Cryopreservation is a long-term storage technique to preserve the biological material without deterioration for extended period of time at least several thousands of years. The ability to preserve and store both maternal and paternal gametes provides a reliable source of fish genetic material for scientific and aquaculture purposes as well as for conservation of biodiversity. Successful cryopreservation of fish sperm have been achieved for more than 200 fish species and many fish species have been adequated for the purpose of cryobanking. Cryopreservation of fish embryo is not viable, mainly because of the same limitations as in fish oocytes, i.e., high chilling sensitivity and low membrane permeability. However, cryopreservation of isolated embryonic... |

We have previously shown that polymorphonuclear neutrophils (PMNs) are present in bovine oviduct fluid under physiological conditions, and that the oviduct provides a microenvironment that protects sperm from phagocytosis by PMNs. Alpha 1-acid glycoprotein (AGP) is a major acute-phase protein produced mainly in the liver that has immunomodulatory functions. AGP mRNA is expressed in extrahepatic organs, such as the lung, kidney, spleen, lymph node, uterus, and ovary. Therefore, in this study, we investigated, 1) the local production of AGP in the bovine oviduct, 2) the effect of AGP on the phagocytic activity of PMNs for sperm and superoxide production and 3) the impact of AGP desialylation on the PMN phagocytosis of sperm. The AGP gene was expressed in... |

The present study was designed to investigate the topographical distribution of seminal plasma (SP) proteins on epididymal and ejaculated bovine sperm. Using immunocytochemistry and confocal microscopy the binding patterns of bovine SP proteins BSP-A3, albumin, transferrin, prostaglandin D-synthase (PGDS) and nucleobindin in ejaculated and cauda epididymal sperm from adult bulls were evaluated. Experiments were performed using sperm from 5 males. Data showed a positive signal, only detected for anti-PGDS, in the acrosomal cap of epididymal and ejaculated sperm. In ejaculated sperm, a very weak signal for nucleobindin 2 in the midpiece and equatorial regions was detected, using the anti-rat nucleobindin. BSP-A3 was detected on all sperm regions studied,... |

n brooding mollusc species that incubate larvae before their release in seawater, the biology of gametes remains poorly explored. The present study describes some biological characteristics of sperm of the European flat oyster, a native species that has been over-exploited in the past and is nowadays classified as an endangered species in Europe. Flat oysters were collected by divers in the Rade de Brest (Finistère, France), during its natural reproduction period. Gonadal pH is acidic (6.31 ± 0.10). Spermatozoa are clustered in spermatozeugmata, an acellular structure in which the sperm heads are embedded. After their transfer in seawater, spermatozeugmata have a mean diameter of 64 ± 3 μm and they release free spermatozoa for a mean duration of... |

The spermatozoon of Pecten maximus is of the primitive type usually associated with species having external fertilization or emitting the sperm in the water. The size of the nucleus is reduced. The middle piece is similar to that reported for several Bivalves. It contains glycogen. But the spermatozoon of P. maximus exhibits its own ultrastructural characteristics: the short acrosome differs in size, shape, position and ultrastructure or by a combination of these from that of many bivalves. No axial rods was observed in contrast to several species of Mytilidae, Veneridae and Ostreidae. These originalities could go to the genus level. |

The biology of cod reproduction is well described in the scientific literature. However, sperm biology and spermatozoa management are poorly studied in this species. Because of its recent farming expansion, a better knowledge of cod gametes is becoming especially useful. This work aimed at establishing tools to study sperm biology in cod, and also investigated the existence of changes in cod sperm quality during the spawning period. We showed that sperm concentration could be assessed using spectrophotometry at 260 nm. Sperm motility significantly decreased after a 168-h storage at 4 degrees C. A 1:9 dilution of sperm in a non-activating medium (1/3 seawater and 2/3 freshwater, osmotic pressure: 360 mOsm kg(-1)) improved sperm storage. Sperm concentration,... |

