This study describes the preparation and use of Roselle (Hibiscus sábddariffa) for the differential staining of testicular tissue sections to find out its staining effect on nuclear, cytoplasmic and other structures. Various treatments using modifications of the plant extract in water were carried out on sections of the rabbit testis. Various levéis of differentiation of nuclear and cytoplasmic structures as well as other structures of this organ was obtained especially when 1% eosin was applied as a counter stain. The best staining result was obtained when iron alum was used to mordant the extract and when the extract, mordanted with potassium alum was acidified using acetic acid and used to stain the sections. Modification of the aqueous extract to an... |

The identification of expressed genes involved in sexual precocity of the mitten crab (Eriocheir sinensis) is critical for a better understanding of its reproductive development. To this end, we constructed a cDNA library from the rapid developmental stage of testis of E. sinensis and sequenced 3,388 randomly picked clones. After processing, 2,990 high-quality expressed sequence tags (ESTs) were clustered into 2,415 unigenes including 307 contigs and 2,108 singlets, which were then compared to the NCBI non-redundant (nr) protein and nucleotide (nt) database for annotation with Blastx and Blastn, respectively. After further analysis, 922 unigenes were obtained with concrete annotations and 30 unigenes were found to have functions possibly related to the... |

Lately, researchers have taken into consideration studies on birds since they represent an excellent nutritional source. There are several classical descriptions of the male reproductive tract, always aiming at establishing a correlation with shape, testicular size, age and sexual maturity. This study analyzed 50 male Gallus domesticus, 1 to 64 weeks old. The birds were collected with 10 days and then weekly until 24 weeks, following 37, 48, 59 and 64 weeks, and sacrificed by cervical displacement. It was observed the sintopies of testis with the other organs. Further, it was done the testicular measurement and then the statistical analysis by following the model of testis weight variation due to the animal age. Our results showed that the maximum weight... |

A study was conducted on 70 embryo/ foetii of goats to observe the descent of testis. These foetii were grouped into I (0-30 days), II (31-60 days), III (61-90 days), IV (91-120days) and V (121 days up to term, having 14 embryos/ foetii in each group. The genital ridge was observed at 1.2cm CRL (23 day) on the ventromedial aspect of mesonephros. At 42nd day, the genital ridge developed into cylindrical structure, the testis, located in the middle of metanephros. The abdominal migration was completed upto 88th day. The testes were situated in the inguinal canal from 89th to 95th day. From 90th day onward, scrotal migration had been observed, however the testis did not reach the base of the scrotum up to term. During the descent of testis, a peritoneal fold... |

Anti-Müllerian hormone (AMH), a glycoprotein secreted from the fetal testis, is responsible for regression of the Müllerian duct in the male fetus. The aim of this study was to evaluate the usefulness of serum AMH as a biomarker for diagnosis of cryptorchidism in horses. Serum AMH concentrations were measured in intact stallions, hemi-castrated unilateral cryptorchid stallions, and geldings. In addition, expression of AMH was characterized in cryptorchid testes by immunohistochemistry. Serum AMH was detected in intact stallions (n=11, 13.3 ± 1.8 ng/ml) and in hemi-castrated cryptorchid stallions (n=8, 17.6 ± 3.0 ng/ml), but not in geldings (n=6, all data were below the limit of detection). Immunolabeling for AMH was detected in Sertoli cells of undescended... |

ABSTRACT This study was done to evaluate the effects of lead nitrate and mercury chloride in testis tissues of Wistar rats. Lead nitrate and mercury chloride are widely used heavy metals in industry. Oral lead and mercury administrations to adult male rats at doses 45 mg/kg bw and 0.02 mg/kg bw, respectively for 4 weeks caused a significant increasing in MDA levels and antioxidant enzyme activities (SOD, CAT, GPx and GST). The MDA levels and acivities of antioxidant enzymes was lower in rats that were administrated by lead nitrate than mercury chloride treated group. Light microscopic analyses revealed that lead nitrate and mercury chloride induced numerous histopathological changes in testis tissues of rats. Histopathological observations of the testis... |

