave 3) the wave amplitude decreases. The combination of these factors drastically limits to the earliest period of motility the ability for spermatozoa to efficiently progress towards egg for fertilization.

Short-term cold storage of sperm from six neotropical characiformes fishes BABT

Sperm of the following Neotropical Characiformes fish species were tested for cold storage: Brycon lundii, Piaractus mesopotamicus, Leporinus elongatus, Leporinus friderici, Prochilodus lineatus and Prochilodus marggravii. Each sperm sample was split into two aliquots. The first was placed into a plastic bag with air or oxygen and the second, in a plastic tube with air. The samples were maintained at temperatures between 1.7-4.9 ºC. The rate of sperm motility was estimated using a 50 mM NaCl solution as the activating solution. Sperm features in turbot (Scophthalmus maximus): a comparison with other freshwater and marine fish species ArchiMer

This paper presents some features of turbot sperm and compares them with data reported in the literature on other freshwater and marine fish. In captivity, turbot spermiation period lasts 6 months. When compared to other fish species, turbot males are characterized by low values of maximum gonadosomatic index (0.6-0.8%) and are poor sperm producers in terms of volume (0.2-2.2 ml), concentration (0.7-11.0 x 10(9) spermatozoa/ml) and total sperm number released per stripping (0.2-12.0 x 10(9) spermatozoa). Sperm Motility and Viability in West African Dwarf Rams Treated with Euphorbia hirta International Journal of Morphology

Four adult sexually matured and clinically healthy West African Dwarf (WAD) rams aged between 24 and 30 months were used for the study. The rams were first used as control and later as experimental animals upon being orally dosed with Euphorbia hirta extract at 400mg/kg body weight for 14 days. Semen samples were collected from the rams a day after the administration of the plant extra and seven days after. The objective of the study was to investigate the effect of Euphorbia hirta on the semen picture of WAD rams. There were significantly differences (P <0.05) in the semen picture as reflected in a reduction of sperm motility from 80% to 47.5% and live-dead ratio from 90.75% to 32.5% in the control and post-experimental stages of the study...