Changes in sperm features during the movement phase are especially interesting to study in external fertilization species whose sperm duration movement is long because this implies a significant adaptation of moving cells to the external medium. This study describes the changes in tetraploid Pacific oyster sperm characteristics in relation to time post activation. Sperm individually collected on three tetraploid males were activated in seawater. Their features were analysed over a 24 h period and compared to a sperm pool collected on three diploid males as a reference. The percentage of motile spermatozoa, the intracellular ATP content, and the fine structure of spermatozoa were studied in relation to time post activation. Furthermore, the fertilisation... |

This study describes morphological changes in the male reproductive system of Miroculis amazonicus (Savage & Peters) from mature nymphs to subimago stages. The sperm ultrastructure of Massartela brieni (Lestage), Farrodes carioca (Domínguez et al) and Miroculis mourei (Savage & Peters), as well as aspects of cell fragments observed in these species' subimagos deferent ducts were described. Sperm from the three species studied are aflagellated and immotile, while those from F. carioca and Ma. brieni are approximately spherical with a homogenous nucleus and acrosome. Sperm of F. carioca present two or three mitochondria located between the nucleus and the acrosome. In Ma. brieni, only one lateral mitochondria was found. Sperm from Mi. mourei... |

Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Two of the heavy metals tested, Cu2+ and Pb2+, did not affect sperm motility when the activating... |

The trade creation and conservation of wild species in Amazon, including collared peccaries (Pecari tajacu), may be favored by artificial insemination, but the time of semen storage may lead to reduced fertility in sperm of some wild animals. Thus, the aim of this research was to evaluate the influence of semen storage at 17 °C over time on seminal features of peccaries in captivity. Eight adult males were sedated and underwent electroejaculation. The ejaculates (n = 65) were evaluated for volume, concentration, pH, sperm motility, vigor, and cell with intact plasma membrane intact (IPM) and sperm morphology. Selected ejaculates (n = 21) were diluted (1:1) in Beltsville Thawing Solution and kept during 48 hours under controlled temperature (17 °C).... |

Representative species of the families Pleuronectidae, Bothidae and Soleidae were studied throughout the year. Changes are described which were seen in the ovaries, and photographs of cross sections of the tissue are shown. The progress of egg release was noted, and in some cases correlated studies were made of gonadal changes in male. The period of egg release was prolonged over 1 one half -2 months for plaice and turbot, and for 6 months for sole. Individual fish showed these extended periods of laying, and the actual months of the year differed for each species. The massive mobilisation of reserves which necessarily accompanied the release of large numbers of eggs was reflected by turbot, in sperm production. In addition, the increase in gonadosomatic... |

Epididymal protein CRISPI is a member of the CRISP (Cysteine-RIch Secretory proteins) family and is involved in sperm-egg fusion through its interaction with complementary sites on the egg surface. Results from our laboratory have shown that this binding ability resides in a 12-amino-acid region corresponding to a highly conserved motif of the CRISP family, named Signature 2 (S2). In addition to this, our results revealed that CRISP1 could also be involved in the previous step of sperm binding to the zona pellucida, identifying a novel role for this protein in fertilization. As another approach to elucidate the participation of CRISP1 in fertilization, a mouse line containing a targeted disruption of CRISP1 was generated. Although CRISP1-deficient mice... |

The oviduct is an active contractile tube that provides the proper environment for sperm transport, capacitation and survival. Oviductal contractions are regulated by autocrine/paracrine secretion of several factors, such as prostaglandins (PGs) and endothelin-1 (EDN-1). We have previously shown that during the preovulatory stage, sperm are exposed to polymorphonuclear neutrophils (PMNs) in the bovine oviduct, and the bovine oviduct epithelial cells (BOECs) secrete molecules including PGE2 that suppress sperm phagocytosis by PMNs in vitro. In this study, we investigated the possible effects of EDN-1 on the phagocytic activity of PMNs toward sperm. The local concentrations of EDN-1 in oviduct fluid and BOEC culture medium ranged from 10–10 to 10–11 M as... |