Bisphenol A [2,2-bis(4-hydroxyphenyl)propane] (BPA), 4-nonylphenol (NP) and di(2-ethylhexyl)phthalate (DEHP), and its metabolite mono-2-ethylhexyl phthalate (MEHP) are chemicals found in plastics, which act as endocrine disruptors (EDs) in animals, including human. EDs act like hormones in the endocrine system, and disrupt the physiologic function of endogenous hormones. Most people are exposed to different endocrine disruptors and concern has been raised about their true effect on reproductive organs. In the testis, they seem to preferentially attack developing testis during puberty rather than adult organs. However, the lack of information about the molecular mechanism, and the apparently controversial effect observed in different models has hampered the... |

Mammalian reproductive axis is regulated by the combination of three fundamental tissues of neuroendocrine system including hypothalamus, hypophysis and gonads. In recent years, pineal gland has been included in this axis. The aim of the present study was to investigate the effect of 12L (Light):12D (Dark) photoperiod and melatonin administration (0.5 mg/kg/day; subcutaneously) on testicular volume and cellular parameters of testis at the pinealectomized (PE) rats. For this aim, twelve adult rats were firstly pinealectomized and then divided into two groups as GI and GII randomly. The GI rats served as control group and received only normal saline, whereas GII rats were the melatonin administered group. It was found that the total testicular volume,... |

The aim of this study was to evaluate the possible beneficial effects of Catuaba Cristal® (CC), an alcoholic drink made from wine and Erythroxylum catuaba Ar. Cam on testis. Wistar rats either received CC solution (n=8) or water (n=9). Results showed significant body weight reduction within the CC group, although, no weight changes were observed for liver, kidney, testis, epididymis, seminal vesicle and prostate. The volumetric proportion and volume of interstitial tissue and lymphatic space were reduced in the treated group. In the CC group, although the nuclear volume of Leydig cells (LC) decreased, the number of LC per testis increased. These results suggested that CC had no beneficial effect on spermatogenesis of Wistar rats. |

Reduction of O2 delivery to tissues damage them, including the seminiferous epithelium. Recently, population working in high altitude has increased, so that the study of hypobaric hypoxia on spermatogenesis becomes of interest. In this study we used two groups of male, sexually mature mice Control (C) (540 meters above sea level (masl)) and chronic simulated hypobaric hypoxia (CSHH) (4,600 masl) exposed during 8, 16, 24 or 33 days. Hematocrit; reticulocytosis; testicular, epididymal and seminal vesicle weight; seminiferous epithelium height, tubular diameter, sperm count and morphology and testicular parenchyme and spermatozoa membranes lipoperoxidation were measured. Weight of testis, epididymis and seminal vesicle were reduced but they recuperate at 33... |

Caveolin-1, the first member of caveolin family reported, is recognized as the structural component of caveola, a plasma membrane invagination or vesicles that are a subcompartment distinct from clathrin-coated pits. This protein is also known to be involved in cholesterol trafficking. The aim of this study was to determine the expression of caveolin-1 in adult rat Leydig cells. Testis sections incubated with an antibody to caveolin-1 showed, by immunohistochemistry, a moderate number of Leydig cells with different degrees of immunoreaction and a strong reaction in endothelial cells and in the lamina propia of seminiferous tubules. Caveolin-1 was detected in the cell cytoplasm with a granular pattern and on the cell surface of Leydig cells cultured 24 h on... |

BACKGROUND: Testis-expressed sequence 101 (TEX101) was found to be highly expressed in testis and involved in acrosome reaction in previous studies. Recently, the metastasis suppressor function of TEX101 in cancer was disclosed, but the comprehensive investigation of its expression has rarely been reported. In this study, the expression features of TEX101 in normal human organs and seminoma were systematically analyzed. RESULTS: Immunohistochemistry demonstrated intense staining of TEX101 in human testis tissues; however, its expression in 27 other types of normal human organs, including the ovary, was negligible. Higher expression of TEX101 was observed in the spermatocytes and spermatids of the testis, but relatively lower staining was detected in... |
