Peanut (Arachis hypogaea L.) is an important crop for the family-based systems in the tropics, mainly in Brazil. In the Brazilian drylands, peanuts are cropped in low technological systems, and cheap and efcient technologies are needed to improve crop yield and sustainability. Despite this importance, few data are available on selecting efcient peanut rhizobia in experiments under diferent edaphoclimatic conditions. This work evaluated the agronomic efciency and the biological nitrogen fxation (BNF) by two elite Bradyrhizobium strains under four diferent felds in the Brazilian semiarid region. We compared a new efcient strain Bradyrhizobium sp. ESA 123 with the reference strain B. elkanii SEMIA 6144, currently used in peanut rhizobial inoculants in Brazil....

Extended serotyping scheme for <i>Vibrio anguillarum</i> with the definition and characterization of seven provisional O-serogroups OMA
Pedersen, K.; Grisez, L.; van Houdt, R.; Tiainen, T.; Ollevier, F.P.; Larsen, J.L..
The present paper summarizes the serotyping scheme of the fish pathogenic bacterium <i>Vibrio anguillarum</i> and defines seven additional O-serogroups. Strains, collected in our laboratories that were nontypable with antisera against the previously defined 16 O-serotypes, were used for generating new antisera and were characterized further by means of LPS profiles, Western blots, and serological reactions. On the basis of the results, it is suggested that the seven new O-serogroups are to be included in the existing serotyping system as serotypes O17-O23. However, the existence of further <i>V. anguillarum</i> strains that were not typable with any of the 23 O-antisera suggested the existence of additional...

In this study, goat fermented milk trials were prepared with six potentially probiotic autochthonous cultures Lactobacillus rhamnosus EM1107, Lactobacillus plantarum (CNPC001, CNPC002, CNPC003 and CNPC004) and Lactobacillus mucosae CNPC007in the presence and absence of a starter culture (Streptococcus thermophilus, ST) and in the presence and absence of inulin (IN) or oligofructose (FOS), consisting of 36 trials. The fermented milk without the starter culture only achieved a lactic acid content above 0.6 g/100 g after 24 h, thus requiring the presence of ST culture for a faster acidification. After the fermentation, three trials with the best fermentative performance and visual firmness (EM1107 + ST + IN, CNPC003 + ST + IN and CNPC007 + ST + IN) were...

Growth, delayed fluorescence and pigment composition of four Prorocentrum minimum strains growing at two salinities Biol. Res.
BERDEN-ZRIMEC,MAJA; FLANDER-PUTRLE,VESNA; DRINOVEC,LUKA; ZRIMEC,ALEXIS; MONTI,MARINA.
Prorocentrum minimum is a potentially harmful and widely distributed marine dinoflagellate. Several P. minimum strains have already been studied, showing phylogenetical relations of strains isolated from the same geographical regions. Similarity among the strains was further examined on the basis of their physiology. Pigment composition and concentration, as well as delayed fluorescence (DF) decay kinetics and intensity, were measured in four P. minimum strains isolated from the Baltic and Adriatic Seas. The strains were grown at two salinities characteristic of the Baltic (8 PSU) and North Adriatic Seas (32 PSU). Strain differences in DF decay kinetics and growth did not always follow their genetic relations. While two strains showed similarities to the...

MALDI-TOF MS Supplementary database for species identification employing the yeast diversity encountered on southern Brazil grapes. Repositório Alice
AGUSTINI, B. C.; SILVA, G. A. da; BOMFIM, T. M. B. B..
The study of grape microflora is of interest when autochthonous yeasts, which are related to typical wine characteristics, are intended to be used in winemaking. The election of matrix-assisted laser desorption-ionization time-of-flight mass spectrometry (MALDI-TOF MS) as the first method for yeast identification was based on its accuracy and rapidity compared to alternative laboratory protocols for identification. The aims of this study are to consolidate the MALDI-TOF MS Supplementary database for environmental yeasts already constructed, to expand it through the addition of standard spectra of not included yeast species, and to discuss the grape microflora encountered in Southern Brazil. A total of 358 strains, isolated from grape berries, were...

Molecular variants of Grapevine rupestris stem pitting-associated virus infecting grapevines (Vitis spp.) in Brazil Ciência Rural
Fajardo,Thor Vinícius Martins; Kin,Aléxis Cardama; Nickel,Osmar.
ABSTRACT: Grapevine rupestris stem pitting-associated virus (GRSPaV) is one of the most common viruses of grapevine. It is involved in the graft-transmissible disease rupestris stem pitting of the rugose wood complex. The objective of the research was to perform the molecular characterization of the coat protein (CP) gene of sixteen Brazilian GRSPaV isolates aiming to determine the occurrence of molecular variants (strains) of this virus. Nine grapevine samples were evaluated, from which dsRNA was extracted. Nucleotide sequences were generated by Next generation sequencing (NGS). Fifteen complete sequences of the GRSPaV CP gene were obtained and phylogenetically analyzed. Multiple alignments of the sequences showed identities of nucleotides ranging from...

Separation of Escherichia coli from natural samples for identification of sources and microcosm inoculation. Repositório Alice
CARNEIRO, M. T.; PEREZ, D. V.; FEITOSA, R. C.; WASSERMAN, J. C..
Obtaining uncultured Escherichia coli from natural waters is an important step in the study of microbes in the environment, which are critical for bacterial decay and microbial source tracking. The quality of the samples used can influence the assays, because high contaminant concentrations, differing cell ages, and physiologic states can impair results. The proposed separation is based on a three-step filtration method applied to replicates of seven samples from a sewage plant affluent, collected in different periods. Aliquots of the leachate were inoculated into microcosms, aiming to observe the cultivability of the cells. The assay resulted in colimetry values ranging between 104 and 105 cells. In the leachate, averages of 1.05% of total coliforms and...
