A comparative expression analysis of gene transcripts in brain tissue of non-transgenic and GH-transgenic zebrafish (Danio rerio) using a DDRT-PCR approach Anais da ABC (AABC)
Alves-Costa,Fernanda A.; Figueiredo,Márcio A.; Lanes,Carlos F.C.; Almeida,Daniela V.; Marins,Luis F.; Wasko,Adriane P..
The presence of higher level of exogenous growth hormone (GH) in transgenic animals could lead to several physiological alterations. A GH transgenic zebrafish (Danio rerio) line was compared to nontransgenic (NT) samples of the species through a DDRT-PCR approach, with the goal of identifying candidate differentially expressed transcripts in brain tissues that could be involved in GH overexpression. Densitometric analyses of two selected amplification products, p300 and ADCY2, pointed to a significant lower gene expression in the transgenic zebrafish (104.02 ± 57.71; 224.10 ± 91.73) when compared to NT samples (249.75 ± 30.08; 342.95 ± 65.19). The present data indicate that p300 and ADCY2 are involved in a regulation system for GH when high circulating...

Changes in Brain Monoamines Underlie Behavioural Disruptions after Zebrafish Diet Exposure to Polycyclic Aromatic Hydrocarbons Environmental Mixtures ArchiMer
Vignet, Caroline; Trenkel, Verena; Vouillarmet, Annick; Bricca, Giampiero; Begout, Marie-laure; Cousin, Xavier.
Zebrafish were exposed through diet to two environmentally relevant polycyclic aromatic hydrocarbons (PAHs) mixtures of contrasted compositions, one of pyrolytic (PY) origin and one from light crude oil (LO). Monoamine concentrations were quantified in the brains of the fish after six month of exposure. A significant decrease in noradrenaline (NA) was observed in fish exposed to both mixtures, while a decrease in serotonin (5HT) and dopamine (DA) was observed only in LO-exposed fish. A decrease in metabolites of 5HT and DA was observed in fish exposed to both mixtures. Several behavioural disruptions were observed that depended on mixtures, and parallels were made with changes in monoamine concentrations. Indeed, we observed an increase in anxiety in fish...

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous contaminants that can be present at high levels as mixtures in polluted aquatic environments. Many PAHs are potent mutagens and several are well-known carcinogens. Despite numerous studies on individual compounds, little is known about the toxicity of PAHs mixtures that are encountered in environmental situations. In the present work, zebrafish were continuously fed from 5 days post-fertilisation to 14 months post-fertilisation (mpf) with a diet spiked with fractions of either pyrolytic (PY), petrogenic light oil (LO), or petrogenic heavy oil (HO) origin at three concentrations. A decrease in survival was identified after 3 mpf in fish fed with the highest concentration of HO or LO, but not for PY. All...

Cytoskeletal and cellular adhesion proteins in zebrafish (Danio rerio) myogenesis BJMBR
Costa,M.L.; Escaleira,R.; Manasfi,M.; Souza,L.F. de; Mermelstein,C.S..
The current myogenesis and myofibrillogenesis model has been based mostly on in vitro cell culture studies, and, to a lesser extent, on in situ studies in avian and mammalian embryos. While the more isolated artificial conditions of cells in culture permitted careful structural analysis, the actual in situ cellular structures have not been described in detail because the embryos are more difficult to section and manipulate. To overcome these difficulties, we used the optically clear and easy to handle embryos of the zebrafish Danio rerio. We monitored the expression of cytoskeletal and cell-adhesion proteins (actin, myosin, desmin, alpha-actinin, troponin, titin, vimentin and vinculin) using immunofluorescence microscopy and video-enhanced,...

Akbulut,Cansu; Öztürk,Burcu; Genc,Hayriye; Zengin,Mustafa; Yön,Nazan Deniz.
ABSTRACT (4S)-2-(4-hydroxy-3-methoxyphenyl)thiazolidine-4-carboxylic acid is new synthesized substance obtained from cysteine and valine. Thiazolidine derivates have important biological responses so scientists work intensively on these compounds in recent years. It is obvious that thiazolidine contained compounds will be used in future in the pharmaceutical industry to treat important diseases. Median lethal concentrations (LC50) for 48 h and 96 h were found as 1.106±0.052 mM and 0.804mM ± 0.102 respectively. According to LC50, exposure doses were determined as control, 0.4 mM, 0.2 mM and 0.1 mM (4S)-2-(4-hydroxy-3-methoxyphenyl)thiazolidine-4-carboxylic acid. Developmental toxicity and apoptotic features on zebrafish development were evaluated in this...

Horri, Khaled.
Persistent organic pollutants (POPs) are a broad group of compounds with varying physicochemical and toxic properties. Among POP, polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) are two families of hydrophobic compounds sharing some similar environmental properties such as hydrophobicity, persistence, bioaccumulation and toxicity. Previous studies have demonstrated that these compounds could affect fish life-history traits such as survival, growth and reproductive success. However, few studies have focused on environmental situations as most previous works investigated the effect of either single congeners, or single families of molecules among POP, or high concentrations, so that results can hardly be transposed to natural...

