Aflatoxin M1 in milk by immunoaffinity column cleanup with TLC/HPLC determination 58

During 2002 and 2003, a total of 107 samples of raw, pasteurized and ultrahigh treated temperature (UHT) milk commercialized in the cities of São Paulo and Marília (SP) were analyzed for the presence of aflatoxin M1 (AFM1). AFM1 was detected in 79 (73.8%) of milk samples, ranging from <0.02 to 0.26 mug/L.The samples were analyzed using an immunoaffinity column for cleanup and a thin layer chromatography for determining AFM1. The parameters, such as recovery, repeatibility, detection and quantification limit were evaluated to optimize this method (in-house). Based on spiked samples, the recovery values ranged from 85.83 to 73.86% at levels of 0.010-0.50 mug/L, respectively, and the relative standard deviation for repeatibility ranged from 7.73 to 2.08%....

Analysis of the composition of Brazilian propolis extracts by chromatography and evaluation of their in vitro activity against gram-positive bacteria 58

Brazilian propolis from São Paulo state was submitted to extraction using several solvents, resulting in extracts with different composition. These extracts were submitted to Thin Layer Chromatography (TLC). Bioauthographic analysis of the TLC plates identified fractions with inhibitory activity, which were then analysed by High Performance Liquid Chromatography (HPLC). In vitro assays, commonly used to evaluate the activity of propolis against Gram-positive bacteria, were compared to determine which rendered the most consistent results. The bactericidal activity of these extracts were analysed by Serial Dilution in Tubes and Agar Plate Diffusion. Construction of a supercritical fluid extraction (SFE) equipment: validation using annatto and fennel and extract analysis by thin layer chromatography coupled to image 116

Abstract The present work describes setting up a laboratory unit for supercritical fluid extraction. In addition to its construction, a survey of cost was done to compare the cost of the homemade unit with that of commercial units. The equipment was validated using an extraction of annatto seeds’ oil, and the extraction and fractionation of fennel oil were used to validate the two separators; for both systems, the solvent was carbon dioxide. The chemical profiles of annatto and fennel extracts were assessed using thin layer chromatography; the images of the chromatographic plates were processed using the free ImageJ software. La presente investigacion se dividió en tres experimentos, dos realizados en el Colegio de Postgraduados en Ciencias Agrícolas, Campus Montecillo Edo. México y la tercera en la Facultad de Agrobiología “Presidente Juárez” Uruapan Michoacan. Dada la importancia de la fertilización foliar como práctica común en los medios de producción y escasa información disponible sobre la absorción de nutrientes a nivel foliar en orquídeas, en el primer estudio se caracterizó la anatomía foliar de Cymbidium sp. e dentificó las rutas de penetración de un fertilizante foliar 10-10-10 NPK. ABSTRACT Potential of turmeric extract and its chemical fractions were evaluated to control the infestation of Bactrocera zonata peach fruit fly in a mortality-based bioassay. The turmeric extract (TE) was taken on Soxhelt's extraction apparatus and chemically fractioned by thin layer followed by column chromatography into 6 fractions (F1 ...F6). Fifty pairs of the flies were fed in cages with 250 and 500 ppm TE and its fractions separately for 20 days along with flies fed on untreated diet to serve as control. Using fecal profiles of bile acids to assess habitat use by threatened carnivores in the Maulino forest of central Chile 97

The distribution and habitat use by carnivores can be assessed by studying their tracks or feces. If these methods are to be used confidently, they should not only unequivocally discriminate among species, but should also render the same patterns of spatial distributions. We assessed the fulfillment of these requirements with five carnivores inhabiting the Maulino forest of central Chile: Galictis cuja, Oncifelis guigna, Pseudalopex culpaeus, Pseudalopex griseus, and Puma concolor.