Exploring the mechanisms of graphene oxide toxicity to zebrafish embryo. Repositório Alice
CLEMENTE, Z.; SILVA, G. H. da; MARTINEZ, D. S. T.; MAURER-MORELLI, C. V.; NUNES, M. C. S.; THOMAZ, A. A.; CASTRO, V. L. S. S. de.
Many applications of graphene oxide (GO) are still in the research phase, but its promising uses have raised concerns about their risks to human health and the environment. Environmental factors, as the presence of humic substances in aquatic environment, can affect nanomaterials behavior and its toxicological effects. A previous study of our group revealed that the exposure of zebrafish embryos to GO is related to a reduction in total body length and in acetylcholinesterase activity. Thus, the aim of this study was to investigate the implications of GO exposure in muscular development and acetylcholinesterase expression. Zebrafish embryos (n=48/group) were exposed to GO (100 mg/L) in the presence or absence of humic acid (HA, 20 mg/L) during 5 days....

Polycyclic aromatic hydrocarbons (PAHs) constitute a large family of organic pollutants emitted in the environment as complex mixtures, the compositions of which depend on origin. Among a wide range of physiological defects, PAHs are suspected to be involved in disruption of reproduction. In an aquatic environment, the trophic route is an important source of chronic exposure to PAHs. Here, we performed trophic exposure of zebrafish to three fractions of different origin, one pyrolytic and two petrogenic. Produced diets contained PAHs at environmental concentrations. Reproductive traits were analyzed at individual, tissue and molecular levels. Reproductive success and cumulative eggs number were disrupted after exposure to all three fractions, albeit to...

Influence of surface characteristics on graphene oxide toxicity for zebrafish embryo. Repositório Alice
CLEMENTE, Z.; CASTRO, V. L. S. S. de; MARTINEZ, D. S..
Despite nanotoxicological studies have been intensified in recent years, gaps remain in the methods used to assess the nanotechnology risks. These gaps are due to the complex nanomaterials behavior in the environment, especially in the presence of organic matter and depending on the nanomaterial characteristics. Studies indicate that humic acid present in the aquatic environment can increase the stability of nanomaterial dispersions and may change its toxicity to aquatic organisms. The adsorption of oxidative debris (carboxylated carbonaceous fragments - CCFs) has also been discussed as an important factor influencing GO properties and behavior. The surface characteristics of GO can influence its biotechnological application as well as its toxicological...

ABSTRACT Mori folium, the leaf of Morus alba L. (Moraceae), has been traditionally used for various medicinal purposes from ancient times to the present. In this study, we examined the effects of water extract of Mori folium (WEMF) on the production of inflammatory mediators, such as nitric oxide (NO) and prostaglandin E2 (PGE2), and reactive oxygen species (ROS) in lipopolysaccharide (LPS)-stimulated murine RAW 264.7 macrophages. Our data indicated that WEMF significantly suppressed the secretion of NO and PGE2 in RAW 264.7 macrophages without any significant cytotoxicity. The protective effects were accompanied by a marked reduction in their regulatory gene expression at the transcription level. WEMF attenuated LPS-induced intracellular ROS production in...

Long-term food-exposure of zebrafish to PCB mixtures mimicking some environmental situations induces ovary pathology and impairs reproduction ability ArchiMer
Daouk, Tarek; Larcher, Thibaut; Roupsard, Francois; Lyphout, Laura; Rigaud, Cyril; Ledevin, Mireille; Loizeau, Veronique; Cousin, Xavier.
Although the use of polychlorinated biphenyls (PCBs) has been banned for several decades, they are still present in the environment and are occasionally mechanically released from sediment or transferred through the trophic chain. Field analyses have established correlations between exposure to PCBs and alterations in fish physiology including reproductive function. Experimental exposures have been mainly performed using dioxin-like PCBs or other congeners at very high concentrations. However, these studies are often difficult to relate to real-life conditions. In the present study, we performed a life-cycle exposure using zebrafish model and mixtures representative of some environmental situations in terms of doses, composition and containing mainly non...

Ovary maturatıon stages and histological investigation of ovary of the Zebrafish (Danio rerio) BABT
Koç (Yön),Nazan Deniz; Aytekin,Yener; Yüce,Rikap.
Microscopic ovary features of Zebrafish were analyzed. Samples were fixed in Bouin’s solution, mounted in parafin and cut into 6-7 µm-thick slices which were stained with Hematoxylin and Eosin (HE), methylene blue and using Periodic-acid Schiff method (PAS). The thin sections in ultramicrotome with the thickness of approximately 0,5 - 0,7 µm were examined by using Jeol electron microscope following contrasting with Uranyl Acetate-Lead Citrate. The zebrafish was an extremely dynamic organ in which follicles underwent asynchronous development. The oocytes of Zebrafish ovary were observed in various phases. The oocyte development of zebrafish was divided into four stages (primary growth, cortical alveolus, vitellogenic and mature oocyte). Oocyte diameters...

Sublethal concentrations of waterborne copper induce cellular stress and cell death in zebrafish embryos and larvae Biol. Res.
Hernandez,Pedro P; Undurraga,Cristian; Gallardo,Viviana E; Mackenzie,Natalia; Allende,Miguel L; Reyes,Ariel E.
Copper is an essential ion that forms part of the active sites of many proteins. At the same time, an excess of this metal produces free radicals that are toxic for cells and organisms. Fish have been used extensively to study the effects of metals, including copper, present in food or the environment. It has been shown that different metals induce different adaptive responses in adult fish. However, until now, scant information has been available about the responses that are induced by waterborne copper during early life stages of fish. Here, acute toxicity tests and LC50 curves have been generated for zebrafish larvae exposed to dissolved copper sulphate at different concentrations and for different treatment times. We determined that the larvae...
